Objective: 1) To describe the prevalence of undiagnosed obstructive sleep apnea (OSA) and depressive symptoms in hypertensive men and women below 65 years of age, and 2) to describe the association of OSA to subjective sleep complaints, depressive symptoms and global perceived health. Design: Cross-sectional design focusing on nursing care outcomes of obstructive sleep apnea. Setting: Four primary care health centres in Sweden. PATIENTS: 411 consecutive patients (52% women), mean age 57.9 years (SD 5.9 years), with diagnosed hypertension (BP > 140/90). Main Outcome Measures: Prevalence of OSA and depressive symptoms, and association of OSA to sleep complaints, depressive symptoms and global perceived health. RESULTS: Mild, moderate and severe OSA was seen among 29%, 16% and 14% of patients, respectively. Depressive symptoms were seen in 16% of the total group, with a higher prevalence among men, compared to women, 21% vs. 12%. No differences were found regarding blood pressure, estimated sleep need, sleep sufficiency index, insomnia symptoms, daytime sleepiness or depressive symptoms with respect to different degrees of OSA. Apnea-hypopnea index was significantly associated to perceived health after adjustment for gender and comorbidities, but when depressive symptoms and non-restorative sleep were added to the model, 33% of the variance in global perceived health was explained. Conclusion: OSA is highly prevalent among patients with hypertension in primary care and does together with sleep complaints and depressive symptoms have a negative impact on global perceived health. Hypertensive patients without subjective sleep complaints or depressive symptoms may still have OSA.

Metabotropic glutamate receptors alter the vulnerability of neurons to excitotoxic damage and are reported to display abnormal expression in the central nervous system of ALS patients. Using reverse transcriptase polymerase chain reaction, we investigated the mRNA expression of specific metabotropic glutamate receptor subtypes in T lymphocytes of 20 patients with sporadic ALS, compared with healthy age-matched control subjects and patients with other neurological disorders. The levels of metabotropic glutamate receptor 2 mRNA were markedly reduced, whereas the expression of other subtypes (1b, 3, 8) was similar to control levels. Our findings may provide a reliable peripheral marker of the glutamatergic dysfunction that characterizes ALS.

Among multiple sclerosis(MS)susceptibility genes,the strongest non-human leukocyte antigen(HLA)signal in the Italian population maps to the TNFSF14 gene encoding LIGHT,a glycoprotein involved in dendritic cell(DC)*** fine-mapping in a large Italian dataset(4,198 patients with MS and3,903 controls),we show that the TNFSF14 intronic SNP rs1077667 is the primarily MS-associated variant in the *** quantitative trait locus(e QTL)analysis indicates that the MS risk allele is significantly associated with reduced TNFSF14 messenger RNA levels in blood cells,which is consistent with the allelic imbalance in RNA-Seq reads(P<0.0001).The MS risk allele is associated with reduced levels of TNFSF14 gene expression(P<0.01)in blood cells from 84 Italian patients with MS and 80 healthy controls(HCs).Interestingly,patients with MS are lower expressors of TNFSF14 compared to HC(P<0.007).Individuals homozygous for the MS risk allele display an increased percentage of LIGHT-positive peripheral blood myeloid DCs(CD11 c+,P=0.035)in 37 HCs,as well as in in vitro monocyte-derived DCs from 22 HCs(P=0.04).Our findings suggest that the intronic variant rs1077667 alters the expression of TNFSF14 in immune cells,which may play a role in MS pathogenesis.

A Real-Time Multi-Stage Architecture for Pose Estimation of Zebrafish Head with Convolutional Neural Networks

In order to conduct optical neurophysiology experiments on a freely swimming zebrafish,it is essential to quantify the zebrafish head to determine exact lighting *** efficiently quantify a zebrafish head's behaviors with limited resources,we propose a real-time multi-stage architecture based on convolutional neural networks for pose estimation of the zebrafish head on *** stage is implemented with a small neural ***,a light-weight object detector named Micro-YOLO is used to detect a coarse region of the zebrafish head in the first *** the second stage,a tiny bounding box refinement network is devised to produce a high-quality bounding box around the zebrafish ***,a small pose estimation network named tiny-hourglass is designed to detect keypoints in the zebrafish *** experimental results show that using Micro-YOLO combined with RegressNet to predict the zebrafish head region is not only more accurate but also much faster than Faster R-CNN which is the representative of two-stage *** with DeepLabCut,a state-of-the-art method to estimate poses for user-defined body parts,our multi-stage architecture can achieve a higher accuracy,and runs 19x faster than it on CPUs.

阿尔茨海默病(AD)与影响神经元和星形胶质细胞功能的β-淀粉样蛋白(Aβ)老年斑有关。大脑活动源于星形胶质细胞-神经元网络中神经元和星形胶质细胞功能的相互协调,而Aβ对星形胶质细胞和神经元网络功能的影响仍然未知。我们同时监测星形胶质细胞钙活动和电神经元活动,将Aβ对AD小鼠感觉诱发皮质活动的影响进行量化。结果显示,在静息状态下,皮质星形胶质细胞表现出与Aβ密度相关的自发性过度活跃。AD小鼠中感觉诱发的星形胶质细胞反应减弱,反应强度取决于Aβ的密度和距离,并且显示钙活动动力学改变。即星形胶质细胞表现出自发性过度活跃,但对感觉刺激呈低反应。此外,AD小鼠表现出感觉诱发的皮质电活动高反应性与星形胶质细胞-神经元网络相互作用的改变相关。我们的研究结果表明,AD小鼠星形胶质细胞网络的功能障碍可能引起皮质电活动失调并导致认知能力下降。

NG2神经胶质细胞在生理和病理条件下显示出较强的增殖和分化潜力。本研究用3月龄Cspg4启动子下由tdTomato标记NG2神经胶质细胞的小鼠,在不同类型的脑疾病模型(局灶性脑缺血、皮质刺伤和脱髓鞘)中研究NG2细胞的增殖和分化潜力,研究首先采用单细胞RT-qPCR和Kohonen自组织映射图分析从未损伤和特定损伤后的皮质/胼胝体分离的tdTomato阳性细胞,比较不同损伤后NG2神经胶质细胞表达概况。这种方法使我们能够区分NG2神经胶质细胞和少突胶质细胞,每个细胞群都包含4个不同的亚群。基因表达谱显示,表达GFAP(反应性星形胶质细胞的标志物)的NG2胶质细胞亚群仅在局灶性脑缺血后短暂出现。然而,在较轻的损伤后(皮质刺伤和脱髓鞘),反映少突胶质细胞成熟不同阶段的亚群明显占优势。免疫组织化学也证实了不同亚群的损伤依赖性发生率。本研究使用单细胞RNA测序分析这种独特的短暂星形胶质细胞样NG2胶质细胞亚群,并采用膜片钳技术揭示了其基本的细胞膜特性。综上所述,我们证明了星形胶质细胞样NG2神经胶质细胞是仅在局灶性脑缺血后短暂出现的NG2神经胶质细胞的特定亚群。这些位于缺血后神经胶质瘢痕中的细胞处于活跃的细胞周期,并显示出与皮质星形胶质细胞相似的电流模式,可能是缺血后胶质瘢痕形成和修复过程中的重要参与者。

Glaucoma,an irreversible optic neuropathy,primarily affects retinal ganglion cells(RGC)and causes vision loss and *** damage to RGCs in glaucoma occurs by various mechanisms,including elevated intraocular pressure,oxidative stress,inflammation,and other neurodegenerative *** the disease progresses,the loss of RGCs leads to vision ***,protecting RGCs from damage and promoting their survival are important goals in managing *** this regard,resveratrol(RES),a polyphenolic phytoalexin,exerts antioxidant effects and slows down the evolution and progression of *** present review shows that RES plays a protective role in RGCs in cases of ischemic injury and hypoxia as well as in ErbB2 protein expression in the ***,RES plays protective roles in RGCs by promoting cell growth,reducing apoptosis,and decreasing oxidative stress in H_(2)O_(2)-exposed *** was also found to inhibit oxidative stress damage in RGCs and suppress the activation of mitogen-activated protein kinase signaling *** could alleviate retinal function impairment by suppressing the hypoxia-i nducible factor-1 alpha/vascular endothelial growth factor and p38/p53 axes while stimulating the PI3K/Akt ***,RES might exert potential therapeutic effects for managing glaucoma by protecting RGCs from damage and promoting their survival.
