HDPairFinder:A data processing platform for hydrogen/deuterium isotopic labeling-based nontargeted analysis of trace-level amino-containing chemicals in environmental water

The combination of hydrogen/deuterium(H/D)formaldehyde-based isotopic methyl labeling with solid-phase extraction and high-performance liquid chromatography–high resolution mass spectrometry(HPLC-HRMS)is a powerful analytical solution for nontargeted analysis of trace-level amino-containing chemicals in water *** the huge amount of chemical information generated in HPLC-HRMS analysis,identifying all possible H/Dlabeled amino chemicals presents a significant challenge in data *** address this,we designed a streamlined data processing pipeline that can automatically extract H/D-labeled amino chemicals from the raw HPLC-HRMS data with high accuracy and ***,we developed a cross-correlation algorithm to correct the retention time shift resulting from deuterium isotopic effects,which enables reliable pairing of H-and D-labeled ***,we implemented several bioinformatic solutions to remove false chemical features generated by in-source fragmentation,salt adduction,and natural13C ***,we used a data mining strategy to construct the AMINES library that consists of over 38,000 structure-disjointed primary and secondary amines to facilitate putative compound ***,we integrated these modules into a freely available R program,*** rationale of each module was justified and its performance tested using experimental H/D-labeled chemical standards and authentic water *** further demonstrated the application of HDPairFinder to effectively extract N-containing contaminants,thus enabling the monitoring of changes of primary and secondary N-compounds in authentic water *** is a reliable bioinformatic tool for rapid processing of H/D isotopic methyl labeling-based nontargeted analysis of water samples,and will facilitate a better understanding of N-containing chemical compounds in water.

Background: Physiological and biochemical processes across tissues of the body are regulated in response to the high demands of intense physical activity in several occupations, such as firefighting, law enforcement, military, and sports. A better understanding of such processes can ultimately help improve human performance and prevent illnesses in the work ***: To study regulatory processes in intense physical activity simulating real-life conditions, we performed a multi-omics analysis of 3 biofluids(blood plasma, urine, and saliva) collected from 11 wildland firefighters before and after a 45 min, intense exercise regimen. Omics profiles post-vs. pre-exercise were compared by Student's t-test followed by pathway analysis and comparison between the different omics ***: Our multi-omics analysis identified and quantified 3835 proteins, 730 lipids and 182 metabolites combining the 3 different types of samples. The blood plasma analysis revealed signatures of tissue damage and acute repair response accompanied by enhanced carbon metabolism to meet energy demands. The urine analysis showed a strong, concomitant regulation of 6 out of 8 identified proteins from the renin-angiotensin system supporting increased excretion of catabolites, reabsorption of nutrients and maintenance of fluid balance. In saliva, we observed a decrease in 3 pro-inflammatory cytokines and an increase in 8 antimicrobial peptides. A systematic literature review identified 6 papers that support an altered susceptibility to respiratory ***: This study shows simultaneous regulatory signatures in biofluids indicative of homeostatic maintenance during intense physical activity with possible effects on increased infection susceptibility, suggesting that caution against respiratory diseases could benefit workers on highly physical demanding jobs.

Protective effects of lupeol and mango extract against androgen induced oxidative stress in Swiss albino mice

Aim： To investigate antioxidant potential of lupeol/mango pulp extract （MPE） in testosterone induced oxidative stress in prostate of male Swiss albino mice. Methods： Oral treatment of lupeol （1 mg/animal） and MPE （1 mL [20% w/v]/ animal） was given separately to animals along with subcutaneous injection of testosterone （5 mg/kg body weight） consecutively for 15 days. At the end of the study period, the prostate was dissected out for the determination of reactive oxygen species （ROS） levels, lipid peroxidation and antioxidant enzymes status （catalase, superoxide dismutase, glutathione reductase, glutathione-S-transferase）. Results： In testosterone treated animals, increased ROS resulted in depletion of antioxidant enzymes and increase in lipid peroxidation in mouse prostate. However, lupeol/MPE treatment resulted in a decrease in ROS levels with restoration in the levels of lipid peroxidation and antioxidant enzymes. Conclusion： The results of the present study demonstrate that lupeol/MPE are effective in combating oxidative stress-induced cellular injury of mouse prostate. Mango and its constituents, therefore, deserve study as a potential chemopreventive agent against prostate cancer. （Asian J Androl 2008 Mar; 10： 313-318）

Histochemical Localization of Glutathione Dependent NBT-Reductase in Mouse Skin

Objective Localization of the glutathione dependent Nitroblue tetrazolium (NBT) reductase in fresh frozen sections of mouse skin and possible dependence of NBT reductase on tissue thiol levels has been investigated. Methods The fresh frozen tissue sections (8m thickness) were prepared and incubated in medium containing NBT, reduced glutathione (GSH) and phosphate buffer. The staining for GSH was performed with mercury orange. Results The activity of the NBT-reductase in mouse skin has been found to be localized in the areas rich in glutathione and actively proliferating area of the skin. Conclusion The activity of the NBT-reductase seems to be dependent on the glutathione contents.

Modulatory effects of diallyl sulfide against testosteroneinduced oxidative stress in Swiss albino mice

Aim： To investigate the protective effect of diallyl sulfide （DAS）, a constituent of garlic, against testosterone-induced oxidative stress in male Swiss albino mice. Methods： The animals were given low （250 mg/animal） and high dose （500 mg/animal） of DAS in corn oil for 7 days along with testosterone （5 mg/kg body weight, i.p.）. At the end of the study period, the prostate and the liver were dissected to determine various antioxidant enzyme levels （catalase, superoxide dismutase, glutathione reductase, glutathione-s-transferase） and lipid peroxidation. Results： In testosterone treated mice, depleted antioxidant enzyme level was accompanied with enhancement in lipid peroxidation in prostate and liver. DAS significantly restored the testosterone-induced antioxidant enzymes and lipid peroxidation in the both organs. These changes appear to be mediated by the antioxidant-enhancing effects of DAS. Conclusion： The results of the present study suggest that DAS is effective in exerting antioxidant effects by inhibiting testosterone-induced oxidative stress and might be helpful in preventing prostate cancer.

Antitumor Activity of Diallyl Sulfide in Two-StageMouse Skin Model of carcinogenesis

It has been reported that diallyl sulfide (DAS), a sulfur-containing volatile compound in garlic (Allium sativum ), exerts anticarcinogenic activity in various rodent tumor models. In the present study, the antitumor property of DAS was tested in Swiss albino mice in the two steqe initiation-promotion mouse skin carcinogenesis. Skin cancers were initiated topically with a single subcarcinOgenic dose (52μg) of 7, 12-dimethyl benz (a) anthracene (DMBA). Promotion was performed by twice weekly applications of 12-O-tetradecanoyl phorbol-13-acetate (TPA) at a dose of 5μg/animal for 32 weeks. DAS was applied topically (250μg/animal) thrice weekly for 3 weeks for anti-initiating and 1 h prior to each promotion treatment for anti-promoting studies. The results showed that the treatment schedule of DAS can effectively delay the onset of tumorigenesis and reduce the cumulative number of tumors and the average number of tumors per mouse. In groups in which DAS applied prior to initiation or promotion, a significant population of the aniinals remained tumor-free till the termination of experiment. These findings suggest that DAS can effectively inhibit chemically induced mouse skincarcinogenesis.

Assessment of Heavy Metal Exposure in Soils of Ihwrekreka Communities, Delta State, Nigeria

Crude oil pollution in the Niger Delta, perpetrated by both local communities and industrial actors, has brought about soil pollution with its consequent ecological, human health and food challenges. The purpose of this research was to examine the concentration and distribution of heavy metals in soil from communities contaminated by crude oil in Niger Delta, and to evaluate the potential health risks to residents from exposure to these contaminants. To achieve this, soil samples were collected from the Ihwrekreka community and analyzed for heavy metal content using inductively coupled plasma mass spectrometry (ICP-MS). The analytical results in mg/kg revealed a significant metals pollution level derived from the oil spill in the soil ranging from 4.85 - 17,078 (Cu), 1.01 - 16.1 (Cd), 0.22 - 36.8 (Cr), 8.28 - 40.9 (Ni), 7.51 - 6474 (Pb), and 8.84 - 12,851 (Zn) respectively. Most of the metals were above the permissible limits of World Health Organization, with Cu, Zn, and Pb as the most contaminating metals. Lead was found to be the main contributor to the hazard index (HI) values for both children and adults in the study area, with its concentration exceeding the permitted limits set by the WHO and the EC. The hazard index (HI) values of Pb, Cu, Zn, Cd, Ni, and Cr were significantly higher than 1. These findings suggest that the release of heavy metals from an oil-contaminated site may pose a risk to human health and the environment.

Objective Calcium Glucarate (Cag), Ca salt of D-glucaric acid is a naturally occurring non-toxic compound present in fruits, vegetables and seeds of some plants, and suppress tumor growth in different models. Due to lack of knowledge about its mode of action its uses are limited in cancer chemotherapy thus the objective of the study was to study the mechanism of action of Cag on mouse skin tumorigenesis. Methods We have estimated effect of Cag on DMBA induced mouse skin tumor development following complete carcinogenesis protocol. We measured, epidermal transglutaminase activity (TG), a marker of cell differentiation after DMBA and/or Cag treatment and [3H] thymidine incorporation into DNA as a marker for cell proliferation. Results Topical application of Cag suppressed the DMBA induced mouse skin tumor development. Inhibition of Carcinogen-induced Activity of γ-Glutamyl Transpeptidase by Certain Dietary Constituents in Mouse Skin

Cancer chemoprevention, a desirable and important facet of biomedical research, provides a practical approach to identify potentially useful inhibitors of cancer development, and offers an opportunity to study the mechanism of carcinogenesis. During the recent past a number of compounds have been tested for their anticarcinogenic potential specially constituents of human diet. The enzyme gamma-glutamyl transpeptidase (GGT), which catalyses the transfer of glutamyl groups of peptides to other peptides and amino acids, has been proposed as a marker of cell proliferation and neoplasia. It also serves as a tool to evaluate the carcinogenic and cocarcinogenic potential of environmental toxicants. In the present investigations, GGT activity induced by carcinogenic polycyclic aromatic hydrocarbons, viz. 7,12-dimethyl-benz(a)anthracene (DMBA) and benzo(a)pyrene (BaP) was significantly inhibited by diallylsulfide (DAS) and indole-3-carbinol (I3C) in mouse skin. DAS and I3C are constituents of garlic. A significant inhibition in GGT levels was also observed in a strong mitogen (12-o-tetradecanoyl phorbol-13-acetate) induced activity by pretreatment of DAS/I3C in mouse skin. Therefore these dietary constituents seem to be strong modifiers of chemically induced carcinogenesis.
