Intermittent fasting can benefit breast cancer patients undergoing chemotherapy or ***,it is still uncertain how to select immunotherapy drugs to combine with intermittent *** we observed that two cycles of fasting treatment significantly inhibited breast tumor growth and lung tissue metastasis,as well as prolonged overall survival in mice bearing 4T1 and 4T07 breast *** this process,both the immunosuppressive monocytic-(M-)and granulocytic-(G-)myeloid-derived suppressor cell(MDSC)decreased,accompanied by an increase in interleukin(IL)7R^(+)and granzyme B^(+)T cells in the tumor ***,we observed that Ly6G^(low)G-MDSC sharply decreased after fasting treatment,and the cell surface markers and protein mass spectrometry data showed potential therapeutic *** investigation revealed that glucose metabolism restriction suppressed the splenic granulocytemonocyte progenitor and the generation of colony-stimulating factors and IL-6,which both contributed to the accumulation of *** the other hand,glucose metabolism restriction can directly induce the apoptosis of Ly6G^(low)G-MDSC,but not Ly6G^(high)*** summary,these results suggest that glucose metabolism restriction induced by fasting treatment attenuates the immune-suppressive milieu and enhances the activation of CD3^(+)T cells,providing potential solutions for enhancing immune-based cancer interventions.

Procyanidin A_1 and its digestive products alleviate acrylamide-induced IPEC-J2 cell damage through regulating Keap1/Nrf2 pathway

Our previous study has revealed that procyanidin A_(1)(A_(1))and its simulated digestive product(D-A,)can alleviate acrylamide(ACR)-induced intestine cell ***,the underlying mechanism remains *** this study,we elucidated the molecular mechanism for and D-A_(1) to alleviate ACR-stimulated IPEC-J2 cell *** slightly activated nuclear factor erythroid 2-related factor 2(Nrf2)signaling and its target genes,but this activation could not reduce intestine cell damage.A_(1) and D-A_(1) could alleviate ACR-induced cell damage,but the effect was abrogated in cells transiently transfected with Nrf2 small interfering RNA(siRNA).Further investigation confirmed that A_(1) and D-A_(1) interacted with Ketch-like ECH-associated protein 1(Keapl),which boosted the stabilization of Nrf2,subsequently promoted the translocation of Nrf2 into the nucleus,and further increased the expression of antioxidant proteins,thereby inhibiting glutathione(GSH)consumption,maintaining redox balance and eventually alleviating ACR-induced cell ***,there was no difference between A_(1) and D-A_(1) treated groups,indicating that A_(1) can tolerate gastrointestinal digestion and may be a potential compound to limit the toxicity of ACR.

Ursolic acid inhibiting excessive reticulophagic flux contributes to alleviate ochratoxin A-induced endoplasmic reticulum stress- and mitochondrial-mediated apoptosis

foods and animal feeds frequently become contaminated with the nephrotoxic ochratoxin A (OTA). Our prior research has indicated that ursolic acid (UA)， which is widely present in fruits and medicinal plants， has the potential to alleviate nephrotoxicity triggered by OTA. Additionally， excessive induction of endoplasmic reticulum (ER)-phagy exacerbates OTA-induced apoptosis. Therefore， further investigation is essential to comprehend whether UA can mitigate OTA-induced apoptosis by influencing ER-phagy. This objective is accomplished through a series of experiments involving assessments of cell viability， apoptosis， fluorescence microscopy， and western blot analysis. The outcomes of these experiments reveal that pre-treatment with 4 μmol/L UA for 2 h can markedly reverse the elevated apoptotic rate， the co-localization of ER and lysosomes， and the protein expressions of GRP78， p-eIF2α， Chop， Bax， and Bak， as well as the reduced cell viability and the protein expressions of Lonp1， Trap1， p62， Tex264， FAM134B， Bcl-2， and Bcl-xl， all caused by exposure to 1 μmol/L OTA for 24 h in human proximal tubule epithelial-originated kidney-2 (HK-2) cells (P induced by OTA is further amplified by UA pre-treatment (P < 0.05). In conclusion， OTA triggers a harmful feedback loop between ER stress (ERS) and excessive ER-phagy， thereby further promoting ERS- and mitochondrial-mediated apoptosis in vitro. However， this effect is significantly mitigated by UA through the inhibition of autophagosome-lysosome fusion， consequently blocking the excessive ER-phagic flux.

Distinct Effects of Three Tea Varieties Made Green Teas on Improving Diabetic Dyslipidemia induced by High-Fat Diet in C57BL/6J Mice

Currently， cultivated tea plants are classified into three varieties， namely， C. sinensis var. sinensis， C. sinensis var. assamica， and C. sinensis var. kucha. Three tea varieties made green teas (TVGTs) are widely consumed worldwide. However， a comparative investigation of the beneficial effects， mechanism， and characteristic compounds of TVGTs has not been reported. Here， three representative tea plant varieties， namely， C. sinensis var. sinensis cv. Fuding (FD)， C. sinensis var. assamica cv. Yunkang 10 (YK)， and C. sinensis var. kucha (KC)， planted in the same garden， were used to make green teas utilizing the same standard procedure. Our findings show that the TVGTs effectively improved hyperglycemia， obesity， dyslipidemia， fatty liver， and mesenteric artery (MA) hypercontractility in mice with diabetes and obesity induced by a high-fat diet (HFD) after 11- and 22-week interventions， with YK being the most effective at 22 weeks. Liver lipidomics indicated that the TVGTs restored the glycerophospholipid and sphingolipid balance and reduced triglycerides， with YK demonstrating superior effects. The TVGTs， particularly YK， suppressed triglyceride synthesis through the ARV1-FXR-SHP-SREBP-1c pathway. A chemical profile analysis revealed that YK green tea had enriched active compounds， including caffeine， quinic acid， L-theanine， gallic acid， and catechins， compared with FD and KC green teas， which may contribute to ameliorating diabetic dyslipidemia via synergistic action. As a representative tea variety of C. sinensis var. assamica， YK green tea was found to be superior to the other two green teas in preventing and treating diabetic dyslipidemia， especially with long-term consumption. These data provide important information for tea plant breeders and tea consumers.

Gut-liver axis and probiotics: Their role in non-alcoholic fatty liver disease

The incidence of obesity and its related conditions, including non-alcoholic fatty liver disease(NAFLD), has dramatically increased in all age groups worldwide. Given the health consequences of these conditions, and the subsequent economic burden on healthcare systems, their prevention and treatment have become major priorities. Because standard dietary and lifestyle changes and pathogenically-oriented therapies(e.g., antioxidants, oral hypoglycemic agents, and lipid-lowering agents) often fail due to poor compliance and/or lack of efficacy, novel approaches directed toward other pathomechanisms are needed. Here we present several lines of evidence indicating that, by increasing energy extraction in some dysbiosis conditions or small intestinal bacterial overgrowth,specific gut microbiota and/or a"low bacterial richness"may play a role in obesity,metabolic syndrome,and fatty *** conditions involving a damaged intestinal barrier("leaky gut"),the gut-liver axis may enhance the natural interactions between intestinal bacteria/bacterial products and hepatic receptors(e.g.,toll-like receptors),thus promoting the following cascade of events:oxidative stress,insulinresistance,hepatic inflammation,and *** also discuss the possible modulation of gut microbiota by probiotics,as attempted in NAFLD animal model studies and in several pilot pediatric and adult human ***,this approach appears to be a promising and innovative add-on therapeutic tool for NAFLD in the context of multi-target therapy.

Pediatric non-alcoholic fatty liver disease: Recent solutions, unresolved issues, and future research directions

Non-alcoholic fatty liver disease(NAFLD) in children is becoming a major health concern. A "multiple-hit" pathogenetic model has been suggested to explain the progressive liver damage that occurs among children with NAFLD. In addition to the accumulation of fat in the liver, insulin resistance(IR) and oxidative stress due to genetic/epigenetic background, unfavorable lifestyles, gut microbiota and gut-liver axis dysfunction, and perturbations of trace element homeostasis have been shown to be critical for disease progression and the development of more severe inflammatory and fibrotic stages [non-alcoholic steatohepatitis(NASH)]. Simple clinical and laboratory parameters, such as age, history, anthropometrical data(BMI and waist circumference percentiles), blood pressure, surrogate clinical markers of IR(acanthosis nigricans), abdominal ultrasounds, and serum transaminases, lipids and glucose/insulin profiles, allow a clinician to identify children with obesity and obesity-related conditions, including NAFLD and cardiovascular and metabolic risks. A liver biopsy(the "imperfect" gold standard) is required for a definitive NAFLD/NASH diagnosis, particularly to exclude other treatable conditions or when advanced liver disease is expected on clinical and laboratory grounds and preferably prior to any controlled trial of pharmacological/surgical treatments. However, a biopsy clearly cannot represent a screening procedure. Advancements in diagnostic serum and imaging tools, especially for the non-invasive differentiation between NAFLD and NASH, have shown promising results, e.g., magnetic resonance elastography. Weight loss and physical activity should be the first option of *** pharmacological treatments are still under development; however, drugs targeting IR, oxidative stress, proinflammatory pathways, dyslipidemia, gut microbiota and gut liver axis dysfunction are an option for patients who are unable to comply with the recommended lifestyle changes

Our previous research studies have shown that Veratrilla baillonii Franch,a food supplement used by ethnic minorities in Southwest China,has multiple pharmacological activities,such as detoxification,antiinflammatory,antioxidant,and anti-insulin ***,the detailed signal pathways for its salutary effect on damages in multiple organs due to type 2 diabetes mellitus(T2DM)remains *** current study is to evaluate the therapeutic effects of *** on T2DM rats and to explore the underlying *** T2DM rat model was successfully established by a high-sugar and high-fat diet(HFD)combination with intraperitoneal injection of a small dose of streptozotocin(STZ,35 mg/kg).Biochemical analysis and histopatholgical examinations were conducted to evaluate the anti-diabetic potential of water extracts of ***(WVBF).The results showed that the WVBF treatment can improve hyperglycemia and insulin resistance,ameliorate the liver,kidney and pancreas injuries via decreasing inflammatory cytokines such as IL-6 and TNF-α,and oxidative *** investigation suggested that WVBF modulates the signal transductions of the IRS1/PI3K/AKT/GLUT4 and AMPK *** findings demonstrate potentials of WVBF in the treatment of T2DM and possible mechanisms for its hepatoprotective activities.

Identification and the molecular mechanism of novel duck liver-derived anti-inflammatory peptides in LPS-induced RAW 264.7 cell model

In this study， 10 novel anti-inflammatory peptides were identified from duck liver， and their molecular mechanism was demonstrated based on machine learning and molecular docking. Using Sephadex G-15 gel chromatography separation， reversed-phase high-performance liquid chromatography purification， liquid chromatography-tandem mass spectrometry identification， and Biopep database comparison， 10 novel anti-inflammatory peptides were initially found. Their splendid ACE inhibition and anti-inflammatory properties were confirmed by machine learning. With binding energies less than -5.0 kcal/mol， molecular docking revealed that they could efficiently bind to the active pockets of TNF-， IL-6， COX-2， and NF-B proteins with efficiency， indicating that the compounds can spontaneously form complexes through hydrogen bonding and hydrophobic interactions with the protein binding pockets. In the LPS-induced RAW 264.7 cell model， the release of NO， TNF-α， and IL-6 and the mRNA expression of inflammatory factors (TNF-α， IL-6， COX-2， and NF-κB) were significantly inhibited by these peptides. We concluded it might be due to their anti-inflammatory effects by inhibiting the protein phosphorylation of IκBα in the cytoplasm and preventing the translocation of NF-κB p65 in the cytoplasm to the nucleus， thereby regulating the NF-κB signaling pathway. This study is essential for the screening of anti-inflammatory peptides and the investigation of the mechanism of action.

Over the past two decades, numerous non-coding RNAs(ncRNAs) have been identified in different biological systems including virology, especially in large DNA viruses such as herpesviruses. As a representative oncogenic alphaherpesvirus, Marek’s disease virus(MDV) causes an important immunosuppressive and rapid-onset neoplastic disease of poultry, namely Marek’s disease(MD). Vaccinations can efficiently prevent the onset of MD lymphomas and other clinical disease, often heralded as the first successful example of vaccination-based control of cancer. MDV infection is also an excellent model for research into virally-induced tumorigenesis. Recently, great progress has been made in understanding the functions of ncRNAs in MD ***, we give a review of the discovery and identification of MDV-encoded viral miRNAs, focusing on the genomics,expression profiles, and emerging critical roles of MDV-1 miRNAs as oncogenic miRNAs(oncomiRs) or tumor suppressor genes involved in the induction of MD lymphomas. We also described the involvements of host cellular miRNAs, lincRNAs, and circRNAs participating in MDV life cycle, pathogenesis, and/or tumorigenesis. The prospects, strategies, and new techniques such as the CRISPR/Cas9-based gene editing applicable for further investigation into the ncRNA-mediated regulatory mechanisms in MDV pathogenesis/oncogenesis were also discussed, together with the possibilities of future studies on antiviral therapy and the development of new efficient MD vaccines.
