Intra-tumor heterogeneity is now arguably one of the most-studied topics in tumor biology,as it represents a major obstacle to effective cancer *** tumor cells are highly diverse at genetic,epigenetic,and phenotypic levels,intra-tumor heterogeneity can be assumed as an important contributing factor to the nullification of chemotherapeutic effects,and recurrence of the *** on the role of heterogeneous subpopulations of cancer cells with varying cell-cycle dynamics and behavior during cancer progression and treatment;herein,we aim to establish a comprehensive definition for adaptation of neoplastic cells against *** discuss two parallel and yet distinct subpopulations of tumor cells that play pivotal roles in reducing the effects of chemotherapy:"resistant"and"tolerant"***,this review also highlights the impact of the quiescent phase of the cell cycle as a survival mechanism for cancer *** understanding the mechanisms under-lying the quiescence,it provides an insightful perspective on cancer stem cells(CsCs)and their dual and intertwined functions based on their cell cycle state in response to ***-over,CSCs,epithelial-mesenchymal transformed cells,circulating tumor cells(CTCs),and disseminated tumor cells(DTCs),which are mostly in a quiescent state of the cell cycle are proved to have multiple biological links and can be implicated in our viewpoint of cell cycle heterogeneity in ***,increasing our knowledge of cell cycle heterogeneity is a key to identifying new therapeutic solutions,and this emerging concept may provide us with new opportunities to prevent the dreadful cancer recurrence.

Microfungi have been explored further as supplement constituents in shrimp culture due to their rich protein,lipid,essential amino acids,fatty acids,and various bioactive *** researchers have evaluated the effects of two microfungi powders(Mucor circinelloides and Trichoderma harzianum)as feed supplements on growth performance,bacterial communities,and the biomass nutritional profile of white shrimp(Litopenaeus vannamei).The results showed that commercial/artificial feed(Feng-Li)with supplementation microfungi powder had a different effect on white shrimp growth performance than control after 62 days of the grow-out *** with 1.5%*** powder(M-2)could improve the growth performance of white shrimp,especially the survival rate(81.33%),final body weight(5.21 g),and shrimp biomass(211.35 g).In contrast,supplementation with 1.5%*** powder(T-2)did not improve the growth performance of white shrimp compared to control(p>0.05).Furthermore,it also affected the profile of the bacterial community in culture water as well as in shrimp *** with both microfungi could reduce the total number of bacteria in culture water at the end of the experiment,where the lowest number was obtained in treatment T-2(3.9×10^(5)CFU/mL),followed by M-2(9.9×10^(5)CFU/mL)and control(4.4×10^(6)CFU/mL).In the culture water,20 isolates of culturable bacteria were found during the trial,with the dominant bacteria in M-2:Bacillus cereus,Bacillus velezensis,Gordonia polyisoprenivorans,T-2:Acinetobacter sp.,Gordonia polyisoprenivorans,Micrococcus *** on average well colour development(AWCD),the highest bacterial activity was in the M-2(0.297-0.707),followed by T-2(0.226-0.717)and control(0.094-0.694).The highest substrate group utilised in M-2 and T-2 were polymers(0.896 and 1.348)and control:amines(0.100).The highest Shannon diversity index was obtained in the M-2(2.508),followed by control(2.286)and the T-2(2.157).As many as

Owing to their limited accuracy and narrow applicability,current antimicrobial peptide(AMP)prediction models face obstacles in industrial *** address these limitations,we developed and improved an AMP prediction model using Comparing and Optimizing Multiple DEep Learning(COMDEL)algorithms,coupled with high-throughput AMP screening method,finally reaching an accuracy of 94.8%in test and 88%in experiment verification,surpassing other state-of-the-art *** conjunction with COMDEL,we employed the phage-assisted evolution method to screen Sortase in vivo and developed a cell-free AMP synthesis system in vitro,ultimately increasing AMPs yields to a range of 0.5-2.1 g/L within ***,by multi-omics analysis using COMDEL,we identified Lactobacillus plantarum as the most promising candidate for AMP generation among 35 edible *** this,we developed a microdroplet sorting approach and successfully screened three *** mutants,each showing a twofold increase in antimicrobial ability,underscoring their substantial industrial application values.

Transcriptional regulation plays a key role in the control of seed dormancy,and many transcription factors(TFs)have been ***,the mechanisms underlying the interactions between different TFs within a transcriptional complex regulating seed dormancy remain largely ***,we showed that TF PHYTOCHROME-INTERACTING FACTOR4(PIF4)physically interacted with the abscisic acid(ABA)signaling responsive TF ABSCISIC ACID INSENSITIVE4(ABI4)to act as a transcriptional complex to promote ABA biosynthesis and signaling,finally deepening primary seed *** pif4 and abi4 single mutants exhibited a decreased primary seed dormancy phenotype,with a synergistic effect in the pif4/abi4 double ***4 binds to ABI4 to form a heterodimer,and ABI4 stabilizes PIF4 at the protein level,whereas PIF4 does not affect the protein stabilization of ***,both TFs independently and synergistically promoted the expression of ABI4 and NCED6,a key gene for ABA *** genetic evidence is also consistent with the phenotypic,physiological and biochemical analysis ***,this study revealed a transcriptional regulatory cascade in which the PIF4–ABI4 transcriptional activator complex synergistically enhanced seed dormancy by facilitating ABA biosynthesis and signaling.

Total Phenols and Flavonoids Content, Antioxidant Capacity and Lipase Inhibition of Root and Leaf Horseradish (<i>Armoracia rusticana</i>) Extracts

Horseradish (Armoracia rusticana Gaertn) is a perennial crop belonging to the Brassicaceae family, widely used as spice in food and as herbal ingredient in ethno-medicine. This study evaluated the phenolic compounds content, antioxidant capacity and anti-lipase activity of methanol, methanol/water (70/30, v/v) and methanol/water (50/50, v/v) extracts of horseradish roots and leaves. Among the extracts tested, both roots and leaves aqueous methanolic (70/30, v/v and 50/50, v/v) extracts showed higher total phenol and flavonoid contents and antioxidant capacity than the corresponding methanol extracts. But extraction yield was high for methanol/ water (50/50, v/v) extracts, in both roots and leaves. The extracts exhibited anti-lipase activity in dose-dependent manner. The results showed that the extraction yield and the antioxidant capacity were strictly dependent on the solvent polarity. The results suggest that A. rusticana could provide opportunities for the development of functional food and further in vivo studies for obesity treatment.

In accordance with the World Health Organization data,cancer remains at the forefront of fatal *** upward trend in cancer incidence and mortality has been observed globally,emphasizing that efforts in developing detection and treatment methods should *** diagnostic path typically begins with learning the medical history of a patient;this is followed by basic blood tests and imaging tests to indicate where cancer may be located to schedule a needle *** initiation of diagnosis is crucial since delayed cancer detection entails higher costs of treatment and ***,there is a need for novel cancer detection methods such as liquid biopsy,elastography,synthetic biosensors,fluorescence imaging,and reflectance confocal *** therapeutic methods,although still common in clinical practice,pose many limitations and are ***,there is a dynamic advancement of clinical research and the development of more precise and effective methods such as oncolytic virotherapy,exosome-based therapy,nanotechnology,dendritic cells,chimeric antigen receptors,immune checkpoint inhibitors,natural product-based therapy,tumor-treating fields,and photodynamic *** present paper compares available data on conventional and modern methods of cancer detection and therapy to facilitate an understanding of this rapidly advancing field and its future *** evidenced,modern methods are not without drawbacks;there is still a need to develop new detection strategies and therapeutic approaches to improve sensitivity,specificity,safety,and ***,an appropriate route has been taken,as confirmed by the approval of some modern methods by the Food and Drug Administration.

Changes in endogenous hormone levels and redox status during enhanced adventitious rooting by rare earth element neodymium of Dendrobium densiflorum shoot cuttings

The effects of neodymium nitrate （Nd^3＋） on the adventitious rooting of Dendrobium densiflorum shoot cuttings were studied. The addition of Nd^3＋（5 μmol/L） to culture medium significantly increased rooting frequency. Histological investigation showed that Nd^3＋ did not change the process of root initiation. Nd^3＋ did not influence total endogenous cytokinin levels, but significantly increased the level of endogenous indole-3-acetic acid （IAA） in the base of shoot cuttings. Compared to the control, the ratio of IAA/cytokinins was very high in the Nd^3＋ treatment. These results suggested that the enhanced rooting frequency may be related to the increase in endogenous IAA level in Nd^3＋ treatment. Analysis of enzyme activities showed that the enhanced accumulation of the endogenous IAA by Nd^3＋ should not be attributed to inhibition of IAA decomposition by IAA oxidase or promotion of cytokinin decomposition by cytokinin oxidase. Besides, Nd^3＋ increased the ratio of reduced glutathione （GSH） and oxidized glutathione （GSSG） in the process of adventitious rooting while the ratio of ascorbate （ASC） to dehydroascorbate （DHA） was not affected.

OBJECTIVE:To investigate the effect of Guijiajiao(Colla Carapacis et Plastri, CCP), the glue of tortoise shell, on bone regeneration in zebrafish and male Wistar ***: In this study, we applied in vitro and in vivo models, tissue section analysis, and quantitative polymerase chain reaction to assess the effects of CCP on bone repair. MG-63 cells were used in alkaline phosphatase(ALP) activity and mineralization assays. Zebrafish and male Wistar rats were used to evaluate the effects of CCP on bone repair in ***: A simple preparation of CCP promoted osteogenesis in vivo and in vitro, and promoted MG-63 cell proliferation as well as ALP activity and mineralization. In addition, CCP activated Akt and extracellular signaling-regulated kinase pathways and significantly increased the expression of ossification-related genes and proteins such as runt-related transcription factor-2, osteocalcin, and osteopontin. The in vivo results revealed promotion of osteogenesis by immersing zebrafish in CCP for 72 *** oral dose of 1.25 g/kg CCP significantly improved skull defects in rats, which was accompanied by an increase in serum ALP ***: One of the ingredients of Guilu Erxianjiao(tortoise shell and deer horn formula) pro-vides a practical alternative therapy for bone regeneration.

Antioxidative, cytotoxic, and anti-metastatic potentials of Laurencia obtusa and Ulva lactuca seaweeds

Objective:To investigate the antioxidant and cytotoxic properties of the most active extracts from Ulva lactuca and Laurencia obtusa against colon and cervical cancer cell ***:The antioxidant radical scavenging activity of the algal extracts was estimated using the 2,2-diphenyl-1-picrylhydrazyl colorimetric ***,the cytotoxic potential of these bioactive extracts was studied against HCT-116 and HeLa cells by 3-(4,5-dimethylthiazole-2-yl)-2,5-diphenyltetrazolium bromide *** healing analysis was adopted to evaluate the antimetastatic effects of protein extracts from both *** cycle analysis was performed by flow cytometry,and apoptotic bodies were detected using 4′,6-diamidino-2-phenylindole fluorescent *** acids of hydrolyzed protein extract were separated and identified chromatographically by high performance liquid ***:The green algae Ulva lactuca had higher lipid content than Laurencia obtusa,whereas the latter had higher protein content with high antioxidant *** extracts had significant dose-and time-dependent cytotoxicity and anti-metastatic activity against HCT-116 *** b extracts of both algae obtained from the chloroform:hexane solvent lipid-free residue caused morphological changes and induced apoptosis of HCT-116 *** analysis revealed that apoptosis induced upon Laurencia obtusa protein b treatment was triggered via the ROS pathway,causing a significant increase in the sub-G_(1) *** and arginine(5.94 and 5.47μM,respectively)were the most common amino acids in Laurencia obtusa protein b ***:Proteins of Laurencia obtusa and Ulva lactuca could be considered as an adjuvant nutraceutical source of pro-apoptotic and anti-metastatic biomolecules against colon cancer.
