Objective: To explore the specific pharmacological molecular mechanisms of Kai Xin San(KXS)on treating Alzheimer’s disease(AD) based on network pharmacology and experimental validation. Methods:The chemical compounds of KXS and their corresponding targets were screened using the Encyclopedia of Traditional chinese Medicine(ETCM) database. AD-related target proteins were obtained from MalaCards database and DisGeNET databases. Key compounds and targets were identified from the compound-targetdisease network and protein-protein interaction(PPI) network analysis. Functional enrichment analysis predicted the potential key signaling pathways involved in the treatment of AD with KXS. The binding affinities between key ingredients and targets were further verified using molecular docking. Finally, the predicted key signaling pathway was validated experimentally. Positioning navigation and space search experiments were conducted to evaluate the cognitive improvement effect of KXS on AD rats. Western blot was used to further examine and investigate the expression of the key target proteins related to the predicted pathway. Results: In total, 38active compounds and 469 corresponding targets of KXS were screened, and 264 target proteins associated with AD were identified. The compound-target-disease and PPI networks identified key active ingredients and protein targets. Kyoto Encyclopedia of Genes and Genomes(KEGG) pathway analysis suggested a potential effect of KXS in the treatment of AD via the amyloid beta(Aβ)-glycogen synthase kinase-3 beta(GSK3β)-Tau pathway. Molecular docking revealed a high binding affinity between the key ingredients and targets. In vivo,KXS treatment significantly improved cognitive deficits in AD rats induced by Aβ1-42, decreased the levels of Aβ, p-GSK3β, p-Tau and cyclin-dependent kinase 5, and increased the expressions of protein phosphatase 1alpha(PP1A) and PP2A(P<0.05 or P<0.01). Third-generation EGFR tyrosine kinase inhibitors(TKIs),exemplified by osimertinib,have demonstrated promising clinical efficacy in the treatment of non-small cell lung cancer(NSCLC).Our previous work has identified ASK120067 as a novel third-generation EGFR TKI with remarkable antitumor effects that has undergone New drug Application(NDA)submission in *** substantial progress,acquired resistance to EGFR-TKIs remains a significant challenge,impeding the long-term effectiveness of therapeutic *** this study,we conducted a comprehensive investigation utilizing high-throughput proteomics analysis on established TKI-resistant tumor models,and found a notable upregulation of branched-chain amino acid transaminase 1(BCAT1)expression in both osimertinib-and ASK120067-resistant tumors compared with the parental TKI-sensitive NSCLC *** depletion or pharmacological inhibition of BCAT1 impaired the growth of resistant cells and partially re-sensitized tumor cells to EGFR ***,upregulated BCAT1 in resistant cells reprogrammed branched-chain amino acid(BCAA)metabolism and promoted alpha ketoglutarate(α-KG)-dependent demethylation of lysine 27 on histone H3(H3K27)and subsequent transcriptional derepression of glycolysis-related genes,thereby enhancing glycolysis and promoting tumor ***,we identified WQQ-345 as a novel BCAT1 inhibitor exhibiting antitumor activity both in vitro and in vivo against TKI-resistant lung cancer with high BCAT1 *** summary,our study highlighted the crucial role of BCAT1 in mediating resistance to third-generation EGFR-TKIs through epigenetic activation of glycolysis in NSCLC,thereby supporting BCAT1 as a promising therapeutic target for the treatment of TKI-resistant NSCLC.

Neglected tropical diseases(NTDs)refer to infectious diseases caused by multiple pathogens that are prevalent in hot,humid climates in tropical *** the global economic growth and the improvement of public health status,eliminating neglected tropical diseases will be of great significance to the healthy development of human ***,the number of drugs and vaccines for NTDs treatment is extremely limited,so it is urgent to develop new *** most NTDs are caused by parasites,this paper selected parasitic diseases with high morbidity and mortality,and focused on new effective therapeutic targets and excellent lead compounds for these ***,human African trypanosomiasis(HAT),Chagas disease,leishmaniasis,filariasis and toxoplasmosis correspond to a series targets such as smHDAC8,thioredoxin glutathione reductase(TGR),*** glucokinase(TcGlcK),phosphofructokinase(PFK),type IB topoisomerase,cell division cycle-2-related Kinase,sterolmethyl transferase,calumenin,dihydrofolate reductase(DHFR)and Toxoplasma gondii farnesyl-diphosphate synthase(TgFPPs).In this paper,the pharmacological effects of typical lead compounds corresponding to each disease,the structural characteristics of the mother nucleus and the pharmacological activities of the *** addition,the binding patterns of some involved targets(such as smHDAC8)with corresponding lead compounds(such as compound 13)and the signaling pathways associated with gluconeogenesis,glycolysis,and pentose phosphate pathways are analyzed in *** this paper,the interaction mechanism between the lead compounds and the target were thoroughly discussed,in order to provide the research ideas of potential anti-parasite compounds,and further improve the understanding and prevention ability of such diseases of NTDs.

Bruton’s tyrosine kinase inhibitors(BTKis)have revolutionized the treatment of B-cell ***,safety issues related to the use of BTKis may hinder treatment continuity and further affect clinical efficacy.A comprehensive and systematic expert consensus from a pharmacological perspective is lacking for safety issues associated with BTKi treatment.A multidisciplinary consensus working group was established,comprising 35 members from the fields of hematology,cardiovascular disease,cardio-oncology,clinical pharmacy,and evidencebased *** evidence-based expert consensus was formulated using an evidence-based approach and the Delphi *** Joanna Briggs Institute Critical Appraisal(JBI)tool and Grading of Recommendations Assessment,Development,and Evaluation(GRADE)approach were used to rate the quality of evidence and grade the strength of recommendations,*** consensus provides practical recommendations for BTKis medication based on nine aspects within three domains,including the management of common adverse drug events such as bleeding,cardiovascular events,and hematological toxicity,as well as the management of drug-drug interactions and guidance for special *** multidisciplinary expert consensus could contribute to promoting a multi-dimensional,comprehensive and standardized management of BTKis.

Exercise training maintains cardiovascular health:signaling pathways involved and potential therapeutics

Exercise training has been widely recognized as a healthy lifestyle as well as an effective non-drug therapeutic strategy for cardiovascular diseases(CVD).Functional and mechanistic studies that employ animal exercise models as well as observational and interventional cohort studies with human participants,have contributed considerably in delineating the essential signaling pathways by which exercise promotes cardiovascular fitness and ***,this review summarizes the beneficial impact of exercise on multiple aspects of cardiovascular *** then discuss in detail the signaling pathways mediating exercise’s benefits for cardiovascular *** exercise-regulated signaling cascades have been shown to confer myocardial protection and drive systemic *** signaling molecules that are necessary for exercise-induced physiological cardiac hypertrophy have the potential to attenuate myocardial injury and reverse cardiac ***-regulated noncoding RNAs and their associated signaling pathways are also discussed in detail for their roles and mechanisms in exercise-induced cardioprotective ***,we address the exercise-mediated signaling pathways and molecules that can serve as potential therapeutic targets ranging from pharmacological approaches to gene therapies in *** also discuss multiple factors that influence exercise’s effect and highlight the importance and need for further investigations regarding the exercise-regulated molecules as therapeutic targets and biomarkers for CVD as well as the cross talk between the heart and other tissues or organs during *** conclude that a deep understanding of the signaling pathways involved in exercise’s benefits for cardiovascular health will undoubtedly contribute to the identification and development of novel therapeutic targets and strategies for CVD.

Defining and predicting deep remission in patients with perianal fistulizing Crohn's disease on anti-tumor necrosis factor therapy

Perianal fistulas can occur to up to one-third of patients with Crohn's disease(CD) leading to significant disabling disease and morbidity. Fistulising perianal CD treatment often necessitates a combined pharmacological and surgical approach. Anti-tumor necrosis factor(anti-TNF) therapy, particularly infliximab, has been shown to be very effective for both perianal and internal fistulising CD. Nevertheless, current data suggest that sustained remission and long-term complete fistula healing can be achieved in only 30% to 50% of patients. Moreover, these percentages refer mostly to clinical rather than deep remission, defined as endoscopic and radiologic remission, which is quickly emerging as the preferred goal of therapy. Unfortunately, the therapeutic options for perianal fistulising CD are still limited. As such, it would be of great value to be able to predict, and more importantly, prevent treatment failure in these patients by early and continued optimization of antiTNF therapy. Similar to ulcerative colitis and luminal CD, recent data demonstrate that higher infliximab concentrations are associated with better clinical outcomes in patients with perianal fistulising CD. This suggests that therapeutic drug monitoring and a treatto-trough therapeutic approach may emerge as the new standard of care for optimizing anti-TNF therapy in patients with perianal fistulising CD.

This work describes the development,optimisation and validation of an analytical method for the rapid determination of 17 priority pharmaceutical compounds and endocrine disrupting chemicals(EDCs).Rather than studying compounds from the same therapeutic class,the analyses aimed to determine target compounds with the highest risk potential(with particular regard to Scotland),providing a tool for further monitoring in different water *** was based on a systematic environmental risk assessment approach,using consumption data;wastewater treatment removal efficiency;environmental occurrence;toxicological effects;and pre-existing regulatory *** process highlighted 17 compounds across various therapeutic classes,which were then quantified,at environmentally relevant concentrations,by a single analytical *** determination was achieved using a single-step solid phase extraction(SPE)procedure followed by high-performance liquid chromatography with tandem mass spectrometry(HPLC-MS/MS).The fully optimised method performed well for the majority of target compounds,with recoveries>71%for 15 of 17 *** limits of quantification for most target analytes(14 of 17)ranged from 0.07 ng/L to 1.88 ng/L in river *** utility of this method was then demonstrated using real water samples associated with a rural hospital/*** compounds were targeted and detected,with the highest levels found for the analgesic,paracetamol(at up to 105,910 ng/L in the hospital discharge).This method offers a robust tool to monitor high priority pharmaceutical and EDC levels in various aqueous sample matrices.
