A basic helix-loop-helix transcription factor,figα,is one of the earliest marker genes of oocyte differentiation in *** the present study,we made figαknockout medaka by CRISPR/Cas9,expecting aborted progress of oogenesis,to see if differentiation of somatic ovarian tissues is ***αknockout male gonads differentiated normally into testes with functional *** females,on the other hand,were sterile;there are oocytes only up to *** growing oocytes in diplotene were *** phenotype was already apparent at 10 days after hatching,when diplotene oocytes start to develop in the ***,several putative target genes of figαwere not expressed in the mutant female *** studies showed that medaka lacking germ cells have morphologically abnormal gonads and female to male sex reversal ***αknockout female gonads differentiated morphologically into ovaries,suggesting that in medaka,figαknockout prevents oogenesis progress into diplotene stage,and ovarian differentiation do not need developed oocytes beyond pachytene *** ovarian phenotype reminded us of"pachytene checkpoint",which blocks meiotic progress into diplotene when chromosomal abnormalities are ***,we further analyzed phenotype of oocytes in the mutant *** number of oocytes at each meiotic stage suggested that oogenesis normally proceeded up to pachytene *** number of apoptotic oocytes in the knockout ovaries was ***,we did not detect any abnormal pairings or unrepaired double-strand breaks by *** together,figαknockout likely stop oocyte growth at or around pachytene via unknown mechanisms other than"pachytene checkpoint"and arrested oocytes remained in the *** is in contrast to figαknockout mice where all oocytes underwent apoptosis quickly after birth.

Improving the nutritional values of yellow mealworm Tenebrio molitor(Coleoptera: Tenebrionidae) larvae as an animal feed ingredient: a review

Yellow mealworm larvae(YML;Tenebrio molitor) are considered as a valuable insect species for animal feed due to their high nutritional values and ability to grow under different substrates and rearing conditions. Advances in the understanding of entomophagy and animal nutrition over the past decades have propelled research areas toward testing multiple aspects of YML to exploit them better as animal feed sources. This review aims to summarize various approaches that could be exploited to maximize the nutritional values of YML as an animal feed ingredient. In addition, YML has the potential to be used as an antimicrobial or bioactive agent to improve animal health and immune function in production animals. The dynamics of the nutritional profile of YML can be influenced by multiple factors and should be taken into account when attempting to optimize the nutrient contents of YML as an animal feed ingredient. Specifically, the use of novel land-based and aquatic feeding resources, probiotics, and the exploitation of larval gut microbiomes as novel strategies can assist to maximize the nutritional potential of YML. Selection of relevant feed supplies, optimization of ambient conditions, the introduction of novel genetic selection procedures, and implementation of effective post-harvest processing may be required in the future to commercialize mealworm production. Furthermore, the use of appropriate agricultural practices and technological improvements within the mealworm production sector should be aimed at achieving both economic and environmental sustainability. The issues highlighted in this review could pave the way for future approaches to improve the nutritional value of YML.

Magnetic field design is essential for the operation of Hall *** study focuses on utilizing a genetic algorithm to optimize the magnetic field configuration of SPT70.A 2D hybrid PIC-DSMC and channel-wall erosion model are employed to analyze the plume divergence angle and wall erosion rate,while a Farady probe measurement and laser profilometry system are set up to verify the simulation *** results demonstrate that the genetic algorithm contributes to reducing the divergence angle of the thruster plumes and alleviating the impact of high-energy particles on the discharge channel wall,reducing the erosion by 5.5%and 2.7%,*** analysis indicates that the change from a divergent magnetic field to a convergent magnetic field,combined with the upstream shift of the ionization region,contributes to the improving the operation of the Hall thruster.

Tuberculosis(TB)remains a leading cause of death among infectious diseases,particularly in poor *** infections,multidrug-resistant and ex-tensively drug-resistant TB strains,as well as the coexistence of chronic illnesses such as diabetes mellitus(DM)greatly aggravate TB morbidity and ***[particularly type 2 DM(T2DM)]and TB have converged making their control even more *** contemporary global epidemics,TB-DM behaves like a syndemic,a synergistic confluence of two highly prevalent diseases.T2DM is a risk factor for developing more severe forms of multi-drug resistant-TB and TB recurrence after preventive *** a bidirectional relationship exists between TB and DM,it is necessary to concurrently treat both,and promote recommendations for the joint management of both *** are also some drug-drug interactions resulting in adverse treatment outcomes in TB-DM patients including treatment failure,and *** addition,autophagy may play a role in these ***,the TB-DM comorbidities present several health challenges,requiring a focus on multidisciplinary collaboration and integrated strategies,to effectively deal with this double *** effectively manage the comorbidity,further screening in affected countries,more suitable drugs,and better treatment strategies are required.

Ripening and rot:How ripening processes influence disease susceptibility in fleshy fruits

Fleshy fruits become more susceptible to pathogen infection when they ripen;for example,changes in cell wall properties related to softening make it easier for pathogens to infect *** need for high-quality fruit has driven extensive research on improving pathogen resistance in important fruit crops such as tomato(Solanum lycopersicum).In this review,we summarize current progress in understanding how changes in fruit properties during ripening affect infection by *** changes affect physical barriers that limit pathogen entry,such as the fruit epidermis and its cuticle,along with other defenses that limit pathogen growth,such as preformed and induced defense *** plant immune system also protects ripening fruit by recognizing pathogens and initiating defense responses involving reactive oxygen species production,mitogen-activated protein kinase signaling cascades,and jasmonic acid,salicylic acid,ethylene,and abscisic acid *** phytohormones regulate an intricate web of transcription factors(TFs)that activate resistance mechanisms,including the expression of pathogenesis-related *** tomato,ripening regulators,such as RIPENING INHIBITOR and NON_RIPENING,not only regulate ripening but also influence fruit defenses against ***,members of the ETHYLENE RESPONSE FACTOR(ERF)family play pivotal and distinct roles in ripening and defense,with different members being regulated by different *** also discuss the interaction of ripening-related and defense-related TFs with the Mediator transcription *** the ripening processes in climacteric and non-climacteric fruits share many similarities,these processes have broad applications across fruiting *** research on the individual contributions of ERFs and other TFs will inform efforts to diminish disease susceptibility in ripe fruit,satisfy the growing demand for high-quality fruit and decrease food waste and related economic losses.

Machine learning algorithm partially reconfigured on FPGA for an image edge detection system

Optimal combination of the correction model and parameters for the precision geometric correction of UAV hyperspectral images

Nowadays,with the rapid development of quantitative remote sensing represented by high-resolution UAV hyperspectral remote sensing observation technology,people have put forward higher requirements for the rapid preprocessing and geometric correction accuracy of hyperspectral *** optimal geometric correction model and parameter combination of UAV hyperspectral images need to be determined to reduce unnecessary waste of time in the preprocessing and provide high-precision data support for the application of UAV hyperspectral *** this study,the geometric correction accuracy under various geometric correction models(including affine transformation model,local triangulation model,polynomial model,direct linear transformation model,and rational function model)and resampling methods(including nearest neighbor resampling method,bilinear interpolation resampling method,and cubic convolution resampling method)were ***,the distribution,number,and accuracy of control points were analyzed based on the control variable method,and precise ground control points(GCPs)were *** results showed that the average geometric positioning error of UAV hyperspectral images(at 80 m altitude AGL)without geometric correction was as high as 3.4041 m(about 65 pixels).The optimal geometric correction model and parameter combination of the UAV hyperspectral image(at 80 m altitude AGL)used a local triangulation model,adopted a bilinear interpolation resampling method,and selected 12 edgemiddle distributed *** correction accuracy could reach 0.0493 m(less than one pixel).This study provides a reference for the geometric correction of UAV hyperspectral images.

Chinese plum(Prunus salicina Lindl.)originates from China and makes a large contribution to the global production of *** ***‘Wushancuili'has a green coloration and high fruit quality and is economically important in eliminating poverty and protecting ecology in the Yangtze River Three Gorges ***,rain-induced cracking(rain-cracking,literally skin cracking caused by rain)is a limitation to‘Wushancuili'fruit production and causes severe *** study reported a high-quality‘Wushancuili'genome assembly consisting of a 302.17-Mb sequence with eight pseudo-chromosomes and a contig N50 of 23.59 Mb through the combination of Illumina sequencing,Pacific biosciences HiFiⅢsequencing,and high-throughput chromosome conformation capture technology.A total of 25109 protein-coding genes are predicted and 54.17%of the genome is composed of repetitive sequences.‘Wushancuili'underwent a remarkable orthoselection during *** identification revealed that loss-of-function in four core MYB10 genes results in the anthocyanin deficiency and absence of red color,revealing the green coloration due to the residual high chlorophyll in fruit ***,the occurrence of cracking is assumed to be closely associated with cell wall modification and frequently rain-induced pathogen enrichment through transcriptomic *** loss of MYB10 genes might render fruit more susceptible to pathogen-mediated cracking by weakening the epidermal strength and reactive oxygen species(ROS)*** findings provided fundamental knowledge regarding fruit coloration and rain-cracking and will facilitate genetic improvement and cultivation management in Chinese plums.
