Probing hydroxyl radical generation from H2O2 upon plasmon excitation of gold nanorods using electron spin resonance： Molecular oxygen-mediated activation

Gold nanostructures are among the noble metal nanomaterials being intensely studied due to their good biocompatibility, tunable localized surface plasmon resonance （SPR）, and ease of modification. These properties give gold nano- structures many potential chemical and biomedical applications. Herein, we demonstrate the critical role of oxygen activation during the decomposition of hydrogen peroxide （H202） in the presence of photoexcited gold nanorods （AuNRs） by using electron spin resonance （ESR） techniques. Upon SPR excitation, 02 is activated first, and the resulting reactive intermediates further activate H202 to produce ,OH. The reactive intermediates exhibit singlet oxygen-like （102-1ike） reactivity, indicated by 102-specific oxidation reaction, quenching behaviors, and the lack of the typical 102 ESR signal. In addition, by using the antioxidant sodium ascorbate （NaA） as an example, we show that hydroxyl radicals from H202 activation can induce much stronger NaA oxidation than that in the absence of H202. These results may have significant biomedical implications. For example, as oxidative stress levels are known to influence tumorigenesis and cancer progression, the ability to control redox status inside tumor microenvironments using noble metal nanostructures may provide new strategies for regulating the metabolism of reactive oxygen species and new approaches for cancer treatment.

Repurposing small molecule drugs and drug candidates is considered as a promising approach to revolutionise the treatment of snakebite *** this study,we investigated the inhibiting effects of the small molecules varespladib(nonspecific phospholipase A2 inhibitor),marimastat(broad spectrum matrix metalloprotease inhibitor)and dimercaprol(metal ion chelator)against coagulopathic toxins found in Crotalinae(pit vipers)snake *** from Bothrops asper,Bothrops jararaca,Calloselasma rhodostoma and Deinagkistrodon acutus were separated by liquid chromatography,followed by nanofractionation and mass spectrometry identification undertaken in *** of the venom toxins were then subjected to a high-throughput coagulation assay in the presence of different concentrations of the small molecules under *** venom toxins were mostly identified as phospholipases A2,while procoagulant venom activities were mainly associated with snake venom metalloproteinases and snake venom serine *** was found to effectively inhibit most anticoagulant venom effects,and also showed some inhibition against procoagulant ***,marimastat and dimercaprol were both effective inhibitors of procoagulant venom activities but showed little inhibitory capability against anticoagulant *** information obtained from this study aids our understanding of the mechanisms of action of toxin inhibitor drug candidates,and highlights their potential as future snakebite treatments.

Rapid and Reproducible Dibutylation Derivatization Coupled with Ultra-High Performance Liquid Chromatography-Tandem Mass Spectrometry for the Simultaneous Determination of Dopamine, Norepinephrine and 5-Hydroxytryptamine in Rat Brain Microdialysates

A rapid and reproducible method has been developed for the simultaneous quantification of dopamine (DA), norepinephrine (NE), and serotonin (5-HT) in rat brain microdialysates. Derivatization was processed through reductive amination with butyraldehyde and sodium cyanoborohydride at 60℃ for 80 min incubation. Dibutylated monoamine neurotransmitters (MANTs) were directly analyzed with ultra-high-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). The chromatographic run time was shorter (2.1 min/sample) than previous studies [15] [18] [20]. Good linearity (R2 > 0.99) was obtained for DA, NE and 5-HT in the range of 25 - 5000 pg/mL, 5 - 1000 pg/mL and 2.5 - 500 pg/mL, respectively. Acceptable precision (CV, 8.5% - 13.4%) as well as accuracy (recovery, 94.1% - 106.8%) could be acquired by analysis of six batches of quality control samples (QCs) at four different concentrations, which demonstrated the reliability and reproducibility of current method. This method was successfully applied to the simultaneous determination of DA, NE and 5-HT in rat brain microdialysates, where basal levels as well as elevated levels after dosing with amphetamine (AMPH) were quantified for all three MANTs. This study provides a simple and rapid way to analyze MANTs in the biofluid in the future.

Infection with Toxoplasma gondii, is one of the most widespread zoonoses in the world. Congenital Toxoplasmosis (CT) is particularly risky due to its fetal complications. Sulfadiazine (SDZ) and Pyrimethamine (PYR) are usually used for CT treatment in Argentina, to prevent morbidity. Due to the lack of commercial pediatric formulations, these must be prepared in the hospital pharmacy. This is the first report of serum concentrations measures in pediatric CT therapy for this combination of drugs. A bioanalytical method was developed for identification and simultaneous quantification of SDZ and PYR by High Performance Liquid Chromatography (HPLC) with UV detection. The validated method was applied to residual serum samples obtained from 6 pediatric patients undergoing treatment with SDZ 42.20 a 93.70 mg/kg/day and PYR 0.77 a 2.70 mg/kg/day. Sample pretreatment consisted on a deproteinization step followed by centrifugation and then injection of supernatant. Limit of Detection (LOD) and Quantification (LOQ) were (0.17 ± 0.02 and 0.13 ± 0.02) μg/mL and (0.46 ± 0.01 and 0.36 ± 0.01) μg/mL for SDZ and PYR respectively, with an appropriate linear range.
