Transplanting human neural progenitor cells is a promising method of replenishing the lost neurons after spinal cord injury (SCI), but differentiating neural progenitor cells into the diverse types of mature functional spinal cord neurons in vivo is challenging. In this study, engineered human embryonic spinal cord-like tissues with dorsal and ventral neuronal characters (DV-SC) were generated by inducing human neural progenitor cells (hscNPCs) to differentiate into various types of dorsal and ventral neuronal cells on collagen scaffold in vitro. Transplantation of DV-SC into complete SCI models in rats and monkeys showed better therapeutic effects than undifferentiated hscNPCs, including pronounced cell survival and maturation. DV-SC formed a targeted connection with the host’s ascending and descending axons, partially restored interrupted neural circuits, and improved motor evoked potentials and the hindlimb function of animals with SCI. This suggests that the transplantation of pre-differentiated hscNPCs with spinal cord dorsal and ventral neuronal characteristics could be a promising strategy for SCI repair.

Maternal uterine immune cells,especially natural killer(NK)cells,are important for successful pregnancy,and an abnormal number or function of uterine NK cells is closely correlated with an impaired endometrial environment and ***,there are currently no noninvasive testing methods using reliable indicators that accurately predict uterine changes leading to ***,we confirmed that before implantation,menstrual blood(MB)from healthy donors had 70%CD49a^(+)tissue-resident NK(trNK)cells,the majority of which were CD49a^(+)Eomes+NK ***,MB from patients with recurrent spontaneous abortion(RSA)had many fewer CD49a^(+)trNK cells than MB from *** of uNK cell populations in MB may better predict an abnormal endometrial environment associated with miscarriage than peripheral blood(PB)***,we suggest using MB-and CD49a-related markers as a promising noninvasive way to evaluate endometrial status.

Timely programmed cell death(PCD) of the tapetum supplying nutrients to microspores is a prerequisite for normal pollen *** we identified a unique mutant of rice(Oryza sativa L.),pollen sterility(post),which showed aborted pollens accompanied with extra-large *** to failure of timely PCD of tapetal cells,post exhibited abnormal pollen wall patterning and defective pollen *** map-based cloning,we identified a causal gene,POST,encoding a novel protein which is ubiquitously localized in *** in situ hybridization showed that POST is highly detected in the tapetum and microspores at stages 8 and *** analysis indicated that POST could function as an important regulator of the metabolic process involved in tapetal *** with wild-type rice,post mutant has an increased cell number resulting from elevated expression of cell cycle associated genes in grain *** of POST inhibits grain size in wild type,while appropriate expression of POST in post mutant can recover the seed fertility but has little effect on the large grains,illustrating that fine-tuning of POST expression could be a potential strategy for rice yield *** connection between cell division and cell death conferred by POST provides novel insights into the understanding of the tapetal PCD process.

Unraveling the mechanisms of synapse formation and axon regeneration: the awesome power of C. elegans genetics

Since Caenorhabditis elegans was chosen as a model organism by Sydney Brenner in 1960's, genetic studies in this organism have been instrumental in discovering the function of genes and in deciphering molecular signaling network. The small size of the organism and the simple nervous system enable the complete reconstruction of the first connectome. The stereotypic developmental program and the anatomical reproducibility of synaptic connections provide a blueprint to dissect the mechanisms underlying synapse formation. Recent technological innovation using laser surgery of single axons and in vivo imaging has also made C. elegans a new model for axon regeneration. Importantly, genes regulating synaptogenesis and axon regeneration are highly conserved in function across animal phyla. This mini-review will summarize the main approaches and the key findings in understanding the mechanisms underlying the development and maintenance of the nervous system. The impact of such findings underscores the awesome power of C. elegans genetics.

FoxP3^(+)regulatory T cells(Tregs)play crucial roles in peripheral immune *** addition,Tregs that reside or accumulate in nonlymphoid tissues,called tissue Tregs,exhibit tissue-specific functions and contribute to the maintenance of tissue homeostasis and *** an experimental mouse model of crescentic glomerulonephritis induced by an anti-glomerular basement membrane antibody,Tregs started to accumulate in the kidney on day 10 of disease onset and remained at high levels(~30–35%of CD4^(+)T cells)during the late stage(days 21–90),which correlated with stable disease *** depletion on day 21 resulted in the relapse of renal dysfunction and an increase in Th1 cells,suggesting that Tregs are essential for disease control during the convalescence *** Tregs that accumulated in the kidney showed tissue Treg phenotypes,including high expression of GATA3,ST2(the IL33 receptor subunit),amphiregulin(Areg),and PPARγ.Although T-bet^(+) Tregs and RORγt^(+) Tregs were observed in the kidney,GATA3^(+) Tregs were predominant during the convalescence stage,and a PPARγagonist enhanced the accumulation of GATA3^(+) Tregs in the *** understand the function of specific genes in kidney Tregs,we developed a novel T cell transfer system to T cell-deficient *** experiment demonstrates that ST2,Areg,and CCR4 in Tregs play important roles in the accumulation of GATA3^(+) Tregs in the kidney and in the amelioration of renal *** data suggest that GATA3 is important for the recruitment of Tregs into the kidney,which is necessary for convalescence after renal tissue destruction.

An Institutional Model for Collaborative Disaster Risk Management in the Southern African Development Community(SADC) Region

This article presents and argues for a collaborative model for disaster risk management in the Southern African Development Community(SADC).The research employed a qualitative study through a literature review and empirical research through focus group interviews to realize its *** a key theory of multinational collaboration,neoliberal institutionalism—a subset of the international relations theory—was used to develop the SADC institutional collaborative *** model combined the theoretical,political,and technical dimensions of collaboration to enhance buy-in for the disaster risk management and reduction function of *** model demonstrates the need for a multidisciplinary approach to achieving disaster risk management and reduction in the SADC and elsewhere,if the developmental objectives of disaster risk reduction are to be realized without interference in the domestic affairs of the member *** model is therefore grounded in seeking consensus and cooperation among cooperating states in a quest to ensure national implementation of the regional framework on disaster risk reduction.

Kinesin is a superfamily of microtubule-based motor proteins that move directionally along microtubules and involved in various essential biological processes in plant ***,few kinesins have been functionally identified in plant embryo and *** function in embryogenesis and seed formation is largely ***,we report NtKRP,a novel motor protein of kinesin-12 family with unique *** predominantly expresses in the tissues where cells are actively dividing,especially in embryo and embryonic root during seed *** RNAi lines displayed a reduction of cell number in meristematic zone and reduced length of embryonic *** size of matured embryo also obviously reduced and this resulted in smaller seeds,the reduced seed yield as ***,we identified a 60S ribosomal protein NtRPL17 as NtKRP interacting ***17was universally,but preferentially expressed in embryo and active-dividing *** phenotypes of NtRPL17 RNAi lines strongly resemble that of NtKRP RNAi lines,indicating they functions in the same *** experiments verified that NtKRP and CDKA:1 were regulated as a couple of co-expressed genes in ***;1 that possess kinase activity could bind to NtKRP at its phosphorylation sites,suggesting NtKRP activity could be regulated by CDKA;1 ***,we show evidences for that cell division delayed at G2-M transition phase,which led to the cell number decreased in embryos of NtKRP-and NtRPLl 7-silenced *** also revealed mat the tail domain of NtKRP that is usually involved in binding cargo lacks of consensus CDKA;1 phosphorylation sites,but can also interact with CDKA;*** unique feature implies its specific regulatory mechanism of the novel NtKRP compared to other members in kinesin-12 *** probably first carries its regulator CDKA;1 to the targeting sites and is then regulated by CDKA;1 phosphorylation for playing its active role in embryogenesis.
