Tobacco(Nicotiana tabacum)and tomato(Solanum lycopersicum)are two major economic crops in *** mosaic virus(TMV;genus Tobamovirus)is the most prevalent virus infecting both ***,some widely cultivated tobacco and tomato cultivars are susceptible to TMV and there is no effective strategy to control this ***-protection can be a safe and environmentally friendly strategy to prevent viral ***,stable attenuated TMV mutants are *** this study,we found that the substitutions in the replicase p126,arginine at position 196(R^(196))with aspartic acid(D),glutamic acid at position 614(E^(614))with glycine(G),serine at position 643(S^(643))with phenylalanine(F),or D at position 730(D^(730))with S,significantly reduced the virulence and replication of ***,only the mutation of S^(643) to F reduced the RNA silencing suppression activity of TMV p126.A double-mutant TMV-E614G-S643F induced no visible symptom and was genetically stable through six successive passages in tobacco ***,our results showed that TMV-E614G-S643F double-mutant could provide effective protection against the wild-type TMV infection in tobacco and tomato *** study reports a promising mild mutant for cross-protection to control TMV in tobacco and tomato plants.

The firmness of table grape berries is a crucial quality parameter. Despite extensive research on postharvest fruit softening, its precise molecular mechanisms remain elusive. To enhance our comprehension of the underlying molecular factors, we initially identified differentially expressed genes(DEGs) by comparing the transcriptomes of folic acid(FA)-treated and water-treated(CK) berries at different time points. We then analyzed the sequences to detect alternatively spliced(AS) genes associated with postharvest softening. A total of 2,559 DEGs were identified and categorized into four subclusters based on their expression patterns, with subcluster-4 genes exhibiting higher expression in the CK group compared with the FA treatment group. There were 1,045 AS-associated genes specific to FA-treated berries and 1,042 in the CK-treated berries, respectively. Gene Ontology(GO) annotation indicated that the AS-associated genes in CK-treated berries were predominantly enriched in cell wall metabolic processes,particularly cell wall degradation processes. Through a comparison between treatment-associated AS genes and subcluster-4 DEGs, we identified eight genes, including Pectinesterase 2(VvPE2, Vitvi15g00704), which encodes a cell wall-degrading enzyme and was predicted to undergo an A3SS event. The reverse transcription polymerase chain reaction further confirmed the presence of a truncated transcript variant of VvPE2 in the FA-treated *** study provides a comprehensive analysis of AS events in postharvest grape berries using transcriptome sequencing and underscores the pivotal role of VvPE2 during the postharvest storage of grape berries.

Fxcessive forest exploitation significantly contributes to land degradation and the creation of human-made *** are sometimes ignored in studies on the biodiversity of coarse woody *** investigated whether the type of stump,*** stumps(naturally created)and cut stumps(formed during forestry operations)had an impact on the species composition and species diversity as well as due to this fact they can be characterized by plant *** research covered 728 spruce stumps(287 broken and 441 cut stumps)that were inhabited by lichens,liverworts,mosses and vascular plants in mountain forest belt(Karkonosze Mts.,Poland).The following types of microhabitats were included in the research:the upper surface and the lateral surface of a stump with both bark and *** are statistically significant compositional differences between the two types of stumps,which was demonstrated by ordination analyses and indicator species *** to the generalized linear models,the probability of occurrence increases in cut stumps in case of liverworts and vascular plants and also along the decomposition *** generalized linear mixed-effects model showed that there was a higher species richness of liverworts,mosses and vascular plants on the cut stumps and less drier but the reverse situation was in *** generalized additive models for their cover showed similar *** all of the taxonomic groups were affected by altitude both in terms of species composition and total cover on the *** concluded that cut stumps are an important type of deadwood for biodiversity and provide a convenient habitat,especially for many lichens and bryophytes.

In a tritrophic context of plant-insect-entomopathogen,plants play important roles in modulating the interaction of insects and their pathogenic ***,the influence of plants on the transmission of insect viruses has been mainly studied on baculoviruses and some RNA viruses,whereas the impact of plants on other insect viruses is largely ***,we identified a new densovirus infecting the green peach aphid Myzus persicae and tested whether and how host plants influence the transmission of the aphid *** complete single-stranded DNA genome of the virus,*** densovirus 2,is 5727 nt and contains inverted terminal *** and phylogenetic analysis indicated that the virus was distinct from other a few identified aphid *** virus abundance was detected highly in the intestinal tract of aphids,compared with the lower level of it in other tissues including head,embryo,and *** and pepper plants had no obvious effect on the vertical transmission and saliva-mediated horizontal transmission of the ***,the honeydew-mediated horizontal transmission among aphids highly depended on host plants(65%on cabbages versus 17%on peppers).Although the virus concentration in the honeydew produced by aphids between 2 plants was similar,the honeydew production of the infected aphids reared on peppers was dramatically *** together,our results provide evidence that plants influence the horizontal transmission of a new densovirus in an aphid population by modulating honeydew secretion of aphids,suggesting plants may manipulate the spread of an aphid-pathogenic densovirus in nature.

The application of anti-drift nozzles,such as air-induction nozzles,is the most common recommendation for reducing spray drift;To reduce the risk of coarse droplets bouncing and rolling produced by anti-drift nozzles,various types of adjuvants were screened by comparing their atomization performance,surface tension and contact angle,in order to identify favourable adjuvants that are compatible with anti-drift *** was found from the study results that the addition of 50%neem crude oil emulsifiable concentrate(neem oil),isomeric alcohol ethoxylates and BYK-405 all significantly decreased distribution span(S)and the percentage of droplet size less than 150μm(ΦVol_(<150μm))values,but significantly promoted the median volume(D_(50))*** the surface tension measurement results showed that all tested adjuvants significantly reduced the surface tension,while neem oil,FC4430 and silwetl-77 were observed with a most significant effects;Furthermore,all tested adjuvants except BYK-051N could also significantly decrease the contact angle,and the difference between neem oil,FC4430 and silwetl-77 was *** tunnel test results clearly demonstrated that the application of IDKA(a combination of air-induction nozzle IDK120-01 and neem oil)substantially decreased the drift deposition amount;In addition,the field experiments revealed that IDKA possessed a significantly improved deposition amount per unit area on canopy or bottom,leading to a more effective *** results suggest that the use of IDK120-01 nozzle and neem oil can effectively reduce spray drift,lessen surface tension and enhance spreading.

Mythimna separata is a notorious phytophagous pest which poses serious threats to cereal crops owing to the gluttony of the *** short neuropeptide F(sNPF)and its receptor sNPFR are involved in a diversity of physiological functions,especially in functions related to feeding in insects,it is a molecular target for pest ***,an sNPF and 2 sNPFRs were identified and cloned from *** analysis revealed that the sNPF and its receptors had a highly conserved RLRFamide C-terminus and 7 transmembrane domains,*** sNPF and its receptor genes were distributed across larval periods and tissues,but 2 receptors had distinct expression *** starvation-induced assay elucidated that sNPF and sNPFR expression levels were downregulated under food deprivation and recovered with subsequent *** interference knockdown of sNPF,sNPFR1,and sNPFR2 by injection of double-stranded RNA into larvae not only suppressed food consumption and increased body size and weight,but also led to decrease of glycogen and total lipid contents,and increase of trehalose compared with double-stranded green fluorescent protein ***,molecular docking was performed on the interaction mode between sNPFR protein and its ligand sNPF based on the 3-dimensional models constructed by AlphaFold;the results indicated that both receptors were presumably activated by the mature peptide *** results revealed that sNPF signaling played a considerably vital role in the feeding regulation of *** and represents a potential control target for this pest.

The cAMP-dependent protein kinase A(PKA)signaling pathway has long been considered critical for long-term memory(LTM)*** studies have mostly focused on the role of PKA signaling in LTM induction by multiple spaced conditioning with less attention to LTM induction by a single ***,we conducted behavioral-pharmacology,enzyme immunoassay and RNA interference experiments to study the role of the PKA signaling pathway in LTM formation in the agricultural pest Bactrocera dorsalis,which has a strong memory capacity allowing it to form a two-day memory even from a single conditioning *** found that either blocking or activating PKA prior to conditioning pretreatment affected multiple spaced LTM,and conversely,they did not affect LTM formed by single *** was further confirmed by enzyme-linked immunosorbent assay(ELISA)and silencing of the protein kinase regulatory subunit 2 and catalytic subunit *** together,these results suggest that activating PKA during memory acquisition helps to induce the LTM formed by multiple spaced conditioning but not by a single *** findings challenge the conserved role of PKA signaling in LTM,which provides a basis for the greater diversity of molecular mechanisms underlying LTM formation across species,as well as possible functional and evolutionary implications.

The rice stem borer(RSB),Chilo suppressalis,a notorious rice pest in China,has evolved a high resistance level to commonly used ***,a new anthranilic diamide insecticide,effectively controls multiple pests,including ***,the potential resistance risk of RSB to tetraniliprole is still *** this study,the tetraniliprole-selection(Tet-R)strain was obtained through 10 continuous generations of selection with tetraniliprole 30%lethal concentration(LC_(30)).The realized heritability(h^(2))of the Tet-R strain was 0.387,indicating that resistance of RSB to tetraniliprole developed rapidly under the continuous selection of *** Tet-R strain had a high fitness cost(relative fitness=0.53).We established the susceptibility baseline of RSB to tetraniliprole(lethal concentration at LC_(50)=0.727 mg/L)and investigated the resistance level of 6 field populations to *** tested strains that had resistance to chlorantraniliprole exhibited moderate-to high-level resistance to tetraniliprole(resistance ratio=27.7-806.8).Detection of ryanodine receptor(RyR)mutations showed that the Y4667C,Y4667D,I4758M,and Y4891F mutations were present in tested RSB field *** mutations were responsible for the cross-resistance between tetraniliprole and ***,the clustered regularly interspaced palindromic repeats(CRISPR)/CRISPR-associated protein 9-mediated genome-modified flies were used to study the contribution of RyR mutations to tetraniliprole *** order of contribution of a single RyR mutation to tetraniliprole resistance was Y4667D>G4915E>Y4667C~~I4758M>*** addition,the I4758M and Y4667C double mutations conferred higher tetraniliprole resistance than single Y4667C *** results can guide resistance management practices for diamides in RSB and other arthropods.
