Background: L-selectin (CD62L) is a cell surface adhesion molecule recently shown to play a critical role in determining endometrial receptivity and implantation in humans. By contrast, the L-selectin ligand is missing from the rodent endometrium. Interestingly, CD62L (L-selectin)-deficient BALB/c mice delivered significantly higher numbers of viable offspring than wild type controls via mechanisms yet to be defined. Methods: Nulliparous CD62L-deficient (8-10-week-old, n = 25) or wild type (n = 18) females were mated with 43 age-matched males. Animals were sacrificed at gestational day (GD) 9.5. Tissue samples were analyzed by immunostaining and flow cytometry. Results: Mating wild type and CD62L-deficient BALB/c mice revealed that the increased birth rate was due to the CD62L deficiency in females. Flow cytometric analysis demonstrated significant differences in the number of natural killer (NK) cells present in the uterus of pregnant CD62L- deficient mice compared to controls. Immunohistochemistry confirmed NK cell accumulation at the fetal-maternal interface. Discussion: Uterine NK cells have been shown to peak at GD 8-10 at the fetal-maternal interface. NK cells might regulate mouse fertility rates by facilitating development of the maternal spiral arteries, thereby stimulating the formation of larger vessels that facilitate intrauterine survival, however, their role is not obligate to spiral artery development. Conclusions: Diminished CD62L expression modified immune cell trafficking into the uterus of pregnant mice generating a microenvironment primarily dominated by NK cells resulting in improved embryonic survival rates.

The chitin is extracted from the cuttlebone of S. inermis and the mineral contents are predictable. The structure and degree of deacetylation of extracted cuttlebone chitin is dogged through Fourier Transform Infrared (FT-IR) spectroscopy. The extracted cuttlebone chitin is used as the substrate for the production of chitinase from Vibrio sp. The extra cellular proteins are concentrated by ammonium sulphate precipitation, dialysed and then purified by using gel (sephadex G-100) chromatography. Among the 40 fractions, only two fractions (active fractions) showed maximum absorbance at 280 nm, which are pooled, dialysed and free-dried. The enzyme activity (0.104 μmoles/ml) and molecular weight (50 - 60 kDa) of purified chitinase is also determined through SDS- PAGE. The optimal condition for chitinase activity is pH between 6.0 - 6.5 and 45℃.

AIM: To identify the altered gene expression patterns in squamous cell carcinoma of esophagus (ESCC) in relation to adjacent normal esophageal epithelium. METHODS: Total RNA was extracted using SV total RNA isolation kit from snap frozen tissues of ESCC samples and normal esophageal epithelium far from the tumor. Radio-labeled cDNA were synthesized from equal quantities of total RNAs of tumor and normal tissues using combinations of 24 arbitrary 13-mer primers and three different anchoring oligo-dT primers and separated on sequencing gels. cDNA with considerable different amounts of signals in tumor and normal tissue were reamplified and cloned. Using southern blot, the clones of each band were controlled for false positive results caused by probable heterogeneity of cDNA population with the same size. Clones that confirmed differential expression by slot blot selected for sequencing and northern analysis. Corresponding full-length gene sequences was predicted using human genome project data, related transcripts were translated and used for various protein/motif searches to speculate their probable functions. RESULTS: The 97 genes showed different levels of cDNA in tumor and normal tissues of esophagus. The expression of mal gene was remarkably down. regulated in all 10 surveyed tumor tissues. Akrlc2, a member of the aldoketo reductase 1C family, which is involved in metabolism of sex hormones and xenobiotics, was up-regulated in 8 out of 10 inspected ESCC samples. Rablla, RPL7, and RPL28 showed moderate levels of differential expression. Many other cDNAs remained to further studies. CONCLUSION: The mal gene which is switched-off in all ESCC samples can be considered as a tumor suppressor gene that more studies in its regulation may lead to valuable explanations in ESCC development. Akrlc2 which is upregulated in ESCC probably plays an important role in tumor development of esophagus and may be proposed as a potential molecular target in ESCC treatments. Differentia

The Ray Wu Society for Life Sciences (RWS) is a non-profit professional society of Chinese-American and Chinese life scientists with the goal to promote scientific communication among the scientists between USA and China and the life science research in China. The members of the RWS, now numbered more than 300, are a group of elite biologists who have reached the level of assistant professor or equivalent in their careers and are experts of their respective fields including molecular biology and genetics, cell and developmental biology, me- dicine, pharmacology, plant biology, and biotechnology. RWS was founded in early 1998 by a group of life scientists who have come from China during the last 22 years. With the encouragement of Dr. Chris Tan, the Director of the molecular biology Institute in Singapore, who proposed to set up a fund in the name of Prof. Ray Wu of Cornell university to recognize his contribution to the academic exchange between China and the USA. A quick positive response was reflected from American Chinese scholars with a further proposal of forming a professional society. This initiative spread rapidly and widely throughout the existing networks of Chinese life-science professionals.
