Aim:Serum amyloid A(SAA)is an important mammalian acute ***,weaim to investigate the effect of SAA on apoptosis and its mechanism of action inhuman amniotic WISH ***:The expression of formyl peptide receptor(FPRL1),which is reported as a SAA receptor,was tested using RT-PCR andligand binding assay with radio-labeled FPRL1 *** effect of SAA onproliferating cell population was evaluated by thymidine incorporation *** protein phosphorylation levels and caspase-3 activity were detected by West-ern blot ***:SAA inhibits thymidine incorporation in human amnioticWISH cells.A SAA-induced decrease of proliferating cell population was accom-panied with nuclear condensation and caspase-3 activation in WISH cells,sug-gesting that SAA induces WISH cell *** FPRL1 has been reportedas a SAA receptor,we investigated the effects of several FRPL1 agonists on aproliferating cell population in WISH *** the tested FPRL1 agonists,only SAA induced a decrease of proliferating cell population in WISH *** downstream signaling of SAA,we found that SAA stimulated extracellularsignal-regulated kinase and p38 kinase,which were not inhibited by pertussistoxin(PTX),ruling out the role of PTX-sensitive *** a SAA-induced decrease of proliferating cell population was not affected by PTX,sug-gesting that SAA inhibits WISH cell apoptosis in a PTX-sensitive G-protein-independent manner.A SAA-induced decrease of a proliferating cell populationwas completely blocked by PD98059 and SB203580,suggesting that mitogen-activated protein kinase activities are essentially required for the ***:SAA is a novel inducer for WISH cell apoptosis,and the PTX-insen-sitive pathway is involved in the process.

Canonical NF-κB signaling regulates satellite stem cell homeostasis and function during regenerative myoge nesis

Skeletal muscle regeneration in adults is attributed to the presence of satellite stem cells that proliferate,differentiate,and eventually fuse with injured ***,the signaling mechanisms that regulate satellite cell homeostasis and function remain less *** IKKp-mediated canonical NF-κB signaling has been implicated in the regulation of myogenesis and skeletal muscle mass,its role in the regulation of satellite cell function during muscle regeneration has not been fully ***,we report that canonical NF-κB signaling is induced in skeletal muscle upon *** cell-specific inducible ablation of IKKβattenuates skeletal muscle regeneration in adult *** ablation of IKKβalso reduces the number of satellite cells in injured skeletal muscle of adult mice,potentially through inhibiting their proliferation and *** also demonstrate that the inhibition of specific components of the canonical NF-κB pathway causes precocious differentiation of cultured satellite cells both ex vivo and in ***,our results highlight that the constitutive activation of canonical NF-κB signaling in satellite cells also attenuates skeletal muscle regeneration following injury in adult ***,our study demonstrates that the proper regulation of canonical NF-κB signaling is important for the regeneration of adult skeletal muscle.

Motilin and ghrelin,members of a structure-function-related hormone family,play important roles in gastrointestinal function,regulation of energy homeostasis and growth hormone *** observed episodic evolution in both of their prehormone gene sequences during primitive placental mammal evolution,during which most of the nonsynonymous changes result in radical *** note,a functional obestatin hormone might have only originated after this episodic evolution *** in placental mammal evolution,a series of biology complexities *** the same time the motilin and ghrelin prehormone genes,which play important roles in several of these processes,experienced episodic evolution with dramatic changes in their coding *** observations suggest that some of the lineage-specific physiological adaptations are due to episodic evolution of the motilin and ghrelin genes.

HER3-targeted therapeutic antibodies and antibody-drug conjugates in non-small cell lung cancer refractory to EGFR-tyrosine kinase inhibitors

Autoimmune hepatitis

Autoimmune hepatitis is a chronic liver disease putatively caused by loss of tolerance to hepatocyte- specific autoantigens. It is characterized by female predilection, elevated aminotransferase levels, autoantibodies, increased y-globulin or IgG levels and biopsy evidence of interface hepatitis. It is currently divided into types 1 and 2, based on expression of autoantibodies. Autoantigenic epitopes have been identified only for the less frequent type 2. Although autoimmune hepatitis occurs in childhood, this review focuses on disease in adults. In the absence of pathognomonic biomarkers, diagnosis requires consideration of clinical, biochemical, serological and histological features, which have been codified into validated diagnostic scoring systems. Since many features also occur in other chronic liver diseases, these scoring systems aid evaluation of the differential diagnosis. New practice guidelines have redefined criteria for remission to include complete biochemical and histological normalization on immunosuppressive therapy. Immunosuppression is most often successful using prednisone or prednisolone and azathioprine; however, the combination of budesonide and azathioprine for non-cirrhotic patients offers distinct advantages. Patients failing standard immunosuppression are candidates for alternative immunosuppressive regimens, yet none of the options has been studied in a randomized, controlled trial. Overlap syndromes with either primary sclerosing cholangitis or primary biliary cirrhosis occur in a minority. Liver transplantation represents a life-saving option for patients presenting with acute liver failure, severely decompensated cirrhosis or hepatocellular carcinoma. Transplant recipients are at risk for recurrent autoimmune hepatitis in the allograft, and de novo disease may occur in patients transplanted for other indications. BACKGROUND： The carcinogenesis of hepatocellular car- cinoma （HCC） is a multi-factorial, multi-step and complex process. Early diagnosis and effective treatments are of utmost importance. This review summarized the recent studies of on- cofetal glypican-3 （GPC-3）, a membrane-associated heparan sulfate proteoglycan, in the diagnosis and treatment of HCC. DATA SOURCES： English-language reports published from Iune 2001 to September 2014 were searched from MEDLINE. The key words searched included： GPC-3, biomarker, target and HCC. The sensitivity, specificity, positive and negative predictive values were extracted, and the effect of GPC-3 tar- geted therapy on HCC was also evaluated. RESULTS： GPC-3 plays a crucial role in HCC cell prolifera- tion and metastasis. It mediates oncogenesis involving signal- ing pathways during hepatocyte malignant transformation. GPC-3 expression is increased in atypical hyperplasia and cancerous tissues. GPC-3 levels in HCC patients are related to HBV infection, TNM stage, periportal cancerous embolus, and extrahepatic metastasis. The diagnostic accuracy of the combination of serum GPC-3 and alpha-fetoprotein in HCC is up to 94.3%. Down-regulation of GPC-3 with specific siRNA or anti-GPC-3 antibody alters cell migration, metastasis and invasion behaviors. The nude mice xenograft tumor growth is inhibited by silencing GPC-3 gene ***： Oncofetal GPC-3 is a highly specific biomark- er for the diagnosis of HCC and a promising target molecule for HCC gene therapy.

Targeting proliferation and survival pathways in head and neck cancer for therapeutic benefit

Head and neck squamous cell carcinomas (HNSCC) are common human malignancies with poor clinical outcomes. The 5-year survival rates for patients with advanced stage HNSCC have not changed appreciably in the past few decades, underscoring a dire need for improved therapeutic options. Recent studies have elucidated a key signaling axis, the EGFR-STAT3-Bcl-XL signaling axis, that is aberrantly activated in a majority of HNSCC and contributes to the proliferation and survival of malignant cells. Considerable effort is being placed on developing highly specific inhibitors of different components of this pathway. This review highlights the progress that is being made towards achieving potent inhibition of the EGFR-STAT3-Bcl-XL signaling axis in HNSCC and the promising therapeutic strategies that are currently under development for this disease.
