AIM:To evaluate the retinal safety of various doses of intravitreal triamcinolone acetonide(TA)in ***:Thirty New Zealand albino rabbits were divided into five groups(six animals each).In group 1(control group),each animal received a single intravitreal injection of 0.1 mL phosphate buffered *** groups2,3,4 and 5,each rabbit received a single intravitreal injection of 4,8,16 and 32mg of TA,*** dose was contained in 0.1 mL phosphate buffered *** ocular examinations were performed before the injection and on the 1st,3rd,10th and 17th postinjection days.A standard dark adapted electroretinogram(ERG)was obtained before injection and on the 3rd,10th and 17th post-injection ***17d,animals were sacrificed and their eyes prepared for pathological ***:By monitoring ERG as a functional index for the retina,intravitreal injection of 4mg TA showed no significant ERG *** doses of 8,16 and 32,hyper-abnormal responses in a-and b-waves of ERG were detected on the 3rd post-injection *** changes gradually returned back to normal limits *** examination of the retina of all animals showed no pathological ***:High doses of intravitreal TA seemed to have enhancing effects on the retinal function with gradual return to normal limits with no pathological changes detected in examined eyes.

AIM： To evaluate effect of hypertension on retinal ganglion cell（RGC） apoptosis, intraocular pressure（IOP）,and the activation of endothelin-1（ET-1） signaling pathway in central retinal artery（CRA） in ***： The experimental study was performed on20 male Sprague Dawley rats that were divided into control group, and hypertension groups. The hypertension was induced by subcutaneous deoxycorticoacetate（DOCA）10 mg/kg twice a week and administered 0.9% Na Cl solution daily for 2, 6, and 10 wk. Blood pressure（BP） was measured using animal BP analyzer. IOP was measured by handheld tonometry. Retinal tissue preparations by paraffin blocks were made after enucleation. The expression of ET-1, e NOS, ET-1 receptor A（ETRA）, ET-1receptor B（ETRB）, and phosphorylated myosin light chain kinase（MLCK）, and caldesmon（Ca D） in CRA and RGC apoptosis were evaluated through immunofluorescent staining method then observed using laser scanning confocal microscopy. RESULTS： BP significantly increased in all of the hypertension groups compared to control（P =0.001）.Peak IOP elevation（7.78±4.14 mm Hg） and RGC apoptosis（576.15±33.28 Au） occurred on 2wk of hypertension. ET-1expression（1238.6±55.1 Au） and e NOS expression（2814.2±70.7 Au） were found highest in 2wk of hypertension,although the ratio of ET-1/e NOS decreased since *** peak expression in 10 wk of hypertension（1219.4 ±6.3 Au）, while ETRBsignificantly increased only in 2 weeks group（1069.2 ±9.6 Au）. The highest MLCK expression（1190.09±58.32 Au）, Ca D（1670.28±18.36 Au）were also found in 2wk of ***： Hypertension effects to activation of ET-1 signaling pathway significantly in CRA, elevation of IOP, and RGC apoptosis. The highest value was achieved at 2wk, which is the development phase of hypertension.

AIM: To evaluate the interchangeability of keratometric and asphericity measurements provided by three measurement systems based on different optical ***: A total of 40 eyes of 40 patients with a mean age of 34.1 y were included. In all cases, a corneal curvature analysis was performed with IOL-Master(IOLM), iDesign 2(ID2), and Sirius systems(SIR). Differences between instruments for flattest(K1) and steepest(K2) keratometric readings, as well as for magnitude and axis of corneal astigmatism were analyzed. Likewise, differences in asphericity(Q) between SIR and ID2 were also evaluated. RESULTS: Mean differences between devices for K1 were 0.20±0.21(P<0.001),-0.12±0.36(P=0.046) and-0.32±0.36 D(P<0.001) for the comparisons IOLM-SIR, IOLM-ID2 and SIR-ID2, respectively. The ranges of agreement for these comparisons between instruments were 0.41, 0.70, and 0.70 D. For K2, mean differences were 0.31±0.33(P<0.001),-0.08±0.43(P=0.265) and-0.39±0.38 D(P<0.001), with ranges of agreement of 0.65, 0.84, and 0.74 D. Concerning magnitude of astigmatism, ranges of agreement were in the limit of clinical relevance(0.49 D, P=0.011; 0.55 D, P=0.386; 0.43 D, P=0.05). In contrast, ranges of agreement were clinically relevant for astigmatic axis(26.68o, 33.83o and 18.37o, P≥0.121) and for Q between SIR and ID2(0.16, P<0.001). CONCLUSION: The keratometric corneal power, astigmatic axis and asphericity measurements provideby the three systems evaluated cannot be considered as interchangeable, whereas measurements of corneal astigmatism obtained with SIR and ID2 can be considered as interchangeable for clinical purposes.

AIM: To investigate the effect of 0.01% atropine sulphate eye gel on myopia progression and axial elongation in a 6-month treatment in ***: Totally 185 children aged 6-12 y with binocular myopia of 3.0 D or less in both eyes were enrolled in this prospective cohort study. The atropine group(n=125) received one drop of 0.01% atropine sulphate eye gel in each eye before bedtime daily. The control group included 60 matched children without drug intervention during the same period. The spherical equivalent and axial length was recorded at baseline and the sixth month of treatment. The efficacy was evaluated by the change of the spherical equivalent and axial length. Adverse events were also ***: The average spherical equivalent and axial length at baseline were not statistically significant between the atropine group(-1.64±0.80 D, 24.13±0.76 mm) and the control group(-1.59±0.94 D, 24.06±0.77 mm, P>0.05). After 6 mo, there was significantly difference in the spherical equivalent progression between the atropine and the control group(-0.27±0.33 vs-0.60±0.35 D, P<0.001),with a relative reduction of 55.0% in myopia progression. The increase in axial elongation in the atropine group was significantly less than control group(0.19±0.14 vs 0.26±0.14 mm, P<0.001), with a relative reduction of 26.9% in axial length. The 84.4% and 38.4% of the eyes progressed by less than 0.50 D and remained stable in the atropine group, compared with 51.7% and 4.2% in the control group. No adverse events were ***: Atropine sulphate eye gel 0.01% can slow down myopia progression and axial elongation in children with a 6-month treatment.

AIM: To identify the novel mutation alleles in the CYP1B1 gene of primary congenital glaucoma(PCG) patients at Shandong Province of China, and investigate their correlation with glaucomatous ***: The DNA from the peripheral blood of 13 congenital glaucoma patients and 50 ethnically matched healthy controls from the affiliated hospital of Qingdao University were extracted. The coding region of the CYP1B1 gene was amplified by PCR and direct DNA sequencing was performed. Disease causing-variants were analyzed by comparing the sequences and the structures of wild type and mutant CYP1B1 proteins by PyMOL ***: Two missense mutations, including A330 F caused by c.988 G>T&c.989 C>T, and R390H caused by c.1169 G>A, were identified in one of the 13 PCG patients analyzed in our study. A330F mutation was observed to be novel in the Chinese Han population, which dramatically altered the protein structure of CYP1B1 gene, including the changes in the ligand-binding pocket. Furthermore, R390H mutation caused the changes in heme-protein binding site of this gene. In addition, the clinical phenotype displayed by PCG patient with these mutations was more pronounced than other PCG patients without these mutations. Multiple surgeries and combined drug treatment were not effective in reducing the elevated intraocular pressure in this ***: A novel A330F mutation is identified in the CYP1B1 gene of Chinese PCG patient. Moreover, in combination with other mutation R390H, this PCG patient shows significant difference in the CYP1B1 protein structure, which may specifically contribute to severe glaucomatous phenotype.
