Studies from nearly 3 decades ago suggested that,in the central nervous system(CNS),myelination of axons by oligodendrocytes not only helps improve axonal conductivity but also stabilizes circuitry(Colello and Schwab,1994).Over the years,myelin sheaths produced by oligodendrocytes have been found to contain multiple molecules that are inhibitory to axonal growth(e.g.,MAG,NogoA,OMgp,Semaphorins)(Yiu and He,2006;Silver et al.,2014).After white matter injury in the adult CNS,myelin debris from damaged axons and dead oligodendrocytes accumulates in the forming glial scar and exposes these myelin-associated axon growth-inhibitory molecules to the injured axonal stumps,thereby contributing to the inhibition of axonal *** development,CNS axons reach their postsynaptic targets and stop growing before oligodendrocytes appear and myelinate them(Foran and Peterson,1992;Dangata et al.,1996).Therefore,myelin-associated axon growth-inhibitory molecules interacting with already grown axons during myelination were thought to block axons from promiscuous sprouting and miswiring,thereby stabilizing neural circuitry in the CNS(Colello and Schwab,1994).

Modeling functional brain connections in methamphetamine and opioid abusers

Substance abuse has become a significant problem *** primary purpose of this study was to model the functional brain connectivity in methamphetamines(Meth)and opioids(Op)groups in comparison with the healthy control(HC)*** data were recorded in Meth(n=8),Op(n=12),and HC groups(n=12)in a closed-eye resting *** NeuCube model based on a spiking neural network was used for EEG data's functional brain connectivity *** NeuCube model was evaluated quantitatively and qualitatively with model descriptive *** results showed the average value of the neuron's activation level,spikes emitted from neurons,and clustering coefficient as a communication parameter in the HC group decreased from 1154,1103,0.026 to 1036,981,0.020 in the Op group and to 511,468,0.009 in the Meth group,*** changes in indices were significant according to the ANOVA test with p〈*** decrease in indices was more significant in the Meth group than in the Op *** research findings showed that the abuse of Meth and Op causes damage to the brain's functional *** damage included shortening and limiting communication routes and decreased neurons'ability to generate membrane potential and emit inhibitory and excitatory *** were reported to be more severe in the Meth group than in the Op group.

Striatal interneurons play a key role in modulating striatal-dependent behaviors, including motor activity and reward and emotional processing. Interneurons not only provide modulation to the basal ganglia circuitry under homeostasis but are also involved in changes to plasticity and adaptation during disease conditions such as Parkinson’s or Huntington’s disease. This review aims to summarize recent findings regarding the role of striatal cholinergic and GABAergic interneurons in providing circuit modulation to the basal ganglia in both homeostatic and disease conditions. In addition to direct circuit modulation, striatal interneurons have also been shown to provide trophic support to maintain neuron populations in adulthood. We discuss this interesting and novel role of striatal interneurons, with a focus on the maintenance of adult dopaminergic neurons from interneuronderived sonic-hedgehog.

Many patients with neurodegenerative diseases,such as Alzheimer’s(AD)and Parkinson’s(PD)diseases suffer from disease progression without any satisfying clinical intervention,likely due to our lack of knowledge on how normal aging impacts the pathogenic mechanisms of these debilitating diseases.A growing body of literature has emerged in recent years that clearly demonstrates the involvement of glycolipids in the protein-oligomerization of neurodegenerative *** hypothesize that changes in glycolipids composition are a common mechanism underlying the shift from healthy brain aging to the neuropathological processes of neurodegenerative diseases.
