A CASE-CONTROL STUDY OF THE ETIOLOGY OF LARYNGEAL CANCER IN LIAONING PROVINCE

A case-control study of laryngeal cancer was conducted in Liaoning Province between January 1991 and April 1992 among 100 subjects with newly diagnosed cancer and 100 controls. The results of one factor analysis, odd ratio (OR), and conditional logistic regression indicated that smoking was the first significant risk factor for laryngeal cancer with an OR value of 16.8. Tobacco smoking ranked the first with an OR value of 30.4. Other factors such as alcoholic indulgence, insufficient intake of vegetable and fruits in winter and spring, air pollution in room and working environment, and fatigue of vocal cords were closely associated with the occurrence of laryngeal cancer.

OBJECTIVE： To investigate the progress in finding, isolation and culture, proliferation and differentiation, and application in neural tissue engineering of adult stem cells （ASCs）. DATA SOURCES ： Using the terms ＂adult stem cells, nerve, tissue engineering＂, we searched the PubMed for adult stem calls-related studies published in English from January 2001 to August 2006. Meanwhile, we also performed a China National Knowledge Infrastructure （CNKI） search for homochronous correlative literatures on the computer by inputting the terms ＂adult stem cells, nerve, tissue engineering＂in Chinese. STUDY SELECTION ： The materials were firstly selected. Studies related to ASCs were involved and their full texts were searched for. Inclusive criteria：（1） Literatures about the sources, distribution, culture, proliferation and differentiation, and application in the repair of neural ASCs by tissue engineering. （2）Articles recommended either by randomized, blind or by other methods were not excluded. Exclusive criteria：（1）Embryonic stem cells. （2） Review, repetitive study, case report, Meta analysis. DATA EXTRACTION： Totally 1 278 articles related to ASCs were collected, 32 were involved and the other 1 246 were excluded. DATA SYNTHESIS ： Adult stem cell has the ability of self-renewal, unceasing proliferation and transdifferentiation. It has wide source, which does not involved in ethical problems. It has advantages over embryonic stem cell. Studies on the isolation and culture, induction and differentiation and application in neural ASCs by tissue engineering contribute to obtaining considerable ASCs, so as to provide experimental and theoretical bases for the application of ASCs in repairing nerve injury in clinical practice.CONCLUSION ： ASCs play a very important role in neural tissue engineering

Objective:To study the efficacy and safety of hepatic arterial infusion of Endostatin(YH-16,Endostar),combined with transcatheter arterial chemoembolization(TACE) on advanced hepatocellular ***:Thirty patients with advanced hepatocellular carcinoma were enrolled in the *** patients received hepatic arterial infusion of Endostar combined with *** efficacy was evaluated strictly after 1-2 cycles according to RECIST criteria and the value of AFP;quality of life(QOL) was evaluated according to Karnofsky *** effects were evaluated ***:29 cases' efficacy was evaluated among the total 30 *** KPS were significantly increased after the treatment(80.39 ± 8.37 vs 73.93 ± 9.22,P = 0.002).Compared with control group,the objective response rate(CR and PR) and the rate of AFP negative changed were significantly higher(P = 0.021,P = 0.046).The adverse effects were not ***:The QOL and preliminary efficiency of patients of advanced hepatocellular carcinoma may be improved by hepatic arterial infusion of Endostar combined with TACE,the rate of AFP negative changed were significantly higher too,and there are little of adverse *** is worthy to clinical generalization and further clinical observation.

AIM: To evaluate the effect of combined antisense oligonucleotides targeting midkine (MK-AS) and chemotherapeutic drugs [cisplatin(DDP), 5-fluorouracil (5-FU) and adriamycin (ADM)] on inhibition of HepG2 cell proliferation, and to analyze the efficacy of MK-AS used in combined ADM in in situ human hepatocellular carcinoma (HCC) model. METHODS: HepG2 cells were treated with MK-AS and/or chemotherapeutic drugs mediated by Lipofectin, and cell growth activity was determined by MTS assay. An in situ HCC model was used in this experiment. MK- AS, ADM and MK-AS + ADM were given intravenously for 20 d, respectively. The animal body weight and their tumor weight were measured to assess the effect of the combined therapy in vivo. RESULTS: Combined treatment with MK-AS reduced the IC50 of DDP, 5-FU and ADM in HepG2 cells. MK-AS significantly increased the inhibition rate of DDP, 5-FU and ADM. Additionally, synergism (Q 1.15) occurred at a lower concentration of ADM, 5-FU and DDP with combined MK-AS. Combined treatment with MK-AS and ADM resulted in the more growth inhibition on in situ human HCC model compared with treatment with chemotherapeutic drugs alone. CONCLUSION: MK-AS increases the chemosensitivity in HepG2 cells and in situ human HCC model, and thecombination of MK-AS and ADM has a much better in vitro and in vivo synergism.

Background Waardenburg syndrome type I （WS1） is an autosomal dominant disorder characterized by sensorineural hearing loss, pigmental abnormalities of the eye, hair and skin, and dystopia canthorum. The gene mainly responsible for WS1 is PAX3 which is involved in melanocytic development and survival. Mutations of PAX3 have been reported in familiar or sporadic patients with WS1 in several populations of the world except Chinese. In order to explore the genetic background of Chinese WS1 patients, a mutation screening of PAX3 gene was carried out in four WS1 pedigrees. Methods A questionnaire survey and comprehensive clinical examination were conducted in four Chinese pedigrees of WSI. Genomic DNA from each patient and their family members was extracted and exons of PAX3 were amplified by PCR. PCR fragments were ethanol-purified and sequenced in both directions on an ABl_Prism 3100 DNA sequencer with the BigDye Terminator Cycle Sequencing Ready Reaction Kit. The sequences were obtained and aligned to the wild type sequence of PAX3 with the GeneTool program. Results Two nonsense PAX3 mutations have been found in the study population. One is heterozygous for a novel nonsense mutation S209X. The other is heterozygous for a previously reported mutation in European population R223X. Both mutations create stop codons leading to truncation of the PAX3 protein. Conclusions This is the first demonstration of PAX3 mutations in Chinese WS1 patients and one of the few examples of an identical mutation of PAX3 occurred in different populations.
