Testicular vascularization at two locations in relation to hormonal levels,and pixel echotexture in bulls at different ages

Objective:To assess the hemodynamic changes that occurred in the main testicular artery at two different locations such as supra and marginal ones in bulls at three different ages.Methods:Eighteen Baladi bulls were divided according to their age into three different age groups:1 year[n=6;groupⅠwith bodyweight(380±10)kg],3 years[n=6;groupⅡwith bodyweight(570±10)kg],and 6 years old[n=6;groupⅢwith bodyweight(650±10)kg]Baladi bulls.Circumference of the scrotum,thickness of the mediastinum,testicular dimensions,vascularity,and hormonal levels were measured.Testicular hemodynamics were assessed by Doppler ultrasound scanning.Results:Testicular width,length,and volume were significantly different among the three age groups,with the highest in bulls of groupⅢ(P<0.05).The bulls in groupⅢhad significantly higher mediastinal line thickness and scrotal circumference than groupⅠ(P<0.05),but there was no significant difference between groupⅡand groupⅢ.Bulls in groupⅡshowed a decline in Doppler indices(resistance and pulsatility indexes)at both testicular branches compared to the other two groups(P<0.05).Supra and marginal end diastolic velocities in the three age groups were significantly different,with the highest in groupⅡ(P<0.05).Time average velocity(cm/s)was significantly elevated in groupⅡcompared to the other two groups(P<0.05).Both testicular echotextures were significantly decreased in groupⅡ(P<0.05).Estradiol and nitric oxide metabolites were significantly high in groupⅡcompared to the other two groups(P<0.05).Meanwhile,the level of plasma testosterone was maximum in groupⅢ(P<0.05).Positive correlations were found between supratesticular artery resistance and pulsatility indexes(r=0.81;P<0.001),while both resistance and pulsatility indexes had correlated negative with estradiol(r=-0.71 and r=-0.91;P<0.001),and nitric oxide metabolites(r=-0.92 and r=-0.72;P<0.001).Conclusions:Three-year old Baladi bull has the lesser Doppler indices with lesser echotexture and greater nitric oxide with estradiol concentrations,which d

Hesperidin attenuates arsenic trioxide-induced cardiac toxicity in rats

Objective:To explore the cardioprotective effect of hesperidin against arsenic trioxide-induced cardiac toxicity in rats.Methods:Cardiac toxicity was induced by oral administration of 4 mg/kg arsenic trioxide for 30 days.Hematological,biochemical,electrocardiography,echocardiography,and histopathological examinations were performed.Results:Hesperidin decreased the neutrophil-to-lymphocyte ratio,calcium,creatine kinase-myoglobin binding,lactate dehydrogenase,IL-6,and lipid peroxidation,as well as increased sodium and potassium concentration and superoxide dismutase and catalase activity in arsenic trioxide-intoxicated rats.Moreover,it reduced peak systolic velocity and end-diastolic velocity while increasing heart rate.Arsenic trioxide-induced histopathological damage to cardiac tissue was prominently alleviated by hesperidin treatment.Conclusions:Hesperidin attenuates arsenic trioxide-induced cardiac toxicity in rats.Therefore,it can be further explored as a cardioprotective agent.

Bones are organs of the skeletal system, providing shape, mechanical support, and protection to the body and facilitating the movement. In addition, bones contribute to the mineral homeostasis of the body and have recently been found to participate in endocrine regulation of energy metabolism. Mechanical properties and in vivo biodegradability of Mg-Zr-Y-Nd-La magnesium alloy produced by a combined severe plastic deformation

Permanent implants are going to be replaced by the implementation of biodegradable magnesium-based implants in fixation of internal bone fractures because of many concerns associated with conventional implants.However,biodegradable magnesium-based biomaterials exhibit higher biodegradation rate and low mechanical properties which are the main challenges.This work aims to almost overcome both disadvantageous by grain refining of a WE43 magnesium alloy containing 93.04 wt% Mg,4.12 wt% Y,2.15 wt% Nd,0.43 wt% Zr,and 0.26 wt%La.In this study,the consequences of combined severe plastic deformation(SPD) on the mechanical properties,microstructure,and in vivo degradation behavior of WE43 magnesium alloy were investigated.To do so,WE43 magnesium alloy was chosen and processed through multipass equal channel angular pressing(ECAP) at 330℃ for up to four passes followed by an extrusion process.The results showed that higher strength and hardness with minimum ductility less was obtained in the sample processed via two-pass ECAP followed by extrusion.In vivo biodegradation experiments showed higher degradation rate for the unprocessed coarse-grained(CG) WE43 sample.The two-pass ECAP and extruded sample with ultrafine-grained(UFG) structure exhibited the lowest in vivo biodegradation rate besides appropriate mechanical properties.It may be concluded that the WE43 magnesium alloy processed via two-pass ECAP and extrusion could be a very promising candidate for biodegradable implants from both mechanical and biocorrosion viewpoints.

Background: Chondrocytes in the growth plate(GP) undergo increases in volume during different cascades of cell differentiation during longitudinal bone growth. The volume increase is reported to be the most significant variable in understanding the mechanism of long bone growth.Methods: Forty-five postnatal Sprague-Dawley rat pups, 7-15 days old were divided into nine age groups(P7-P15). Five pups were allocated to each group. The rats were sacrificed and tibia and metatarsal bones were harvested. Bone lengths were measured after 0, 24, 48, and 72 hours of ex vivo incubation. Histology of bones was carried out, and GP lengths and chondrocyte densities were determined.Results: There were significant differences in bone length among the age groups after 0 and 72 hours of incubation. Histological sectioning was possible in metatarsal bone from all age groups, and in tibia from 7-to 13-day-old rats. No significant differences in tibia and metatarsal GP lengths were seen among different age groups at 0 and 72 hours of incubation. Significant differences in chondrocyte densities along the epiphyseal GP of the bones between 0 and 72 hours of incubation were observed in most of the age groups.Conclusion: Ex vivo growth of tibia and metatarsal bones of rats aged 7-15 days old is possible, with percentage growth rates of 23.87 ± 0.80% and 40.38 ± 0.95%measured in tibia and metatarsal bone, respectively. Histological sectioning of bones was carried out without the need for decalcification in P7-P13 tibia and P7-P15 metatarsal bone. Increases in chondrocyte density along the GP influence overall bone elongation.

Ultrasound guided testicular fine needle aspiration in buck (Capra hircus)-An animal model

Objective:To establish ultrasound guided testicular fine needle aspiration (TFNA) as well as to assess the effectiveness of uni-directional (UD) and multi-directional (MD) TFNA in buck according to testicular cells, echotexture and gross changes of testicle, age of buck was considered.Methods:A total of 120 samples were collected with both directions (UD, n=60) and (MD,n=60) suction from testis of 10 apparently healthy bucks. All slides were stained with May-Grünwald-Giemsa and examined under light microscope with 1000× magnifications to count spermatogenic cells, spermatozoa and sertoli cells. The percentage of spermatozoa and sertoli cells were expressed as spermatic index and sertoli cell index.Results:Results revealed no difference in the presence of various spermatogenic and sertoli cells in cell cluster of slides made either unidirectional TFNA or multidirectional TFNA. Early spermatids were the most numerous, followed by late spermatids, primary spermatocytes, spermatogonia. Sertoli cell index was higher in TFNA smears of young bucks prepared from (7-13 mo) of age and spermatic index was higher in adult bucks (14-24 mo) of age. No echogenic change was observed in the echotexture of testisafter TFNA.Conclusion: It seems that TFNA has no serious ill effect on the buck testis when uni-direction aspiration is performed. Moreover, the possibility to standardize this method might provide a greater impulse to the clinical diagnostics of male animal infertility.

Endoscopic monitoring the estrous phase of Arabian mares with assaying its biopsy

Objective:To evaluate the changes occur in the vagina, cervix and endometrium during the pre- ovulatory period in Arabian mares by using endoscope as a diagnostic tool as well as the availability of taking an endoscopic biopsy during this stage.Methods:Ten cyclic and healthy arabian mares were subjected to ultrasound scanning of their ovaries to estimate the diameter of the preovulatory follicles, collection of blood samples for estimating the hormonal levels (Estradiol-17β and progestrone) for confirming the estrous phase, endoscopic monitoring their vagina, cervix and endometrium, the availability of taking biopsical samples and histological examination of the obtained samples.Results:The obtained data revealed that, the mean diameter of the preovulatory follicles was (3.95±0.34) cm, the plasma estradiol and progestrone levels were (85.98±5.26) pg/mL and (0.73±0.23) ng/mL, respectively, endoscopic monitoring the vagina, cervix and endometrium revealed hyperemic vaginal mucous membrane, soft and opened cervical ostium with abundant estrous mucous and folding, hyperemic and edematous appearance of the endometrium, respectively and the histological features of the biopsical endometrial samples revealed that, The lamina propria-sub mucosa was packed with simple or branched tubular glands lined with simple columnar epithelium showing oval or rounded dark nuclei with different degrees of secretory activities of the glands.Conclusions: The endoscope proved to be a reliable tool for confirming the estrous phase in mares and monitoring the gross features of the vagina, the cervix and the endometrium, in addition to the availability of taking biopsical smears.

Aldehyde dehydrogenase activity helps identify a subpopulation of murine adipose-derived stem cells with enhanced adipogenic and osteogenic differentiation potential

AIM To identify and characterize functionally distinct subpopulation of adipose-derived stem cells(ADSCs).METHODS ADSCs cultured from mouse subcutaneous adipose tissue were sorted fluorescence-activated cell sorter based on aldehyde dehydrogenase(ALDH) activity, a widely used stem cell marker. Differentiation potentials were analyzed by utilizing immunocytofluorescece and its quantitative analysis.RESULTS Approximately 15% of bulk ADSCs showed high ALDH activity in flow cytometric analysis. Although significant difference was not seen in proliferation capacity, the adipogenic and osteogenic differentiation capacity was higher in ALDHHi subpopulations than in ALDHLo. Gene set enrichment analysis revealed that ribosome-related gene sets were enriched in the ALDHHi subpopulation. CONCLUSION High ALDH activity is a useful marker for identifying functionally different subpopulations in murine ADSCs. Additionally, we suggested the importance of ribosome for differentiation of ADSCs by gene set enrichment analysis.

In vitro and in vivo degradability,biocompatibility and antimicrobial characteristics of Cu added iron-manganese alloy

In recent years,iron(Fe)based degradable metal is explored as an alternative to permanent fracture fixation devices.In the present work,copper(Cu)is added in Fe-Mn system to enhance the degradation rate and antimicrobial properties.Fe-Mn-x Cu(x=0.9,5 and 10 wt.%)alloys are prepared by the meltingcasting-forging route.XRD analysis confirms austenite phase stabilization due to the presence of Mn and Cu.As predicted by Thermo-Calc calculations,Cu rich phase precipitations are noticed along the austenite grain boundaries.Degradation behaviours of Cu added Fe-Mn alloys are investigated through static immersion and electrochemical polarization where enhanced degradation is found for higher Cu added alloys.When challenged against E.Coli bacteria,the Fe-Mn-Cu alloy media extract shows a significant bactericidal effect compare to the base alloy.In vitro cytocompatibility studies,as determined using MG63 and MC3T3-E1 cell lines,indicate increased cell density as a function of time for all the alloys.When implanted in rabbit femur,the newly developed alloy does not show any kind of tissue necrosis around the implants.Better osteogenesis and higher new bone formation are observed with Fe-Mn-10 Cu alloy as evident from micro-computed tomography(μ-CT)and fluorochrome labelling.
