Flavonoids from mulberry leaves inhibit fat production and improve fatty acid distribution in adipose tissue in finishing pigs

This study evaluated the effects of flavonoids from mulberry leaves (FML) on plasma biochemical indices,serum activities of lipid metabolism-related enzymes,fat morphology,fatty acid composition,and lipid metabolism in different adipose tissues of finishing pigs.We used 120 Chinese hybrid barrows of Berkshire and Bama mini-pigs with an average initial body weight of 45.11±4.23 kg.The pigs were randomly assigned to five treatment groups and fed a control diet based on corn,soybean meal,and wheat bran or a control diet supplemented with 0.02%,0.04%,0.08%,or 0.16%FML.Each experimental group had six replicates (pens),with four pigs per pen.After a 7-d adaptation period,the feeding trial was conducted for 58 d.Blood and adipose tissue samples were collected from 30 pigs (one pig per pen)at the end of the test.The results showed that FML supplementation significantly decreased the feed intake to body gain ratio,the plasma concentrations of total cholesterol and free fatty acids,and the serum activity of 3-hydroxy-3-methylglutaryl coenzyme A reductase (linear or quadratic effects,P<0.05),and decreased the plasma triglyceride concentration (quadratic,P=0.07).Increasing FML supplementation increased the average daily gain and serum activities of lipoprotein lipase (linear and quadratic effects,P<0.05) and adipose triglyceride lipase (linear,P<0.05).Dietary FML supplementation decreased the adipocyte area in the dorsal subcutaneous adipose (DSA) tissue of finishing pigs (linear,P=0.05) and increased the adipocyte area in the visceral adipose tissue (quadratic,P<0.01).Increasing FML supplementation decreased the C20:1 content in DSA,abdominal subcutaneous adipose,and visceral adipose tissues of finishing pigs (P<0.05) and increased the C18:3n3 and n-3 PUFA contents(P<0.05).The lipid metabolism genes were regulated by the PPARγ-LXRa-ABCA1 signaling pathway,and their expressions differed in different adipose tissues.These findings suggest that FML improved growth performance,regulated lipid metabolism,inhibited fat production

Negative enthalpy alloys and local chemical ordering:a concept and route leading to synergy of strength and ductility

Solid solutions are ubiquitous in metals and alloys. Local chemical ordering(LCO) is a fundamental sub-nano/nanoscale process that occurs in many solid solutions and can be used as a microstructure to optimize strength and ductility. However, the formation of LCO has not been fully elucidated, let alone how to provide efficient routes for designing LCO to achieve synergistic effects on both superb strength and ductility. Herein, we propose the formation and control of LCO in negative enthalpy alloys. With engineering negative enthalpy in solid solutions, genetic LCO components are formed in negative enthalpy refractory high-entropy alloys(RHEAs). In contrast to conventional ‘trial-and-error' approaches, the control of LCO by using engineering negative enthalpy in RHEAs is instructive and results in superior strength(1160 MPa) and uniform ductility(24.5%) under tension at ambient temperature, which are among the best reported so far. LCO can promote dislocation cross-slip, enhancing the interaction between dislocations and their accumulation at large tensile strains; sustainable strain hardening can thereby be attained to ensure high ductility of the alloy. This work paves the way for new research fields on negative enthalpy solid solutions and alloys for the synergy of strength and ductility as well as new functions.

Categorizing methods for integrating machine learning with executable specifications

Deep learning(DL),which includes deep reinforcement learning(DRL),holds great promise for carrying out real-world tasks that human minds seem to cope with quite readily.That promise is already delivering extremely impressive results in a variety of areas.However,while DL-enabled systems achieve excellent performance,they are far from perfect.It has been demonstrated,in several domains,that DL systems can err when they encounter cases they had not hitherto encountered.Furthermore,the opacity of the produced agents makes it difficult to explain their behavior and ensure that they adhere to various requirements posed by human engineers.At the other end of the software development spectrum of methods,behavioral programming(BP) facilitates orderly system development using self-standing executable modules aligned with how humans intuitively describe desired system behavior.In this paper,we elaborate on different approaches for combining DRL with BP and,more generally,machine learning(ML) with executable specifications(ES).We begin by defining a framework for studying the various approaches,which can also be used to study new emerging approaches not covered here.We then briefly review state-of-the-art approaches to integrating ML with ES,continue with a focus on DRL,and then present the merits of integrating ML with BP.We conclude with guidelines on how this categorization can be used in decision making in system development,and outline future research challenges.

Combined Treatment with Bojungikgi-tang(Buzhong Yiqi Decoction) and Riluzole Attenuates Cell Death in TDP-43-Expressing Cells

Objective: To examine the effect of combined treatment with Bojungikgi-tang(BJIGT, Buzhong Yiqi Decoction) and riluzole(RZ) in transactive response DNA-binding protein 43(TDP-43) stress granule(SG) cells, a amyotrophic lateral sclerosis(ALS) cell line using transcriptomic and molecular techniques. Methods: TDP-43 SG cells were pretreated with BJIGT(100 μg/mL), RZ(50 μmol/L), and combined BJIGT(100 μg/mL)/RZ(50 μmol/L) for 6 h before treatment with lipopolysaccharide(LPS, 200 μmol/L). Cell viability assay was performed to elucidate cell toxicity in TDP-43 SC cells using a cell-counting kit-8(CCK8) assay kit. The expression levels of cell death-related proteins, including Bax, caspase 1, cleaved caspase 3 and DJ1 in TDP-43 SG cells were examined by Western blot analysis. The autophagy-related proteins, including pmTOR/mTOR, LC3b, P62, ATG7 and Bcl-2-associated athanogene 3(Bag3) were investigated using immunofluorescence and immunoblotting assays. Results: Cell viability assay and Western blot analysis showed that combined treatment with BJIGT and RZ suppressed LPS-induced cell death and expression of cell death-related proteins, including Bax, caspase 1, and DJ1(P<0.05 or P<0.01). Immunofluorescence and immunoblotting assays showed that combined treatment with BJIGT and RZ reduced LPS-induced formation of TDP-43 aggregates and regulated autophagy-related protein levels, including p62, light chain 3b, Bag3, and ATG7, in TDP-43-expressing cells(P<0.05 or P<0.01). Conclusion: The combined treatment of BJIGT and RZ might reduce inflammation and regulate autophagy dysfunction in TDP-43-induced ALS.

What can we learn from quality assurance badges in open-source software?

In the development of open-source software(OSS), many developers use badges to give an overview of the software and share some key features/metrics conveniently. Among various badges, quality assurance(QA) badges make up a large proportion and are the most prevalent because QA is of vital importance in software development, and ineffective QA may lead to anomalies or defects. In this paper, we focus on QA badges in open-source projects, which present quality assurance information directly and instantly,and aim to produce some interesting findings and provide practical implications. We collect and analyze 100000 projects written in popular programming languages from GitHub and conduct a comprehensive empirical study both inside and outside QA badges. Inside QA badges, we build a category classification for all QA badges based on the properties they focus on, which shows the types of QA badges developers use. Then,we analyze the frequency of the properties that QA badges focus on, and property combinations, too, which present their use status. We find that QA badges focus on various properties while developers give different preferences to different properties. The use status also differs between different programming languages. For example, projects written in C focus on Security to a great extent. Our findings also provide implications for developers and badge providers. Outside QA badges, we conduct a correlation analysis between QA badges and some software metrics that have potential relationships with code quality, contribution quality, and popularity. We find that QA badges have statistically significant correlations with various software metrics.

Topological magnetoelectric response in ferromagnetic axion insulators

The topological magnetoelectric effect （TME） is a hallmark response of the topological field theory,which provides a paradigm shift in the study of emergent topological phenomena.However,its direct observation is yet to be realized due to the demanding magnetic configuration required to gap all surface states.Here,we theoretically propose that axion insulators with a simple ferromagnetic configuration,such as the MnBi2Te4/（Bi2Te3）nfamily,provide an ideal playground to realize the TME.In the designed triangular prism geometry,all the surface states are magnetically gapped.Under a vertical electric field,the surface Hall currents give rise to a nearly half-quantized orbital moment,accompanied by a gapless chiral hinge mode circulating in parallel.Thus,the orbital magnetization from the two topological origins can be easily distinguished by reversing the electric field.Our work paves the way for direct observation of the TME in realistic axion-insulator materials.

ELK3-ID4 axis governs the metastatic features of triple negative breast cancer

Purpose:Cancer cell metastasis is a multistep process,and the mechanism underlying extravasation remains unclear.ELK3 is a transcription factor that plays a crucial role in regulating various cellular processes,including cancer metastasis.Based on the finding that ELK3 promotes the metastasis of triple-negative breast cancer(TNBC),we investigated whether ELK3 regulates the extravasation of TNBC by forming the ELK3-ID4 axis.ID4 functions as a transcriptional regulator that interacts with other transcription factors,inhibiting their activity and subsequently influencing various biological processes associated with cell differentiation,survival,growth,and metastasis.Methods:We assessed the correlation between the expression of ELK3 and that of ID4 in TNBCs using bioinformatics analyses,QRT-PCR,western blot analysis,luciferase reporter assays,and chromatin immunoprecipitation.Migration,adhesion,invasion,and lung metastasis assays were employed to determine whether the ELK3-ID4 axis regulates the metastatic features of TNBC.Results:We found that ELK3 binds directly to a binding motif close to the ID4 promoter to repress promoter activity.The expression of E-cadherin in TNBC was regulated by the ELK3-ID4 axis.In vitro and in vivo analyses showed that inhibiting ID4 expression in ELK3-knockdown MDA-MB-231(ELK3KD)cells restored the ability to extravasate and metastasize.Conclusion:The results indicate that the ELK3 regulates ID4 promoter activity,and that the ELK3-ID4 axis regulates the metastatic characteristics of TNBC cells.Additionally,the data suggest that the ELK3-ID4 axis regulates metastasis of TNBCs by modulating expression of E-cadherin.

Controllable multi-domain semantic artwork synthesis

We present a novel framework for the multidomain synthesis of artworks from semantic layouts.One of the main limitations of this challenging task is the lack of publicly available segmentation datasets for art synthesis.To address this problem,we propose a dataset called ArtSem that contains 40,000 images of artwork from four different domains,with their corresponding semantic label maps.We first extracted semantic maps from landscape photography and used a conditional generative adversarial network(GAN)-based approach for generating high-quality artwork from semantic maps without requiring paired training data.Furthermore,we propose an artwork-synthesis model using domain-dependent variational encoders for high-quality multi-domain synthesis.Subsequently,the model was improved and complemented with a simple but effective normalization method based on jointly normalizing semantics and style,which we call spatially style-adaptive normalization(SSTAN).Compared to the previous methods,which only take semantic layout as the input,our model jointly learns style and semantic information representation,improving the generation quality of artistic images.These results indicate that our model learned to separate the domains in the latent space.Thus,we can perform fine-grained control of the synthesized artwork by identifying hyperplanes that separate the different domains.Moreover,by combining the proposed dataset and approach,we generated user-controllable artworks of higher quality than that of existing approaches,as corroborated by quantitative metrics and a user study.

Remarkable N_(2)-selectivity enhancement of NH_(3)-SCR over HPMo modified MnCo-BTC@SiO_(2) catalyst

In this work,the phosphomolybdate(HPMo)modification strategy was applied to improve the N_(2) selectivity of Mn Co-BTC@SiO_(2)catalyst for the selective catalytic reduction of NO_(x),and further,the mechanism of HPMo modification on enhanced catalytic performance was explored.Among Mn Co-BTC@SiO_(2-x) catalysts with different HPMo concentrations,Mn CoBTC@SiO_(2)-0.75 catalyst exhibited not only the highest NH_(3)-SCR performance(95% at 200-300℃)but also the best N_(2)selectivity(exceed 80% at 100-300℃)due to the appropriate redox capacity,greater surface acidity.X-ray photoelectron spectrometer(XPS)and temperature programmed reduction of H_(2)(H_(2)-TPR)results showed that the modification with HPMo reduced the oxidation-reduction performance of the catalyst due to electron transfer from Mo^(5+)to Mn^(4+)/Mn^(3+)and prevent the excessive oxidation of ammonia adsorption species.NH_(3)temperature-programmed desorption of(NH_(3)-TPD)results showed that the modification with HPMo could significantly improve the surface acidity and NH_(3)adsorption,which enhance the catalytic activity and N_(2)selectivity.In-situ diffused reflectance infrared Fourier transform spectroscopy(in-situ DRIFTS)revealed that modification with HPMo increased significantly the amount of adsorbed NH_(3)species on the Bronsted acid site and C_(B)/C_(L),it suppressed the production of N_(2)O by inhibiting the production of NH species,the deep dehydrogenation of ammonia adsorption species.This study provided a simple design strategy for the catalyst to improve the low-temperature catalytic performance and N_(2)selectivity.

Nanoparticle Exsolution on Perovskite Oxides:Insights into Mechanism,Characteristics and Novel Strategies

Supported nanoparticles have attracted considerable attention as a promising catalyst for achieving unique properties in numerous applications,including fuel cells,chemical conversion,and batteries.Nanocatalysts demonstrate high activity by expanding the number of active sites,but they also intensify deactivation issues,such as agglomeration and poisoning,simultaneously.Exsolution for bottomup synthesis of supported nanoparticles has emerged as a breakthrough technique to overcome limitations associated with conventional nanomaterials.Nanoparticles are uniformly exsolved from perovskite oxide supports and socketed into the oxide support by a one-step reduction process.Their uniformity and stability,resulting from the socketed structure,play a crucial role in the development of novel nanocatalysts.Recently,tremendous research efforts have been dedicated to further controlling exsolution particles.To effectively address exsolution at a more precise level,understanding the underlying mechanism is essential.This review presents a comprehensive overview of the exsolution mechanism,with a focus on its driving force,processes,properties,and synergetic strategies,as well as new pathways for optimizing nanocatalysts in diverse applications.
