Repeated inoculation with rumen fluid accelerates the rumen bacterial transition with no benefit on production performance in postpartum Holstein dairy cows

Background The dairy cow’s postpartum period is characterized by dramatic physiological changes,therefore imposing severe challenges on the animal for maintaining health and milk *** dynamics of the ruminal microbiota are also tremendous and may play a crucial role in lactation *** aim to investigate the potential benefits of early microbial intervention by fresh rumen microbiota transplantation(RMT)and sterile RMT in postpartum dairy *** fistulated peak-lactation dairy cows were selected to be the donors for rumen fluid *** postpartum cows were divided into 3 groups as the transplantation receptors respectively receiving 10 L fresh rumen fluid(FR),10 L sterile rumen fluid(SR),or 10 L saline(CON)during 3 d after *** Production performance,plasma indices,plasma lipidome,ruminal microbiome,and liver transcriptome were *** fresh and sterile RMT,we found that the molar proportion of propionic acid was increased on d 7 in the FR and SR groups and the bacterial composition was also significantly changed when compared with the CON group.A similarity analysis showed that the similarities between the CON group and FR or SR group on d 7 were 48.40%or 47.85%,whereas the similarities between microbiota on d 7 and 21 in the FR and SR groups were 68.34%or 66.85%.Dry matter intake and feed efficiency were not affected by ***β-hydroxybutyrate concentration in the FR group was decreased and significantly different lipids mainly included phosphatidylcholine and lysophosphatidylcholine containing polyunsaturated fatty *** transcriptomics analysis indicated acutephase response pathways were upregulated in the SR *** Our study suggests that RMT can shorten the transition process of the ruminal microbiota of postpartum dairy cows with no benefit on dry matter intake or feed *** with rumen fluid may not be a useful approach to promote the recovery of postpartum dairy cows.

Halogenated natural products(HNPs)are considered to be emerging contaminants whose environmental distribution and fate are only incompletely ***,several persistent and bioaccumulative HNP groups,together with manmade polychlorinated biphenyls(PCBs)and polybrominated diphenyl ethers(PBDEs),were quantified in the blubber of nine sperm whales(Physeter macrocephalus)stranded on the coast of the Mediterranean Sea in *** naturally occurring polybrominated hexahydroxanthene derivatives(PBHDs;sum of TetraBHD and TriBHD)were the most prominent substance class with up to 77,000 ng/g *** mean PBHD content(35,800 ng/g blubber)even exceeded the one of PCBs(28,400 ng/g blubber),although the region is known to be highly contaminated with manmade *** on mean values,Q1∼PBDEs>MeO-BDEs∼2,2′-diMeO-BB 80 and several other HNPs followed with decreasing *** blubber samples contained an abundant compound whose molecular formula(C_(16)H_(19)Br_(3)O_(2))was verified using high-resolution mass *** only plausible matching isomer was(2S,4′S,9R,9′S)-2,7-dibromo-4′-bromomethyl-1,1-dimethyl-2,3,4,4′,9,9′-9,9′-hexahydro-1H-xanthen-9-ol(OH-TriBHD),a hydroxylated secondary metabolite previously detected together with TriBHD and TetraBHD in a sponge known to be a natural producer of *** estimated mean amount of the presumed OH-TriBHD was 3000 ng/g blubber,which is unexpectedly high for hydroxylated compounds in the lipids of marine mammals.

Maternal resveratrol improves the intestinal health and weight gain of suckling piglets during high summer temperatures: The involvement of exosome-derived microRNAs and immunoglobin in colostrum

Previous studies have shown that maternal resveratrol improved growth performance and altered the microbial composition of suckling piglets under hot summer conditions. However, it remains unclear how maternal resveratrol improves growth performance of suckling piglets during high summer temperatures. A total of 20 sows(Landrace × Large White; three parity) were randomly assigned to 2 groups(with or without 300 mg/kg resveratrol) from d 75 of gestation to d 21 of lactation during high ambient temperatures(from 27 to 30℃). The results showed that maternal resveratrol supplementation increased total daily weight gain of piglets under hot summer conditions, which is consistent with previous studies. Furthermore, we found that maternal resveratrol improved the intestinal morphology and intestinal epithelial proliferation in suckling piglets. Dietary resveratrol supplementation affected the characteristics of exosome-derived microRNAs(miRNAs) in sow colostrum, as well as the genes targeted by differentially produced miRNAs. MiRNAs are concentrated in the tight junction pathway. As a result, the expression of intestinal tight junction proteins was increased in suckling piglets(P < 0.05).Notably, maternal resveratrol increased the intestinal secretory immunoglobulin A(sIgA) levels of suckling piglets via colostrum immunoglobin(P < 0.05), which could increase the abundance of beneficial microbiota to further increase the concentration of short chain fatty acids(SCFA) in suckling piglets' intestine(P < 0.05). Finally, our correlation analysis further demonstrated the positive associations between significantly differential intestinal microbiota, intestinal sIgA production and SCFA concentrations,as well as the positive relation between total daily weight gain and intestinal health of suckling *** together, our findings suggested that maternal resveratrol could promote intestinal health to improve piglet growth during high summer temperatures, which might be associa

The present study was to evaluate the effect of trace minerals(Zn, Mn, and Cu) from complexed amino acid minerals(ZMCAA) and bis-glycinate chelated minerals(ZMCGly) in laying hen diets on performance,internal and external egg quality, yolk mineral deposition, intestinal morphometry, and bone characteristics. From 78 to 98 weeks of age, 400 White LSL-Lite strain laying hens were distributed in a randomized design with 4 treatments with 10 replicates per treatment. Treatments were distributed in a 2 ×2 factorial arrangement using either Zn, Mn, and Cu of ZMCAA or ZMCGly source at 2 levels: low(20, 20,and 3.5 mg/kg of Zn, Mn, and Cu, respectively) or high(40, 40, and 7 mg/kg of Zn, Mn, and Cu,respectively). The analysis of variance was performed, and in cases where differences were observed, the means were compared using Tukey?s test(P feed conversion ratio(P =0.05) compared to those fed ZMCGly. The ZMCAA supplementation showed higher albumen height(P =0.01), albumen weight(P = 0.01), and eggshell thickness(P < 0.01). The deposition of Zn(P < 0.01), Mn(P< 0.01), and Cu(P < 0.01) in the egg yolk was greater for hens received ZMCAA. Tibia weight(P = 0.04)and bone densitometry(P < 0.01) in the tibia were higher with ZMCAA supplementation. In the small intestine, ZMCAA resulted in longer villi(P = 0.02) and shorter crypt depth(P = 0.01) in the *** and ileum measurements were influenced by the level and source of trace minerals(P < 0.05).Laying hens fed ZMCAA exhibited superior performance, egg quality, deposition of trace minerals in the egg yolk, and bone density compared to hens fed ZMCGly. In this study, older laying hens supplemented with ZMCAA at lower levels demonstrated adequate levels of supplementation.

Gut microbiota intervention attenuates thermogenesis in broilers exposed to high temperature through modulation of the hypothalamic 5‑HT pathway

AIM Influence of chronic hyperglycemia on chemical coding of enteric neurons in stomach using pig as a model for human diabetic *** Ten pigs were divided into two groups: diabetic(D group,n = 5) and control(C group,n = 5). Pigs constituting the experimental group were given streptozotocin(150 mg/kg). Animals were euthanized six weeks after the induction of diabetes. The samples of stomach were collected from animals of both groups. The cryostat sections were processed for double immunofluorescence staining using primary antisera directed towards pan-neuronal marker(Hu C/D) proteins and/or neuronal isoform of nitric oxide synthase(n NOS),vasoactive intestinal peptide(VIP) and galanin(GAL). RESULTS In the control group in the myenteric ganglia(MG) of the corpus we have noted 22.28% ± 1.19% of n NOS positive neurons,while in diabetic group we have found 40.74% ± 2.22% of n NOS immunoreactive perikarya(increase by 82.85 %). In turn in the pylorus we have observed 15.91% ± 0.58% n NOS containing neurons in control animals and 35.38% ± 1.54% in the diabetes group(increase by 122.37%). In the MG of the antrum and submucosal ganglion(SG) in the corpus hyperglycemia did not cause statistically significant changes. With regard to VIP-positive cell bodies in the antrum MG in the control animals we have noted 18.38 ± 1.39% and 40.74% ± 1.77% in the experimental group(increase by 121.65%). While in the corpus we have observed 23.20% ± 0.23% in the control and 30.93% ± 0.86% in the diabetes group(increase by 33.31%). In turn in the pylorus VIP positive cells bodies constituted 23.64% ± 1.56% in the control group and 31.20% ± 1.10% in the experimental group(increase by 31.97%). In the submucosal ganglion in the corpus we have noted 43.61% ± 1.06% in the control animals and 37.00% ± 1.77% in the experimental group(decrease by 15.15%). The study was carried out in Matiranga Upazila,Bandarban district,which is considered one of Bangladesh’s major hill tract *** study aimed to identify present farming conditions,prospects,and problems and suitable solutions to the problems of fish *** participatory rapid appraisal tools such as focus group discussion,field visits,and farmers’interviews were used to collect relevant fish farming information in the study *** polyculture method was the most practised aquaculture *** the selected farms,eleven fish species were identified as culture *** transportation cost for seeds purchased from remote areas was high(11%of total seed cost).The average transportation-related mortality of fish fry and fingerlings was 7.2%and 2.8%.feed cost was found as the major cost(58.4%)in *** and revenue for fish production per decimal varied from BDT 311 to 3528 and BDT 23 to *** of vacant lake,youth and women engagement,integrated aquaculture,the desire of people to engage in aquaculture,and available human resources were found as significant *** major problems identified were lack of hatchery,high feed cost,lack of stakeholder linkage,lack of capital for fish farming,and lack of proper knowledge,guidelines,and consultancy for fish culture.A community-based aquaculture model was proposed to overcome the present challenges and problems in the study *** model may help to expand aquaculture and thus improve the livelihood and socio-economic status of the people of the hilly areas of Bangladesh.

Fishmeal is the most preferred source of protein in aquafeeds, but it is expensive and scarce. Hence, Achatina fulica meat meal (AFM), which is much less preferred for human consumption out of three species of African giant land snails, was tested as a fishmeal substitute for Clarias gariepinus growth. Five iso-nitrogenous and iso-calorific diets were formulated, in which AFM substituted fish meal at 0% (control or Diet A), 25% (Diet B), 50% (Diet C), 75% (Diet D) and 100% (Diet E). These dietary treatments were each replicated thrice in a completely randomized design experiment, using 36-L plastic tanks in which the fish were fed daily rations corresponding to 5% of their body weight, for 8 weeks. Water quality parameters in the tanks were monitored. Proximate analyses were conducted on the fish meal, snail meal and experimental diets before the feeding trials. Cost-benefit analysis of the different diets was performed. The crude protein content of AFM (69.18%) was significantly higher than that of fish meal (55.81%). There was no significant difference (P > 0.05) in the mean weight gain, specific growth rate, feed conversion ratio, protein efficiency ratio and survival rate in fish fed Diet A and Diet B. The best protein efficiency ratio (0.77) was recorded in fish fed Diet B. Furthermore, the survival rate of fish increased with increased levels of AFM substitution. Water quality parameters were within a suitable range for tropical fish culture, indicating that the AFM did not pollute the water. The fish fed 25% AFM diet significantly (P Clarias gariepinus diets at a 25% substitution level. The aquaculture industry can thus exploit the availability of this feed resource.
