＜正＞The steroid hormone estrogen （E） plays a crucial role during early pregnancy by coordinating a complex series of interactions between the implanting embryo and the uterus. The cellular actions of E are mediated through estrogen receptor （ER） proteins, which are hormone-inducible transcription factors. It is believed that hormone-occupied ER regulates the expression of specific gene networks in different cell types within the uterus and the products of these genes mediate the hormonal effects during early pregnancy. To identify these E-regulated genes, we utilized a delayed implantation mouse model in which embryo attachment to the uterus is dependent on administration of E to P-primed pregnant uteri. Using gene chip technology, we analyzed E-regulated gene expression profiles in these delayed uteri. We found that in response to E, expression of mRNAs corresponding to 56 known genes was up regulated at least two-fold in the P-treated delayed mouse uterus. The E-regulated genes included transcription factors, regulators of cell cycle and cell differentiation, proteases and others. We have confirmed the steroid receptor regulation of several of these genes and have examined the spatio-temporal expression of these genes in the uterus during early pregnancy. Among these genes, we have performed a detailed analysis of CCAAT/Enhancer Binding Protein （C/EBP beta）, a transcription factor that is rapidly induced in response to E in the P-treated delayed mice. Our study showed that C/EBP beta is a novel steroid hormone regulated marker of decidualization and is an essential functional signal for this process.

Hypertrichosis is one of the most common side effects of systemic cyclosporine A *** has been previously shown that cyclosporine A induces anagen and inhibits catagen development in *** the present study,to explore the mechanisms of cyclosporine A,we investigated the effects of cyclosporine A on hair shaft elongation,hair follicle cell proliferation,apoptosis,and mRNA expression of selected growth factors using an organ culture model of mouse *** this model,cyclosporine A stimulated hair growth of normal mouse vibrissae follicles by inhibiting catagen-like development and promoting matrix cell *** addition,cyclosporine A caused an increase in the expression of vascular endothelial growth factor（VEGF）,hepatocyte growth factor（HGF）,and nerve growth factor（NGF）,and inhibited follistatin *** findings provide an explanation for the clinically observed effects of cyclosporine A on hair *** mouse vibrissae organ culture offers an attractive model for identifying factors involved in the modulation of hair growth.

Glutamate triggers the expression of functional ionotropic and metabotropic glutamate receptors in mast cells

Mast cells are emerging as players in the communication between peripheral nerve endings and cells of the immune ***,it is not clear the mechanism by which mast cells communicate with peripheral *** previously found that mast cells located within healing tendons can express glutamate receptors,raising the possibility that mast cells may be sensitive to glutamate *** evaluate this hypothesis,we stimulated primary mast cells with glutamate and showed that glutamate induced the profound upregulation of a panel of glutamate receptors of both the ionotropic type(NMDAR1,NMDAR2A,and NMDAR2B)and the metabotropic type(mGluR2 and mGluR7)at both the mRNA and protein *** binding of glutamate to glutamate receptors on the mast cell surface was ***,glutamate had extensive effects on gene expression in the mast cells,including the upregulation of proinflammatory components such as IL-6 and *** also induced the upregulation of transcription factors,including Egr2,Egr3 and,in particular,*** extensive induction of FosB was confirmed by immunofluorescence *** receptor antagonists abrogated the responses of the mast cells to glutamate,supporting the supposition of a functional glutamate–glutamate receptor axis in mast ***,we provide in vivo evidence supporting a functional glutamate–glutamate receptor axis in the mast cells of injured ***,these findings establish glutamate as an effector of mast cell function,thereby introducing a novel principle for how cells in the immune system can communicate with nerve cells.

Participatory Evaluation of the Relative Livestock Populations and the Assessment of the Status and Impacts of Newcastle Disease in Rural Communities of Two Northeastern States, Nigeria

Livestock is an important component of food security in many developing nations. However, animal diseases continue to undermine animal production and public health efforts thus widening poverty gap. Unfortunately, the conventional and what seems to be inefficient “top to bottom” disease surveillance and control policies are heavily relied upon such that negative impacts of diseases are underestimated. Participatory disease surveillance (PDS) techniques using semi-structured questioning assisted by key informants targeting focus groups were employed in 60 randomly selected villages of two Nigerian States during the years 2012 to 2014. Haemagglutination and ELISA tests to detect antibodies to Newcastle disease (ND) were conducted on 950 poultry sera. The status and economic burden of Newcastle disease and the relative livestock populations and some ethno-veterinary practices of these livestock farming communities were brought to the fore. Poultry, goats, cattle, sheep and pigs in descending order were the major livestock species kept in the study areas to which Kendalls Coefficient of Concordance (W = 0.9) agreed strongly. Accordingly, ND, lousiness, fowl pox and coccidiosis with percentage scores of ND-52%, Fowl Pox-31%, lousiness-17% and Salmonellosis-10% were important causes of poultry morbidity and mortality. ND sero-prevalence was 39%, relative morbidity;mortality and case fatality rates of 95%, 78% and 82% respectively were appraised. Again ND, coccidiosis, ectoparasitism and fowl pox were reported as seasonal poultry diseases (W = 0.6). Solanum nodiflorum and Momordica balsalmina were used to treat ND. Major livestock kept, and the ND status and effects in poultry in these livestock farming communities are here reported. Institutionalization of PDS would better inform strategic livestock policy reforms, and improve national food security and diseases surveillance and reporting system in Nigeria.

Construction and Characterization of a Novel Bacmid AcBac-Syn Based on a Synthesized Baculovirus Genome

Baculoviruses are large DNA viruses which have been widely used as expression vectors and biological *** recombination and Bac-to-Bac system have been the main methods for manipulating the baculovirus ***, we generated a synthetic baculovirus Ac MNPV-WIV-Syn1 which fully resembled its parental virus Autographa californica multiple nucleopolyhedrovirus(Ac MNPV). Here, we report the modification of Ac MNPV-WIV-Syn1 into a novel bacmid, Ac Bac-Syn, which can be used as a backbone for Bac-to-Bac system. To achieve this, a vector contained a Lac Z:att Tn7 and egfp cassette was constructed, and recombined with a linearized Ac MNPV-WIV-Syn1 genome by transformation-associated recombination in yeast to generate bacmid Ac Bac-Syn. The bacmid was then transfected to insect cells and the rescued virus showed similar biological characteristics to the wild-type virus in terms of the kinetics of budded virus production, the morphology of occlusion bodies, and the oral infectivity in insect larvae. For demonstration, a red fluorescent protein gene Dsred was transposed into the att Tn7 site by conventional Bac-to-Bac method, and the transfection and infection assays showed that Ac Bac-Syn can be readily used for foreign gene insertion and *** Bac-Syn has several advantages over the conventional Ac MNPV bacmids, such as it contains an egfp reporter gene which facilitates visualization of virus propagation and titration;its DNA copy numbers could be induced to a higher level in E. coli;and the retaining of the native polyhedrin gene in the genome making it an attractive system for studying the functions of gene related to occlusion body assembly and oral infection.

Biodegradable zinc-iron alloys: Complex study of corrosion behavior,mechanical properties and hemocompatibility

Zn-Fe alloys have been extensively investigated in this study with a view to their application as biodegradable bone implants. Biogenic element zinc is a very appropriate metal because of the ideal degradation rate compared to those of Mg and Fe. Studied alloys were made by compressing metallic powders in a content ratio of 100% Zn,Zn-1% Fe, Zn-2% Fe, Zn-5% Fe and Zn-10% Fe and sintering at 350°C for 1 h. Prepared samples were examined by optical microscopy, SEM and XRD. Corrosion behavior, mechanical testing and hemocompatibility were observed subsequently. The electrochemical performance of such materials was studied in the simulated body fluids. The enhanced corrosion rate was observed for all samples after iron addition due to the micro-galvanic effect between the pure Zn and Zn;Fe intermetallic phase. The corrosion rate of the Zn-5% Fe alloyed sample was more than 20-times higher（2.89 mmpy） compared to the pure Zn. However, alloying with more than 5 wt %of iron diminished the mechanical performance of the material. Therefore, the performed mechanical and hemocompatibility tests showed acceptable biocompatibility of zinc and Zn-1% Fe and Zn-2% Fe samples.

Biodegradation of phenol by free and immobilized Acinetobacter *** PD12

A new phenol-degrading bacterium with high biodegradation activity and high tolerance of phenol, strain PD 12, was isolated from the activated sludge of Tianjin Jizhuangzi Wastewater Treatment Facility in China. This strain was capable of removing 500 mg phenol/L in liquid minimal medium by 99.6% within 9 h and metabolizing phenol at concentrations up to 1100 mg/L. DNA sequencing and homologous analysis of 16S rRNA gene identified PD12 to be an Acinetobacter sp. Polyvinyl alcohol （PVA） was used as a gel matrix to immobilize Acinetobacter sp. strain PDI2 by repeated freezing and thawing. The factors affecting phenol degradation of immobilized cells were investigated, and the results showed that the immobilized cells could tolerate a high phenol level and protected the bacteria against changes in temperature and pH. Storage stability and reusability tests revealed that the phenol degradation functions of immobilized cells were stable after reuse for 50 times or storing at 4℃ for 50 d. These results indicate that immobilized Acinetobacter sp. strain PD 12 possesses a good application potential in the treatment of phenol-containing wastewater.

Antibacterial and Antioxidant Activities of ZnO Nanoparticles Synthesized Using Extracts of Allium sativum,Rosmarinus officinalis and Ocimum basilicum

Evaluation of the anti-diabetic properties of Mucuna pruriens seed extract

Objective:To explore the antidiabetic properties of Mucuna pruriens(***).Methods: Diabetes was induced in Wistar rats by single intravenous injection of 120 mg/kg of alloxan monohydrate and different doses of the extract were administered to diabetic *** blood glucose level was determined using a glucometer and results were compared with normal and untreated diabetic *** acute toxicity was also determined in albino ***:Results showed that the administration of 5,10,20,30,40,50,and 100 mg/kg of the crude ethanolic extract of *** seeds to alloxan-induced diabetic rats(plasma glucose > 450 mg/dL) resulted in 18.6%,24.9%,30.8%,41.4%,49.7%,53.1%and 55.4%reduction,respectively in blood glucose level of the diabetic rats after 8h of treatment while the administration of glibenclamide (5 mg/kg/day) resulted in 59.7%*** administration of the extract resulted in a significant dose dependent reduction in the blood glucose level(P<0.001).It also showed that the antidiabetic activity of *** seeds resides in the methanolic and ethanolic fractions of the *** toxicity studies indicated that the extract was relatively safe at low doses,although some adverse reactions were observed at higher doses(8-32 mg/kg body weight),no death was ***,oral administration of *** seed extract also significandy reduced the weight loss associated with ***:The study clearly supports the traditional use of *** for the treatment of diabetes and indicates that the plant could be a good source of potent antidiabetic drug.
