Protein microarrays based on fluorescence detection have been widely utilized for high-throughput functional proteomic analysis. However, a drawback of such assays has been low sensitivity and narrow dynamic range, limiting their capabilities, especially for detecting low abundance biological molecules such as cytokines in human samples. Here, we present fluorescence-enhancing microarrays on plasmonic gold films for multiplexed cytokine detection with up to three orders of magnitude higher sensitivity than on conventional nitrocellulose and glass substrates. Cytokine detection on the gold plasmonic substrate is about one to two orders of magnitude more sensitive than enzyme-linked immunosorbent assay （ELISA） and can be multiplexed. A panel of six cytokines （Vascular endothelial growth factor （VEGF）, Interleukin 1β （IL-1β）, Interleukin 4 （IL-4）, Interleukin 6 （IL-6）, Interferon γ （IFN-γ）, and Tumor necrosis factor （TNF）） were detected in the culture media of cancer cells. This work establishes a new method of high throughput multiplexed cytokine detection with higher sensitivity and dynamic range than ELISA.

The relationship of the immune system and tumour cells is complex; although recognised that the immune system can protect the host against tumour development, the immune system also facilitates tumour progression through immune suppression. Pro-inflammatory mediators associated with chronic inflammation are responsible for the expansion and activation of myeloid derived suppressor cells(MDSCs); a heterogeneous group of cells that originates from myeloid progenitor cells but does not complete the final stages of differentiation. A causal relationship between chronic inflammation and tumour progression relies on the accumulation and maintenance of MDSCs as its linchpin; responsible for immunosuppression through the down-regulation of antitumour responses. MDSCs cause immunosuppression through a number of mechanisms; inhibiting the proliferation of CD4^+ and CD8^+ T cells, blocking natural killer cell activation and limiting dendritic cell maturation and function. As well as using various mechanisms to inhibit adaptive and immune responses, MDSCs also have non-immunological functions that aid tumour spread; including directly promoting tumour proliferation and metastasis by having an important role in tumour angiogenesis, secretion of matrix metalloproteinases and induction of epithelial-mesenchymal transition. Breast cancer is the most common cancer among women in the United Kingdom with 44540 new cases of invasive carcinoma in 2013 and results in the second highest cancer mortality rate in women, with 11600 deaths in 2012. Considering this, the need for novel therapeutic interventions is higher than ever. This review summarises the rationale for the targeting of MDSCs in breast cancer as a realistic avenue to increase survival from breast cancer.

SARS-CoV-2 causes the pandemic of COVID-19 and no effective drugs for this disease are available thus *** to the high infectivity and pathogenicity of this virus,all studies on the live virus are strictly confined in the biosafety level 3(BSL3)laboratory but this would hinder the basic research and antiviral drug development of SARS-CoV-2 because the BSL3 facility is not commonly available and the work in the containment is costly and *** this study,we constructed a reverse genetics system of SARS-CoV-2 by assembling the viral cDNA in a bacterial artificial chromosome(BAC)vector with deletion of the spike(S)*** of the cDNA into cells results in the production of an RNA replicon that keeps the capability of genome or subgenome replication but is deficient in virion assembly and infection due to the absence of S ***,such a replicon system is not infectious and can be used in ordinary biological *** confirmed the efficient replication of the replicon by demonstrating the expression of the subgenomic RNAs which have similar profiles to the wild-type *** mutational analysis of nsp12 and nsp14,we showed that the RNA polymerase,exonuclease,and cap N7 methyltransferase play essential roles in genome replication and sgRNA *** also created a SARS-CoV-2 replicon carrying a luciferase reporter gene and this system was validated by the inhibition assays with known anti-SARS-CoV-2 ***,such a one-plasmid system is biosafe and convenient to use,which will benefit both fundamental research and development of antiviral drugs.

On the basis of data mining,systematic pharmacology,molecular docking,and experiment validation,the oxidative-inflammatory molecular targets of Coicis Semen in the therapy of osteoarthritis(OA)were ***:The association rule analysis was effectively applied to highlight the correlation between Coicis Semen and oxidative inflammation *** random walk model was subsequently used to evaluate the clinical efficacy of Coicis *** pharmacology was used to predict network *** binding affinity of the active ingredient in Coicis Semen to the key target of OA was also successfully ***:Coicis Semen showed a significant reduction in oxidative-inflammatory indicators of OA.A total of 108 promising targets were predicted for the 24 bioactive compounds in Coicis *** target genes were considered core target *** enrichment analysis predicts that Coicis Semen may activate the interleukin(IL)-17,mitogen-activated protein kinase(MAPK),and nuclear factor kappa B(NF-kappa B)signaling *** docking demonstrated that stigmasterol,2-monoolein,sitosterol,and sitosterol alpha1 had free binding energies to oxidative and inflammatory targets(MAPK1,Estrogen Receptor 1[ESR1],and Peroxisome Proliferator-Activated Receptor Alpha[PPARA]).Both clinical trials and in vitro cell experiments revealed that Coicis Semen could increase ESR1 and PPAR-αlevels while decreasing MAPK1 ***:Coicis Semen has a remarkable anti-OA ***,the major components of Coicis Semen,including stigmasterol,sitosterol alpha1,sitosterol,and 2-monoolein,specifically inhibit MAPK1,ESR1,and PPARA to reduce the inflammatory response and oxidative damage in OA.

Objective: To evaluate the in vitro and in vivo effect of the Algerian propolis ethanolic extract(EEP) against Echinococcus granulosus(E. granulosus) infection. Methods: In vitro scolicidal activity of EEP was investigated on the protoscolices of hydatid cyst. This in vitro study was conducted by using an in vivo assay. BALB/c mice were inoculated with E. granulosus and treated with propolis for three months. Hydatid cysts development was assessed. Nitric oxide(NO), tumor necrosis factor alpha(TNF-α) production and inducible NO synthase, NF-κB, and TNF-α spleen expression were estimated by Griess method and immunofluorescence ***: Our study revealed that EEP has a high scolicidal activity against E. granulosus. Oral administration of EEP decreased TNF-α, NF-κB and inducible NO synthase expression in the spleen tissues in the CE+EEP group, in comparison with the CE group. Concomitantly, EEP treatment caused an important systemic decrease in NO and TNF-α levels. These findings are associated with the reduction of CE development. Conclusions: This is the first report demonstrating with interest the antihydatic and immunomodulatory effects of the Algerian EEP, suggesting its therapeutic potential for the hydatid disease treatment.
