To the Editor:China has the largest number of intracerebral hemorrhage(ICH)patients of any single country,[1]and efforts to mine,utilize,and share the relevant data are warranted.As a structured diagnosis and treatment pipeline for ICH has been demonstrated to improve patient outcomes,there is an urgent need to construct a large medical database to store relevant data and facilitate data mining,analysis,and sharing.However,traditional stroke databases are dominated by text data without comprehensive imaging data,and few studies have focused on the construction of databases on ICH.[2,3]To integrate multimodal data on ICH and standardize data processing and feedback,the Chinese Intracranial Hemorrhage Imaging Database(CICHID)was established by Peking Union Medical College Hospital in 2019 and is under continuous refinement.Differing from traditional text-based databases,CICHID is a large multimodal data warehouse implementing comprehensive clinical data storage and multidimensional imaging data analysis.

Circular RNAs(circRNAs)are a new and large group of non-coding RNA molecules that are abundantly expressed in the central nervous system.However,very little is known about their roles in traumatic brain injury.In this study,we firstly screened differentially expressed circ RNAs in normal and injured brain tissues of mice after traumatic brain injury.We found that the expression of circ Lphn3 was substantially decreased in mouse models of traumatic brain injury and in hemin-treated b End.3(mouse brain cell line)cells.After overexpressing circ Lphn3 in b End.3 cells,the expression of the tight junction proteins,ZO-1,ZO-2,and occludin,was upregulated,and the expression of mi R-185-5 p was decreased.In b End.3 cells transfected with mi R-185-5 p mimics,the expression of ZO-1 was decreased.Dual-luciferase reporter assays showed that circ Lphn3 bound to mi R-185-5 p,and that mi R-185-5 p bound to ZO-1.Additionally,circ Lphn3 overexpression attenuated the hemin-induced high permeability of the in vitro b End.3 cell model of the blood-brain barrier,while mi R-185-5 p transfection increased the permeability.These findings suggest that circ Lphn3,as a molecular sponge of mi R-185-5 p,regulates tight junction proteins'expression after traumatic brain injury,and it thereby improves the permeability of the blood-brain barrier.This study was approved by the Animal Care and Use Committee of Chongqing Medical University of China(approval No.2021-177)on March 22,2021.

Pyroptosis plays an important role in hemorrhagic stroke.Excessive endoplasmic reticulum stress can cause endoplasmic reticulum dysfunction and cellular pyroptosis by regulating the nucleotide-binding oligomerization domain and leucine-rich repeat pyrin domain-containing protein 3(NLRP3)pathway.However,the relationship between pyroptosis and endoplasmic reticulum stress after intraventricular hemorrhage is unclear.In this study,we established a mouse model of intraventricular hemorrhage and found pyroptosis and endoplasmic reticulum stress in brain tissue.Intraperitoneal injection of the selective GPR120 agonist TUG-891 inhibited endoplasmic reticulum stress,pyroptosis,and inflammation and protected neurons.The neuroprotective effect of TUG-891 appears related to inhibition of endoplasmic reticulum stress and pyroptosis activation.

BACKGROUND： Deep-brain stimulation has proven to be beneficial in the treatment of Parkinson＇s disease （PD） patients. OBJECTIVE： To investigate the effects of high-frequency stimulation （HFS） to the subthalamic nucleus （STN） on neuronal apoptosis and apoptosis-related gene expression in the substantia nigra pars compacta, and to analyze the neuroprotective effect of HFS-STN. DESIGN, TIME AND SETTING： Neuronal morphology experiments were performed in the Beijing Neurosurgical Institute from May to December in 2005. MATERIALS： Forty healthy, adult, Sprague Dawley rats were used to establish a PD model with a unilateral microinjection of 6-hydroxydopamine into two target areas of the right medial forebrain bundle. 6-hydroxydopamine was purchased from Sigma （USA）; high-frequency electrical stimulator was produced by World Precision Instruments （USA）; Terminal deoxynucleotidyl transferase dUTP nick end labeling （TUNEL） kit was a product of Nanjing Jiancheng Technology Co., Ltd. （China）; and Bcl-2 and Bax protein assay kit were purchased from Wuhan Boster Bioengineering Co., Ltd. （China）. METHODS： Forty rats were randomly divided into three groups. The stimulation group （n = 15） received HFS-STN on the day of PD modeling. The PD model group （n = 15） was used to establish the PD model. The control group （n = 10） was injected with normal saline containing 0.2 g/L ascorbic acid into two areas of the right medial forebrain bundle. MAIN OUTCOME MEASURES： Survival of dopaminergic neurons in the substantia nigra pars compacta was determined using Nissl staining. Apoptosis of dopaminergic neurons was detected using TUNEL techniques. Expression of anti-apoptotic protein, Bcl-2, and pro-apoptotic protein, Bax, were assayed by immunohistochemistry. RESULTS： Following 6-hydroxydopamine injection, the number of substantia nigra pars compacta neurons was reduced in the stimulation and PD model groups, compared to the control group. Mild hypothermia for treatment of diffuse axonal injury: a quantitative analysis of diffusion tensor imaging

Fractional anisotropy values in diffusion tensor imaging can quantitatively reflect the consistency of nerve fibers after brain damage, where higher values generally indicate less damage to nerve fibers. Therefore, we hypothesized that diffusion tensor imaging could be used to evaluate the effect of mild hypothermia on diffuse axona[ injury. A total of 102 patients with diffuse axonal injury were randomly divided into two groups： normothermic and mild hypothermic treatment groups. Patient＇s modified Rankin scale scores 2 months after mild hypothermia were significant- ly lower than those for the normothermia group. The difference in average fractional anisotropy value for each region of interest before and after mild hypothermia was 1.32-1.36 times higher than the value in the normothermia group. Quantitative assessment of diffusion tensor imaging indicates that mild hypothermia therapy may be beneficial for patients with diffuse axonal injury.

Studies have shown that downregulation of nuclear-enriched autosomal transcript 1(Neat1)may adversely affect the recovery of nerve function and the increased loss of hippocampal neurons in mice.Whether Neat1 has protective or inhibitory effects on neuronal cell apoptosis after secondary brain injury remains unclear.Therefore,the effects of Neat1 on neuronal apoptosis were observed.C57 BL/6 primary neurons were obtained from the cortices of newborn mice and cultured in vitro,and an oxygen and glucose deprivation cell model was established to simulate the secondary brain injury that occurs after traumatic brain injury in vitro.The level of Neat1 expression in neuronal cells was regulated by constructing a recombinant adenovirus to infect neurons,and the effects of Neat1 expression on neuronal apoptosis after oxygen and glucose deprivation were observed.The experiment was divided into four groups:the control group,without any treatment,received normal culture;the oxygen and glucose deprivation group were subjected to the oxygen and glucose deprivation model protocol;the Neat1 overexpression and Neat1 downregulation groups were treated with Neat1 expression intervention techniques and were subjected to the in oxygen and glucose deprivation protocol.The protein expression levels of neurons p53-induced death domain protein 1(PIDD1,a pro-apoptotic protein),caspase-2(an apoptotic priming protein),cytochrome C(a pro-apoptotic protein),and cleaved caspase-3(an apoptotic executive protein)were measured in each group using the western blot assay.To observe changes in the intracellular distribution of cytochrome C,the expression levels of cytochrome C in the cytoplasm and mitochondria of neurons from each group were detected by western blot assay.Differences in the cell viability and apoptosis rate between groups were detected by cell-counting kit 8 assay and terminal deoxynucleotidyl transferase dUTP nick-end labeling assay,respectively.The results showed that the apoptosis rate,PIDD1,caspase-2,and cleaved caspase-3 expressi

Traumatic brain injury(TBI)remains a major cause of disability among young adults in both civilian and military settings contributing to a high burden on healthcare systems(Badhiwala et al.,2019).Sequel of TBI,even mild injuries,include motor and sensory dysfunction,neurocognitive decline,neuropsychiatric complications,as well as increased risk of neurodegenerative and neurovascular events such as Alzheimer’s disease and stroke(Breunig et al.,2013;Burke et al.,2013;Li et al.,2017).Despite the acute nature of the insult in TBI,pathological changes in the traumatized brain are better recognized as a chronic rather than an acute neurological disease,a phenomenon that remains under-investigated.

Background: An epidemic of COVID-19 broke out in Wuhan, China, since December 2019. The ordinary medical services were hindered. However, the emergency cases, including aneurysmal subarachnoid hemorrhage (aSAH), still required timely intervention. Thus, it provoked challenges to the routine management protocol. In this study, we summarized our experience in the emergency management of aSAH (Beijing Tiantan Protocol, BTP) in Beijing, China.Methods: Demographic, clinical, and imaging data of consecutive emergency aSAH patients who underwent craniotomy clipping during the COVID-19 epidemic season were reviewed and compared with the retrospective period last year. Subgroup analysis was further performed to assess the outcomes of different screening results and several detailed protocols. Neurological outcomes were evaluated by the modified Rankin Scale (mRS).Results: A total of 127 aSAH were referred to our emergency department, and 42 (33.1%) underwent craniotomy clipping between January 20, 2020, and March 25, 2020. The incidence of preoperative hospitalized adverse events and the perioperative outcomes were similar (-0.1, 95% CI - 1.0 to 0.8,P = 0.779) to the retrospective period last year (January 2019-March 2019). After the propensity score matching (PSM), there were still no statistical differences in prognostic parameters between the two groups. Eight (19.0%) of the 42 individuals were initially screened as preliminary undetermined COVID-19 cases, in which 2 of them underwent craniotomy clipping in the negative pressure operating room (OR). The prognosis of patients with varied COVID-19 screening results was similar (F(2, 39) = 0.393,P = 0.678). Since February 28, 12 cases (28.6%) received COVID-19 nucleic acid testing (NAT) upon admission, and all showed negative. The false-negative rate was 0.0%. The preoperative hospitalized adverse events and postoperative prognosis were still similar between patients with and without COVID-19 NAT (-0.3, 95% CI - 1.4 to 0.9,P = 0.653).Conclusions: Our emergency surgery man

Background:A safe,effective,and ethically sound animal model is essential for preclinical research to investigate spinal medical devices.We report the initial failure of a porcine spinal survival model and a potential solution by fixating the spine.Methods:Eleven female Dutch Landrace pigs underwent a spinal lumbar interlaminar decompression with durotomy and were randomized for implantation of a medical device or control group.Magnetic resonance imaging(MRI)was performed before termination.Results:Neurological deficits were observed in 6 out of the first 8 animals.Three of these animals were terminated prematurely because they reached the predefined humane endpoint.Spinal cord compression and myelopathy was observed on postoperative MRI imaging.We hypothesized postoperative spinal instability with epidural hematoma,inherent to the biology of the model,and subsequent spinal cord injury as a potential cause.In the subsequent 3 animals,we fixated the spine with Lubra plates.All these animals recovered without neurological deficits.The extent of spinal cord compression on MRI was variable across animals and did not seem to correspond well with neurological outcome.Conclusion:This study shows that in a porcine in vivo model of interlaminar decompression and durotomy,fixation of the spine after lumbar interlaminar decompression is feasible and may improve neurological outcomes.Additional research is necessary to evaluate this hypothesis.
