Background Receptor activator of nuclear factor kappa B （NF-κB） ligand （RANKL） and osteoprotegerin （OPG） have been recently shown to play important roles in bone resorption. The aim of this study was to investigate the possible association between the expression of bone resorption regulators （RANKL and OPG） and inflammatory cell infiltration in chronic apical *** The samples of chronic periapical lesions （n=40） and healthy periapical tissues （n=10） were examined for immunohistochemical analysis of RANKL and OPG. Lesion samples were further analyzed for the inflammatory infiltration condition. The inflammatory cell infiltration was scored in relation to immunohistochemical reactivity for CD3, CD20 and *** The number of RANKL-positive cells and the ratio of RANKL/OPG in chronic apical periodontitis were significantly higher than those in healthy periapical tissues （P＜0.001）. The number of RANKL-positive cells was higher in lesions with severe inflammatory infiltration than in those with light inflammatory infiltration （P＜0.05）. Significantly increased RANKL expression was found with T lymphocytes （CD3＋）, macrophages （CD68＋） and B lymphocytes （CD20＋）infiltration （P＜0.05）. No association was found between the ratio of RANKL/OPG and inflammatory cell *** RANKL expression was increased with T, B lymphocytes and macrophages infiltration, respectively in chronic periapical lesions. RANKL appears to be closely related to periapical inflammatory infiltrates. The relative ratio of RANKL/OPG may be a key determinant of RANKL-mediated bone resorption.

Initial Stability Study of Calcium Phosphate Coated Dental Implants

Thin film of biodegradable calcium phosphate coated on threaded commercially pure titanium （ cp- Ti ） dental implants has been investigated as one of alternatives to eliminate the problem of the long- term instability of plasma sprayed HA coated implants. In order to compare in-vivo bone-to-implant response behavior among as-machined, HA coated and CMP coated groups, each group was implanted into New Zealand white mature male rabbits for 2 and 6 weeks, and then in-vivo biological behavior was examined in terms of H＆E staining. Initial stability and removable torques of implants were compared among three groups. Measured removable torque of CMP coated specimen at 6 weeks after inplantation was significantly higher than that of non-coated group, but slightly lower than that of HA coated group, without any inflammatory response at the surrounding of the implants. The initial stability （ ISQ value ; implant stability quotient ） of CMP coated specimen at 2 weeks after implantation was slightly lower than that of HA coated group and significantly higher than that of non-coated group. However, after 6 weeks, ISQ value of CMP coated group was slightly higher than that of HA coated group and significantly higher than that of non-coated group.

With the discovery of the pivotal role of macrophages in tissue regeneration through shaping the tissue immune microenvironment, various immunomodulatory strategies have been proposed to modify traditional biomaterials. Decellularized extracellular matrix (dECM) has been extensively used in the clinical treatment of tissue injury due to its favorable biocompatibility and similarity to the native tissue environment. However, most reported decellularization protocols may cause damage to the native structure of dECM, which undermines its inherent advantages and potential clinical applications. Here, we introduce a mechanically tunable dECM prepared by optimizing the freeze-thaw cycles. We demonstrated that the alteration in micromechanical properties of dECM resulting from the cyclic freeze-thaw process contributes to distinct macrophage-mediated host immune responses to the materials, which are recently recognized to play a pivotal role in determining the outcome of tissue regeneration. Our sequencing data further revealed that the immunomodulatory effect of dECM was induced via the mechnotrasduction pathways in macrophages. Next, we tested the dECM in a rat skin injury model and found an enhanced micromechanical property of dECM achieved with three freeze-thaw cycles significantly promoted the M2 polarization of macrophages, leading to superior wound healing. These findings suggest that the immunomodulatory property of dECM can be efficiently manipulated by tailoring its inherent micromechanical properties during the decellularization process. Therefore, our mechanics-immunomodulation-based strategy provides new insights into the development of advanced biomaterials for wound healing.

This study aimed to evaluate the efficacy of comprehensive therapy based on traditional Chinese medicine （TCM） patterns on older patients with chronic obstructive pulmonary disease （COPD） through a four- center, open-label, randomized controlled trial. Patients were divided into the trial group treated using conventional western medicine and Bu-Fei Jian-Pi granules, Bu-Fei Yi-Shen granules, and Yi-Qi Zi-Shen granules based on TCM patterns respectively; and the control group treated using conventional western medicine. A total of 136 patients ≥ 65 years completed the study, with 63 patients comprising the trial group and 73 comprising the control group. After the six-month treatment and the 12-month follow-up period, significant differences were observed between the trial and control groups in the following aspects： frequency of acute exacerbation （P ≤ 0.040）, duration of acute exacerbation （P = 0.034）, symptoms （P ≤ 0.034）, 6-min walking distance （6MWD）（P ≤ 0.039）, dyspnea scale （P ≤ 0.036）; physical domain （P ≤ 0.019）, psychological domain （P ≤ 0.033）, social domain （P ≤ 0.020）, and environmental domain （P ≤ 0.044） of the WHOQOL-BREF questionnaire; and daily living ability domain （P ≤ 0.007）, social activity domain （P ≤ 0.018）, depression symptoms domain （P ≤ 0.025）, and anxiety symptoms domain （P ≤ 0.037） of the COPD-QOL. No differences were observed between the trial and control groups with regard to FVC, FEV1, and FEV1%.
