Glucagon-like peptide 1 agonists are potentially useful drugs for treating metabolic dysfunction-associated steatotic liver disease

In this editorial,we comment on Yin et al’s recently published Letter to the *** particular,we focus on the potential use of glucagon-like peptide 1 receptor agonists(GLP-1RAs)alone,but even more so in combination therapy,as one of the most promising therapies in metabolic dysfunction-associated steatotic liver disease(MASLD),the new definition of an old condition,non-alcoholic fatty liver disease,which aims to better define the spectrum of steatotic *** is well known that GLP-1RAs,having shown outstanding performance in fat loss,weight loss,and improvement of insulin resistance,could play a role in protecting the liver from progressive *** clinical trials have shown that,among GLP-1RAs,semaglutide is a safe,well-studied therapeutic choice for MASLD patients;however,most studies demonstrate that,while semaglutide can reduce steatosis,including steatohepatitis histological signs(in terms of inflammatory cell infiltration and hepatocyte ballooning),it does not improve *** of therapies with different but complementary mechanisms of action are considered the best way to improve efficiency and slow disease progression due to the complex pathophysiology of the *** particular,GLP-1RAs associated with antifibrotic drug therapy,dual glucose-dependent insulinotropic polypeptide(GIP)/GLP-1RA or GLP-1 and glucagon RAs have promoted greater improvement in hepatic steatosis,liver biochemistry,and non-invasive fibrosis tests than ***,although to date there are no definitive indications from international drug agencies,there is the hope that soon the therapeutic lines in the most advanced phase of study will be able to provide a therapy for MASLD,one that will certainly include the use of GLP-1RAs as combination therapy.

Identification of research gaps to improve care for healthy ageing:a scoping review

Objective Several research gaps affect the improvement of care for healthy *** identification is crucial to developing a specific research prioritisation agenda supporting progress at the micro(clinical),meso(service delivery)and macro(system)*** achieve this,a scoping review was carried out to describe the most significant gaps impeding the improvement of care for healthy *** A scoping review of the literature was conducted according to the Joanna Briggs Institute *** selected articles were analysed to identify topics or areas essential for improving care for healthy ageing but requiring further support from *** criteria Every type of scientific article,except for randomised controlled trials,was considered of potential interest without restrictions on publication date,type of publication and *** sources A systematic search(last search:6 December 2023)was conducted using PubMed,MEDLINE and *** Overall,1558 articles were retrieved from the *** these,310 were finally retained for this work.A total of 1195 research gaps were identified(average:3.85 per article)and clustered into the 13 primary areas:ageing,care approach,caregivers,health economics,health,interventions,policies,research,settings,training,technology,specific populations and understanding the older *** particular,research for improving the person-centred approach(n=38),better considering cultural diversities(n=27),implementing integrated care(n=25)and ensuring access to care(n=25)were the most prevalent priorities reported in the *** A wide range of factors spanning multiple disciplines,from clinical to policy levels,require special consideration,exploration and *** findings of this scoping review represent an essential step in identifying gaps for developing a research prioritisation agenda to improve care for healthy ageing.

Objective Anemia is a common public health concern in patients with type 2 diabetes *** study aimed to identify the prevalence of anemia among patients with *** Electronic databases,including PubMed,Scopus,Web of sciences,and Google Scholar,were searched systematically for studies published between 2010 and *** removing duplicates and inappropriate reports,the remaining manuscripts were reviewed and appraised using theNewcastleOttawa Scale(NOS)tool.A random-effects model was used to calculate the pooled estimates of the extracted data using Stata version *** of the studies was assessed using the Q *** A total of 51 articles containing information on 26,485 patients with diabetes were included in this *** articles were mainly from Asia(58.82%)and Africa(35.29%).The overall prevalence of anemia was 35.45%(95%CI:30.30–40.76),with no evidence of heterogeneity by *** the two continents with the highest number of studies,the prevalence of anemia in patients with diabetes was significantly higher in Asia[40.02;95%CI:32.72–47.54]compared to Africa[28.46;95%CI:21.90–35.50](P for heterogeneity=0.029).Moreover,there has been an increasing trend in the prevalence of anemia in patients with diabetes over time,from[15.28;95%CI:9.83–22.21]in 2012 to[40.70;95%CI:10.21–75.93]in *** Globally,approximately 4 in 10 patients with diabetes suffer from ***,routine anemia screening and control programs every 3 months might be useful in improving the quality of life of these patients.

In this editorial,we comment on the article published in the recent issue of the World Journal of *** liver failure(ALF)is a fatal disease that causes uncontrolled massive hepatocyte death and rapid loss of liver *** and pyroptosis,cell death forms that can be initiated or blocked concurrently,can play significant roles in developing inflammation and various ***,their roles in ALF remain *** article discovered the positive feedback between ferroptosis and pyroptosis in the progression of ALF,and revealed that the silent information regulator sirtuin 1(SIRT1)inhibits both pathways through p53,dramatically reducing inflammation and protecting *** suggests the potential use of SIRT1 and its downstream molecules as therapeutics for ***,we will discuss the role of ferroptosis and pyroptosis in ALF and the crosstalk between these cell death ***,we address potential treatments that could alleviate ALF by simultaneously inhibiting both cell death pathways,as well as examples of SIRT1 activators being used as disease treatment strategies,providing new insights into the therapy of ALF.

The miR-9-5p/CXCL11 pathway is a key target of hydrogen sulfide-mediated inhibition of neuroinflammation in hypoxic ischemic brain injury

We previously showed that hydrogen sulfide(H2S)has a neuroprotective effect in the context of hypoxic ischemic brain injury in neonatal ***,the precise mechanism underlying the role of H2S in this situation remains *** this study,we used a neonatal mouse model of hypoxic ischemic brain injury and a lipopolysaccharide-stimulated BV2 cell model and found that treatment with L-cysteine,a H2S precursor,attenuated the cerebral infarction and cerebral atrophy induced by hypoxia and ischemia and increased the expression of miR-9-5p and cystathionineβsynthase(a major H2S synthetase in the brain)in the prefrontal *** also found that an miR-9-5p inhibitor blocked the expression of cystathionineβsynthase in the prefrontal cortex in mice with brain injury caused by hypoxia and ***,miR-9-5p overexpression increased cystathionine-β-synthase and H2S expression in the injured prefrontal cortex of mice with hypoxic ischemic brain injury.L-cysteine decreased the expression of CXCL11,an miR-9-5p target gene,in the prefrontal cortex of the mouse model and in lipopolysaccharide-stimulated BV-2 cells and increased the levels of proinflammatory cytokines BNIP3,FSTL1,SOCS2 and SOCS5,while treatment with an miR-9-5p inhibitor reversed these *** findings suggest that H2S can reduce neuroinflammation in a neonatal mouse model of hypoxic ischemic brain injury through regulating the miR-9-5p/CXCL11 axis and restoringβ-synthase expression,thereby playing a role in reducing neuroinflammation in hypoxic ischemic brain injury.
