Pain inhibition through transplantation of fetal neuronal progenitors into the injured spinal cord in rats

Neuropathic pain after spinal cord injury(SCI) is a complex condition that responds poorly to usual treatments. Cell transplantation represents a promising therapy;nevertheless, the ideal cell type in terms of neurogenic potential and effectiveness against pain remains largely controversial. Here, we evaluated the ability of fetal neural stem cells(fNSC) to relieve chronic pain and, secondarily, their effects on motor recovery. Adult Wistar rats with traumatic SCI were treated, 10 days after injury, with intra-spinal injections of culture medium(sham) or fNSCs extracted from telencephalic vesicles(TV group) or the ventral medulla(VM group) of E/14 embryos. Sensory(von Frey filaments and hot plate) and motor(the Basso, Beattie,Bresnahan locomotor rating scale and inclined plane test) assessments were performed during 8 weeks. Thereafter, spinal cords were processed for immunofluorescence and transplanted cells were quantified by stereology. The results showed improvement of thermal hyperalgesia in the TV and VM groups at 4 and 5 weeks after transplantation, respectively. Moreover, mechanical allodynia improved in both the TV and VM groups at 8 weeks. No significant motor recovery was observed in the TV or VM groups compared with sham. Stereological analyses showed that ~70% of TV and VM cells differentiated into NeuN+ neurons,with a high proportion of enkephalinergic and GABAergic cells in the TV group and enkephalinergic and serotoninergic cells in the VM group. Our study suggests that neuronal precursors from TV and VM, once implanted into the injured spinal cord, maturate into different neuronal subtypes, mainly GABAergic, serotoninergic, and enkephalinergic, and all subtypes alleviate pain, despite no significant motor recovery. The study was approved by the Animal Ethics Committee of the Medical school of the university of S?o paulo(protocol number 033/14) on March 4, 2016.

The prevalence of torus mandibularis, Eagle’s syndrome and parafunctional activity was reported to be higher in patients with temporomandibular joint dysfunction and might be useful as an indicator of increased risk of temporomandibular disorders. This study case report was to evaluate a 62-year-old female patient, with limited mandibular and cervical movements, articular and muscular pain, ear fullness, irritation of the lingual mucosa, discomfort associated with movement of her tongue, involving temporomandibular dysfunction, torus mandibularis and Eagle’s syndrome. The treatment comprehended the collection of data of Helkimo’s Clinical Dysfunction Index, electromyographic activity of the masseter and temporalis muscles in rest position and bite force, masticatory efficiency, speech evaluation and oclusal splint, after the excision of the stylohyoid process and mandibularis torus. The authors found a decrease in electromyographic activity of masticatory muscles, a reduction of painful symptoms throughout the region muscular and joint pains, an increase in the amplitude of mandibular movements, an improvement in masticatory efficiency and an increase in bite force.

Objective: The present study aimed to investigate and to compare the electrophysiological changes in bipolar patients and healthy subjects during the execution of a saccade task. Materials and Methods: The subjects had to respond to a fixed visual stimulus presented by a LEDs bar. We executed an ANOVA (one-way) and post hoc Scheffé test to examine the difference of absolute alpha power and reaction time among four groups: control, depression, manic and euthymic. We observed the frontal, parietal and occipital regions. The paired t test was realized on each electrode and group to compare the pre and post moment in the task. Results: We observed a statistical difference among the groups for the behavioral variable—saccade reaction time. For the electrophysiological variable—absolute alpha power, we did not find significant difference between the moments (pre and post stimulus presentation) for none of the electrodes of each bipolar group. However, the results pointed out to a difference between the moments for F3, P3, O1 and O2 electrodes for control group. Conclusion: We cannot affirm that the task influences the cortical activity of the patients. It is possible that the method used to analyze the data is not the adequate. The time-frequency analysis could be better to analyze the present data. We also observed that the absolute alpha power could be considered a marker of bipolar disorder, but not of the different states (i.e., mania, depression, euthymia).
