Nowadays,presynaptic dopaminergic positron emission tomography,which assesses deficiencies in dopamine synthesis,storage,and transport,is widely utilized for early diagnosis and differential diagnosis of *** review provides a comprehensive summary of the latest developments in the application of presynaptic dopaminergic positron emission tomography imaging in disorders that manifest *** conducted a thorough literature search using reputable databases such as PubMed and Web of *** criteria involved identifying peer-reviewed articles published within the last 5 years,with emphasis on their relevance to clinical *** findings from these studies highlight that presynaptic dopaminergic positron emission tomography has demonstrated potential not only in diagnosing and differentiating various Parkinsonian conditions but also in assessing disease severity and predicting ***,when employed in conjunction with other imaging modalities and advanced analytical methods,presynaptic dopaminergic positron emission tomography has been validated as a reliable in vivo *** validation extends to screening and exploring potential neuropathological mechanisms associated with dopaminergic *** summary,the insights gained from interpreting these studies are crucial for enhancing the effectiveness of preclinical investigations and clinical trials,ultimately advancing toward the goals of neuroregeneration in parkinsonian disorders.

Extrapulmonary benign and malignant lesions avid for 18F-fluoro-deoxyglucose by multivariate regression model identification

Whether extrapulmonary lesions are avid for18F-fluorodeoxyglucose(18F-FDG) could help to differentiate the benign or malignant lung *** this trial,the 199 consecutive patients with newly diagnosed lung lesions(169 malignant and 36 benign lesions) were imaged by whole body18F-FDG pet/*** and clinic results served as the reference *** malignancy likelihood were conducted by ctscores;the maximum standardized uptake value(SUVmax) of lung lesions,and pet on FDG negative or positive,as well as metastasis index(MI),by pet combined with ct *** data were analyzed by stepwise logistic regression and *** malignancy predictive probability(P) was obtained by P =ex/(1+ex),where x= –1.16+0.87(ctscore) +0.15(SUVmax)+0.27(MI).The area under curve(AUC) for the fitted logistic model was 0.82±0.04,this was superior and significantly different from SUVmax(AUC,0.73±0.05) and ctscores(AUC,0.71±0.05).The fitted logistic model could improve the diagnostic *** MI could help for differential diagnosis.

Development and Characterization of Novel FAP-Targeted Theranostic Pairs: A Bench-to-Bedside Study

Fibroblast activation protein (FAP) is among the most popular targets in nuclear medicine imaging and cancer theranostics. Several small-molecule moieties (FAPI-04, FAPI-46, etc.) are used for developing FAP-targeted theranostic agents. Nonetheless, the circulation time of FAP inhibitors is relatively short, resulting in rapid clearance via kidneys, low tumor uptake, and associated unsatisfactory treatment efficacy. To address the existing drawbacks, we engineered 3 peptides named FD1, FD2, and FD3 with different circulation times through solid-phase peptide synthesis. All the 3 reported peptides bind to human and murine FAP with single-digit nanomolar affinity measured by surface plasmon resonance. The diagnostic and therapeutic potential of the agents labeled with 68Ga and 177Lu was assessed in several tumor models exhibiting different levels of FAP expression. While radiolabeled FD1 was rapidly excreted from kidneys, radiolabeled FD2/FD3 have significantly prolonged circulation, increased tumor uptake, and decreased kidney accumulation. Our findings indicated that [68Ga]Ga-DOTA-FD1 positron emission tomography (pet) effectively detected FAP dynamics, whereas [177Lu]Lu-DOTA-FD2 and [177Lu]Lu-DOTA-FD3 exhibited remarkable therapeutic efficacy in FAP-overexpressing tumor models, including pancreatic cancer cell models characterized by abundant stroma. Moreover, a pilot translational investigation demonstrated that [68Ga]Ga-DOTA-FD1 had the capability to identify both primary and metastatic tumors with precision and distinction. In summary, we developed [68Ga]Ga-DOTA-FD1 for same-day pet imaging of FAP dynamics and [177Lu]Lu-DOTA-FD2 and [177Lu]Lu-DOTA-FD3 for effective radioligand therapy of FAP-overexpressing tumors.

Metabolic assessement of spleen infiltration by non-Hodgkin lymphoma using ^(18)F-FDG pet/ct:Preliminary clinical results

18F-FDG pet/ct was used for evaluation of spleen infiltration in patients with B-cell non-Hodgkin lymphoma(NHL).Five patients with histological diagnosed B-cell NHL underwent1 8F-FDG pet/ct *** integrated pet/ct image,spleen infiltration was considered when pet images revealed a discrete margin of solid splenic masses or diffuse lesion of spleen with higher maximum standardized uptake value(SUV) greater than those of normal liver *** images demonstrating a positive splenic index(>480 mL)or focal hypodensities were classified as positive for spleen *** the patients underwent systemic ***1 8F-FDG pet and ct results were compared with final *** patients had spleen infiltration originating from B-cell NHL at final *** diagnoses,which were confirmed by clinical and ct(n=3) or1 8F-FDG pet/ct(n=1) follow-up in 4 patients and biopsy of 1 *** integrated pet/ct image,1 8F-FDG pet was true-positive in all 5 patients with spleen *** was true-positive in 4 of the 5 patients with spleen infiltration and false-negative in 1 patients (spleen infiltration without morphology changes).The accuracies of1 8F-FDG pet and ct for evaluating the spleen infiltration were 100% and 80% at staging,*** preliminary results suggested metabolic imaging of1 8F-FDG pet/ct may be helpful in the diagnosis of spleen infiltration in B-cell NHL *** patients may benefit from1 8F-FDG pet/ct in diagnosis,when spleen infiltration without morphology changes,which may not be diagnosed exactly by conventional image.

^(18)F-FDG pet/ct for malignant small intestinal neoplasms

In this paper, 18F-FDG pet/ct data of 19 malignant SINs (small intestinal neoplasms) were consecutively reviewed. Nnine patients accepted pet/ct scan for preoperative diagnosis and staging, while ten patients presented follow-up after treatment and restaging. The results were correlated with abdomen enhanced ct and surgical pathological findings. Abdominal pain and weight loss were the most common findings. About 16% SINs located in the duodenum, 52% in the jejunum and 32% in the ileum. Lymphoma was the most frequent neoplasms. pet/ct revealed residual or recurrent malignant SINs in 5 patients who had negative or non-definite findings by abdomen ct and demonstrated extra-abdomen metastasis in 3 patients. Clinical decisions of treatment were changed for 6 patients after pet/ct examinations. The 18F-FDG pet/ct were better than ct in accuracy, negative predictive value and positive predictive value (89.5% vs 68.4%, 100% vs 66.7%, and 81.8% vs 69.2%, respectively). Whole body 18F-FDG pet/ct may be an effective molecular imaging method for staging and restaging of malignant SINs.

Metabolic imaging for guidance of curative treatment of isolated pelvic implantation metastasis after resection of spontaneously ruptured hepatocellular carcinoma: A case report

Spontaneous rupture of hepatocellular carcinoma(HCC) is a life-threatening complication and its prognosis is significantly poor because of the high recurrence rate after initial hepatectomy. Resection of isolated extrahepatic metastasis of HCC has been advocated to obtain a possibility of long-term survival. However, it is a challenge for clinicians to detect implantation metastasis of spontaneously ruptured HCC. Accurate re-staging plays the most important role in making a decision on isolated metastasis resection. 18F-fluorodeoxyglucose(18F-FDG) positron emission tomography/computed tomography(pet/ct) is useful in detecting intraabdominal implantation metastasis from a variety of malignancies and shows superior accuracy to conventional imaging modalities in determining the location of metastasis. We present one patient with a new isolated pelvic implantation metastasis detected by 18F-FDG pet/ct and pathologically confirmed by pet/ct-guided percutaneous biopsy, who had a history of resection of spontaneously ruptured HCC two years ago. The patient's condition was stable at the 6-mo follow-up after resection of the isolated pelvic metastasis.

Comparison of diagnostic accuracy between ^(18)F-FDG pet and pet/ct for pulmonary neoplasm

Aimed at comparing diagnostic accuracy of 18F-FDG pet with pet/ct for pulmonary neoplasm, a study based on multi-center clinical trial of the diagnoses, in randomized and semi-blind ways, was executed from January 2006 to June 2007. It included 55 patients, i.e. 16 with histopathologically proved lung tumors, 16 with tuberculosis and 23 with benign lesions (inflammation, pseudotumor, granuloma, fibrosis and others). The histopathologic and clinic results were served as reference standard. Statistical significances in pulmonary nodule diagnosis between 18F-FDG pet and pet/ct were determined with 95% confidence interval obtained by ROC analysis. The 18F-FDG pet detected lung neoplasm with a sensitivity of 87.5% (14/16), a specificity of 59.0% (23/39), an accuracy of 67.3% (37/55) and a positive-likelihood ratio of 2.13. The 18F-FDG pet/ct detected lung neoplasm with a sensitivity of 93.8% (15/16), a specificity of 61.5% (24/39), an accuracy of 70.9% (39/55) and a positive-likelihood ratio of 2.43. The area under curves (AUC) of 18F-FDG pet and pet/ct were 0.803±0.068 and 0.799±0.063, respectively. It can be concluded that the diagnostic accuracy for malignant pulmonary nodules between 18F-FDG pet and pet/ct was not statistically different.

Brain dopaminergic system changes in drug addiction:a review of positron emission tomography findings

Dopamine （DA） is considered crucial for the rewarding effects of drugs of abuse, but its role in addiction remains unclear. Positron emission tomography （pet） is the first technology used for in vivo measurement of components of the dopaminergic system in the human brain. In this article, we review the major findings from pet imaging studies on the involvement of DA in drug addiction, including presynaptic DA synthesis, vesicular monoamine transporter 2, the DA transporter, and postsynaptic DA receptors. These results have corroborated the role of DA in addiction and increased the understanding of its underlying mechanisms.

Diagnostic accuracy of fluorine-18-fluorodeoxyglucose positron emission tomography in gallbladder cancer: A meta-analysis

AIM: To meta-analyze published data about the diagnostic accuracy of fluorine-18-fluorodeoxyglucose(18F-FDG) positron emission tomography(pet) and pet/computed tomography(pet/ct) in the evaluation of primary tumor in patients with gallbladder cancer(GBCa).METHODS: A comprehensive literature search of studies published through 30 th June 2014 regarding the role of 18F-FDG pet and pet/ct in the evaluation of primary gallbladder cancer(GBCa) was performed. All retrieved studies were reviewed. Pooled sensitivity and specificity of 18F-FDG pet or pet/ct in the evaluation of primary GBCa were calculated. The area under the summary receiving operator characteristics curve(AUC) was calculated to measure the accuracy of these methods. Sub-analyses considering the device used(pet vs pet/ct) were carried ***: Twenty-one studies comprising 495 patients who underwent 18F-FDG pet or pet/ct for suspicious GBCa were selected for the systematic review. The meta-analysis of 13 selected studies provided the following results: sensitivity 87%(95%CI: 82%-92%),specificity 78%(95%CI: 68%-86%). The AUC was 0.88. Improvement of sensitivity and specificity was observed when pet/ct was ***: 18F-FDG-pet and pet/ct demonstrated to be useful diagnostic imaging methods in the assessment of primary tumor in GBCa patients,nevertheless possible sources of false-negative and false-positive results should be kept in mind. pet/ct seems to have a better diagnostic accuracy than pet alone in this setting.
