Many polyurethanes(PUs)are blood-contacting materials due to their good mechanical properties,fatigue resistance,cytocompatibility,biosafety,and relatively good *** functionalization of the PUs using chemical synthetic methods is especially attractive for expanding their ***,a series of catechol functionalized PU(CPU-PTMEG)elastomers containing variable molecular weight of polytetramethylene ether glycol(PTMEG)soft segment are reported by stepwise polymerization and further introduction of *** the molecular weight of PTMEG fragment enables a regulable catechol content,mobility of the chain segment,hydrogen bond and microphase separation of the C-PUPTMEG elastomers,thus offering tunability of mechanical strength(such as breaking strength from 1.3 MPa to 5.7 MPa),adhesion,self-healing eficiency(from 14.9%to 96.7%within 2 hours),anticoagulant,antioxidation,anti-inflammatory properties and cellular growth *** cardiovascular stent coatings,the C-PU-PTMEGs demonstrate enough flexibility to withstand deformation during the balloon dilation *** special importance is that the C-PU-PTMEG-coated surfaces show the ability to rapidly scavenge free radicals to maintain normal growth of endothelial cells,inhibit smooth muscle cell proliferation,mediate inflammatory response,and reduce thrombus *** the universality of surface adhesion and tunable multifunctionality,these novel C-PU-PTMEG elastomers should find potential usage in artificial heart valves and surface engineering of stents.

Prophylactic effects of alkaloids from Ba lotus seeds on L-NNA-induced hypertension in mice

Alkaloids from Ba lotus seeds(ABLS) are a kind of important functional compounds in lotus seeds. The present study was designed to determine its hypertension prophylactic effects in the L-NNA-induced mouse hypertension model. The mice were treated with ABLS, the serum and tissues levels of NO, MDA, ET-1, VEGF, and CGRP were determined using the experimental kits, the mRNA levels of various genes in the heart muscle and blood vessel tissues were further determined by RT-PCR assay. ABLS could reduce the systolic blood pressure(SBP), mean blood pressure(MBP), and diastolic blood pressure(DBP), compared to that of the model control group. After ABLS treatment, the NO(nitric oxide) contents in serum, heart, liver, kidney and stomach of the mice were higher than that of the control mice, but the MDA(malonaldehyde) contents were lower than that of the control mice. The serum levels of ET-1(endothelin-1), VEGF(vascular endothelial growth factor) were decreased after ABLS treatment, but CGRP(calcium gene related peptide) level was increased. The ABLS treated mice had higher mRNA expressions of HO-1, nNOS, and eNOS and lower expressions of ADM, RAMP2, IL-1β, TNF-α, and iNOS than the control mice. Higher concentration of ABLS had greater prophylactic effects, which were close to that of the hypertension drug captopril. These results indicated the hypertension prophylactic effects of ABLS could be further explored as novel medicine or functional food in the future.

目的探索深度神经网络(DNN)辅助的心肌声学造影(MCE)定量分析对经皮冠状动脉介入治疗(PCI)成功开通梗死相关动脉后的急性ST段抬高型心肌梗死(STEMI)患者短期预后的预测价值。方法回顾性纳入2021年6-11月于武汉大学人民医院接受PCI成功开通梗死相关动脉(心肌梗死再灌注后血流分级3级)的STEMI患者97例。PCI后48 h内进行MCE检查,随访至术后120 d。主要心血管不良事件定义为心源性死亡及因充血性心力衰竭、再发心肌梗死、卒中、再发心绞痛再入院。心肌分割框架由基于U-net和LSTMs的组成,输出各心肌节段平台期增强强度(A)、灌注速率常数(β)和微循环血流量(MBF),并进行MCE定量参数重复性检验。通过MBF将患者等分为低MBF组与高MBF组,比较两组间基线特征、心血管不良事件的差异。其他纳入变量包括常规生化检查、室壁运动分析、MCE定性分析及左室射血分数等。通过Cox风险回归分析探索各变量与预后的关系。绘制ROC曲线以评估模型预测效能,使用综合判别改善指数(IDI)进行模型间的预测效能比较。结果自动心肌分割所有帧(3810帧)耗时377 s。其中92.89%、7.11%的帧由经验医师评价为"良好分割"和"需要校正"。A、β、MBF的观察者内及观察者间组内相关系数为0.97~0.99。20例患者在随访期间出现心血管不良事件,多因素Cox回归分析显示,梗死相关动脉支配区域MBF每增加1 IU/s,短期发生心血管不良事件的风险降低6%(HR 0.94,95%CI=0.91~0.98)。低MBF组短期内发生心血管不良事件的风险比高MBF组高4.5倍(HR 5.50,95%CI=1.55~19.49)。在MCE传统定性分析中加入DNN辅助的MCE定量分析后,预后预测IDI为15%(AUC 0.86,敏感性0.78,特异性0.73)。结论STEMI患者PCI术后的梗死相关动脉支配区域微循环血流量是短期心血管不良事件的独立保护因素。基于DNN的MCE心肌分割框架可快速、客观、可重复地输出微循环定量灌注参数。在传统定性分析中加入DNN的定量分析对预后预测提供了增量价值,有望成为STEMI-PCI术后患者风险分层与临床随访的有力工具。
