We describe a new species of Discostella from Guiyang based on light and scanning electron microscopic observations,as well as molecular *** species is morphologically distinguished from other species by a relatively smooth and slightly recessed central area with marginal striae of unequal lengths forked near the *** fultoportulae and a single rimoportula are situated between every two costae or on the *** morphological characters and the concatenated data of rbc L and SSU rDNA indicate that Discostella guiyangensis is a unique and new *** to divergence time estimation analysis,Discostella guiyangensis is the oldest species among the Discostella species sequenced *** addition,we complement molecular data of Discostella asterocostata and Discostella stelligera.

Mescaline,among the earliest identified natural hallucinogens,holds great potential in psychotherapy ***,despite the existence of a postulated biosynthetic pathway for more than half a century,the specific enzymes involved in this process are yet to be *** this study,we investigated the cactus Lophophora williamsii(Peyote),the largest known natural producer of the phenethylamine *** employed a multi-faceted approach,combining de novo whole-genome and transcriptome sequencing with comprehensive chemical profiling,enzymatic assays,molecular modeling,and pathway engineering for pathway *** identified four groups of enzymes responsible for the six catalytic steps in the mescaline biosynthetic pathway,and an N-methyltransferase enzyme that N-methylates all phenethylamine intermediates,likely modulating mescaline levels in ***,we reconstructed the mescaline biosynthetic pathway in both Nicotiana benthamiana plants and yeast cells,providing novel insights into several challenges hindering complete heterologous mescaline *** together,our study opens up avenues for exploration of sustainable production approaches and responsible utilization of mescaline,safeguarding this valuable natural resource for future generations.

Ethylene (C2H4) methoxycarbonylation (EMC) with CO and methanol (CH3OH) to methyl propanoate (MP) is industrially important due to its 100%atom efficiency [1].However, the heavy reliance on homogeneous Pd-based catalysts and strong acid media over commercial EMC processes still causes economic and environmental concerns[2]. At present, supported single-atom catalysts (SACs) can serve as bridges for accessing homogeneous catalysts in traditional heterogeneous catalytic processes. SACs have been extensively studied because they may provide new opportunities for exploring heterogeneous catalysts for the EMC process [3]. However, the mechanism of the heterocatalytic EMC process on SACs is still unclear. Therefore,developing efficient SACs and unveiling their working mechanism are highly important for identifying the heterocatalytic EMC field.

The electroreduction of carbon dioxide to generate multicarbon products is significant for achieving a carbon-neutral economy. However, the selectivity for multi-carbon products faces several challenges, such as the intricate nature of the multistep hydrogenation processes, slow reaction kinetics and the bicarbonate precipitate issue caused by alkaline conditions. These factors hinder the extensive utilization of this reaction. Herein, we successfully developed and implemented a tandem electrolyzer cascade to separate and optimize the complex elementary reactions involved in the process of multi-carbon production. This approach enables us to enhance the Faradaic efficiency (FE) of each step, consequently improving the overall performance of the system. G protein-coupled receptors(GPCRs) are a large family of cell membrane proteins that are important targets for drug discovery. Nanobodies, also known as VHH(variable domains of heavy chain-only antibodies, HcAbs) antibodies, are small antibody fragments derived from camelids that have gained significant attention as potential therapeutics for targeting GPCRs due to their advantages over conventional antibodies. However, there are challenges in developing nanobodies targeting GPCRs, among which epitope accessibility is the most significant because the cell membrane partially shields the GPCR surface. We developed a universal protocol for making nanobodies targeting GPCRs using the cell membrane extract of GPCR-overexpressing HEK293 cells as the llama/alpaca immunization antigen. We constructed an immune VHH library and identified nanobodies by phage display bio-panning. The monoclonal nanobodies were recombinantly expressed in Escherichia coli(E. coli) and purified to characterize their binding potency.

Osteoarthritis and psoriasis arthritis are two degenerative forms of arthritis that share similar yet also different manifestations at the histological,cellular,and clinical *** have marked them as two entirely distinct *** recent dis-coveries in disease initiation and progression,potential mechanisms,cellular signaling path-ways,and ongoing clinical therapeutics,there are now more opportunities for discovering osteoarthritis *** review summarized the osteoarthritis and psoriasis arthritis signaling pathways,crosstalk between BMP,WNT,TGF-β,VEGF,TLR,and FGF signaling pathways,bio-markers,and anatomical *** bench research,we demonstrated that regen-erative medicine is a promising alternative for treating osteoarthritis by highlighting significant scientific discoveries on entheses,multiple signaling blockers,and novel molecules such as immunoglobulin new antigen receptors targeted for potential drug ***,we offered valuable therapeutic approaches with a multidisciplinary strategy to treat patients with osteoarthritis or psoriasis arthritis in the coming future in the clinic.

The innate immune regulator stimulator of interferon genes(STING)mediates self-DNA sensing and leads to the induction of type I interferons and inflammatory cytokines,which promotes the progression of various inflammatory and autoimmune *** immune system plays a critical role in regulating obesity-induced islet dysfunction,whereas the potential effect of STING signaling is not fully ***,we demonstrate that STING is mainly expressed and activated in islet macrophages upon high-fat diet(HFD)***^(-/-)alleviates HFD-induced islet inflammation by inhibiting the expression of pro-inflammatory cytokines and the infiltration of ***,palmitic acid incubation promotes mitochondrial DNA leakage into the cytosol and subsequently activates STING pathway in ***,STING activation in macrophages impairs glucose-stimulated insulin secretion by mediating the engulfment ofβcell insulin secretory *** inhibiting STING activation enhances insulin secretion to control ***,our results reveal a regulatory mechanism in controlling the islet inflammation and insulin secretion in dietinduced obesity and suggest that selective blocking of the STING activation may be a promising strategy for treating type 2 diabetes.

The high mortality of patients with lung squamous cell carcinoma(LUSC)results from metastasis rather than primary tumors,1 whereas the molecular pathogenesis of cancer metastasis remains poorly *** encodes urokinase-type plasminogen activator(uPA)which is closely related to tumor diagnosis,treatment target,and prognosis of patients.2.3 We previously predicted that PLAU is a key gene negatively associated with overall survival by integrated multidimensional analyses of The Cancer Genome Atlas(TCGA)datasets from 504 samples of LUSC tumor tissues and 46 samples of adjacent non-tumorous lung tissues.4 However,whether the role of PLAU contributes to the metastasis of LUC remains ***,we analyzed the association between uPA levels and LUSC and found that uPA levels were associated with pathological variations in LUSC patients.
