Echinococcosis or hydatid disease （HD） is a zoonosis caused by the larval stages of taeniid cestodes belong- ing to the genus Echinococcus. Hepatic echinococcosis is a life-threatening disease, mainly differentiated into alveolar and cystic forms, associated with Echinoc- cus multilocularis （E. multi/ocular/s） and Echinococcus granulosus （E. granulosus） infection, respectively. Cys- tic echinococcosis （CE） has a worldwide distribution, while hepatic alveolar echinococcosis （AE） is endemic in the Northern hemisphere, including North America and several Asian and European countries, like France, Germany and Austria. E. granulosus young cysts are spherical, unilocular vesicles, consisting of an internal germinal layer and an outer acellular layer. Cyst expansion is associated with a host immune reaction and the subsequent development of a fibrous layer, called the per/cyst; old cysts typically present internal septa- tions and daughter cysts. E. multilocularis has a tumor-like, infiltrative behavior, which is responsible for tissue destruction and finally for liver failure. The liver is the main site of HD involvement, for both alveolar and cystic hydatidosis. HD is usually asymptomatic for a long period of time, because cyst growth is commonly slow; the most frequent symptoms are fatigue and abdominal pain. Patients may also present jaundice, hepatomegaly or anaphylaxis, due to cyst leakage or rupture. HD diagnosis is usually accomplished with the combined use of ultrasonography and immunodiagnosis; furthermore, the improvement of surgical techniques, the introduction of minimally invasive treatments [such as puncture, aspiration, injection, re-aspiration （PAIR）] and more effective drugs （such as benzoimidazoles） have deeply changed life expectancy and quality of life of patients with HD. The aim of this article is to provide an up-to-date review of biological, diagnostic, clinical and therapeutic aspects of hepatic echinococcosis.

Canine hypoadrenocorticism may be characterized by insufficiency of adrenocortical hormonal secretion of glucocorticoids and mineralocorticoids. Clinical signs are nonspecific such as dehydration, hypovolemic shock, bradycardia, hypotension, emesis, diarrhea, among other clinical and laboratory changes, and can mimic other diseases, and because of this, the disease can go unnoticed by the veterinarian, increasing the mortality and morbidity of patients seen in emergency consultations. Despite being of low occurrence, hypoadrenocorticism is often underdiagnosed, and should be considered in the differential diagnosis in the presence of gastrointestinal clinical signs or hypovolemic shock, and low Sodium:Potassium ratio. The definitive diagnosis is based on the dosage of cortisol before and after the application of the adrenocorticotrophic hormone. Acute treatment consists of patient stabilization, and chronic includes replacement of glucocorticoids and mineralocorticoids. Prognosis is usually favorable if the diagnosis is early and the correct treatment is performed. This article is a review focusing on clinical signs, laboratory findings, diagnosis and treatment on canine hypoadrenocorticism, to increase the knowledge about the disease to veterinarians.

【目的】设计乳酸杆菌属特异性T-RFLP技术(末端限制性片段长度多态性分析)对14株乳酸杆菌进行分型。【方法】采用源于16S-23S rRNA基因间隔区序列的乳酸杆菌属特异性引物LAB-rev,乳酸杆菌的属特异性引物,6-FAM荧光标记后结合16S上游通用引物7f用于乳酸杆菌的PCR扩增。【结果】选取HaeⅢ和HhaⅠ进行限制性酶切,最后对酶切后的产物末端测序得到T-RFLP峰谱图,该图谱能够快速准确地对不同种的乳酸杆菌进行定性、定量的分析。【结论】实验成功搭建T-RFLP技术用于微生态环境中乳酸杆菌检测的平台,对于在功能性食品、乳酸饮料和药物对肠道微生态的影响及菌种鉴定等领域有重大意义。

Bacterial genomic DNA is highly organized into one or few compacted bodies known as nucleoid, which is composed of DNA, RNA and several DNA-binding proteins. These DNA-binding proteins require essential alterations in their expression during stationary phase of growth in order to re-spond to stressful environmental conditions. Dps (DNA-binding protein from starved cells) is one of such DNA-binding proteins, which accumulates most when E. coli cells reach to the stationary phase. Here, we have characterized Dps protein under various growth phases. Immunofluorescent microscopic observation reveals that Dps plays a key role in final round of genome compaction during the stationary phase. Similar results are also obtained by Western immunoblot analysis, after quantification of Dps protein from the exponential phase and early stationary phase nucleoid bound fractions, separated by sucrose density gradient centrifugation. Our results support the conclusion that Dps occupies more than half of the stationary phase nucleoid in E. coli.

Subtype and sequence analysis of HIV-1 strains in Heilongjiang Province

Methicillin-resistant Staphylococcus aureus(MRSA) causing nosocomial infection is the most common pathogen spreading rapidly among people causing skin and soft tissue infection, life-threatening endocarditis and sepsis. With remarkable resistance to many conventional antibiotics, infection caused by MRSA are becoming increasingly difficult to combat in clinic, especially in patients with sepsis in ICU. To address this issue, traditional Chinese medicine(TCM) which is widely applied to prevent and treat various diseases for thousands of years, is increasingly attracting attention worldwide and has gradually been used as an alternative therapy. Xuebijing injection(XBJ) is a traditional Chinese herbal prescription made from Chishao(Radix Paeoniae Rubra), Danggui(Radix Angelica Sinensis), Chuanxiong(Rhizoma Chuanxiong), Honghua(Flos Carthami), and Danshen(Radix Salviae Miltiorrhizae). XBJ is an intravenous injection widely used in the conventional management of sepsis in China. Whether it has certain effects in the treatment of sepsis caused by MRSA and its molecular mechanism are still unknown. Pam3 CSK4 is a synthetic tripalmitoylated lipopeptide that mimics the acylated amino terminus of bacterial lipoproteins and can induce anti-bacterial immune response through TLR1/2. This study indicated that XBJ decreased Pam3 CSK4-induced production of cytokines(IL-6, TNF-α, etc.) in Raw264.7 cells, peritoneal macrophages and dendritic cells. XBJ also reduced the production of IL-6 and TNF-α triggered by heat-killed MRSA(HK-MRSA) in peritoneal macrophages and immortalized bone marrow-derived macrophages(iBMDM). Meanwhile, XBJ had no apparente effects of antibacterium to MRSA in vitro bacteriostasis experiments. In vivo study, low-dose of XBJ could protect mice from lethal dose of sepsis infected by MRSA. In conclusion, these results indicate that XBJ has certain effects in the treatment of sepsis caused by MRSA through inhibiting the expression of inflammatory cytokines.

Background Human immunodeficiency virus （HIV-1） is divided into two types, HIV-1 （groups M, N and O） and HIV-2. Heilongjiang Province located in the northeast of China, and the feature of the subtype distribution and sequence characteristics of HIV-1 strains prevalent in Heilongjiang Province is still uncertain. The aim of this study was to investigate the subtype distribution and genetic characteristics of HIV-1 strains in one hospital in Heilongjiang Province. Methods HIV-1 env gene was amplified by nested-PCR from peripheral blood mononuclear cells （PBMCs） obtained from 19 HIV-1 seropositive individuals in Heilongjiang Province. The C2-V3 region was sequenced. Aligned the nucleotide sequence of 19 samples with CLUSTAL W （BioEdit） software, results were acquired and used for phylogenetic tree analysis after artificial adjustment. Reference sequence, downloaded from Los Alamos HIV Sequence Database, was used to identify the subtype of obtained sequence. Genetic distance between sequences was assessed using the software MEGA 3.1 Kimura 2-parameter, and the Phylogenetic tree was reestablished with Neighbor-Joining method. Results Phylogenetic tree analysis showed that 19 Heilongjiang strains clustered closely to subtype B strain from Thailand and were far from other international subtype reference strains. Statistical test showed no significant discrepancy between the genetic distance of interclass and intra-class （P〉0.05）. The analysis of V3 loop amino sequence of 19 Heilongjiang B strains revealed that V3 tip motif of 10 samples （52.63%） was GPGQ, and of 4 samples （21.53%） was GPGR. Conclusions The subtype of 19 HIV-1 seropositive individuals in Heilongjiang Province is B′, and it is introduced from He＇nan Province. V3 tip motifs of the HIV-1 isolates are mainly GPGQ and GPGR.

AIM: To estimate the prevalence of gastric cancer(GC) in a cohort of patients diagnosed with GC and to compare it with patients diagnosed with all other types of gastro-intestinal(GI) cancer during the same ***: Between 2008 and 2013,five-year period,the medical records of all GI cancer patients who underwent medical care and confirm diagnosis of cancer were reviewed at the National Referral Hospital,Thimphu which is the only hospital in the country where surgical and cancer diagnosis can be made. Demographic information,type of cancer,and the year of diagnosis were ***: There were a total of 767 GI related cancer records reviewed during the study period of which 354(46%) patients were diagnosed with GC. There were 413 patients with other GI cancer including; esophagus,colon,liver,rectum,pancreas,gall bladder,cholangiocarcinoma and other GI tract cancers. The GC incidence rate is approximately 0.9/10000 per year(367 cases/5years per 800000 people). The geographic distribution of GC was the lowest in the south region of Bhutan 0.3/10000 per year compared to the central region 1.4/10000 per year,Eastern region 1.2/10000 per year,and the Western region 1.1/10000 per year. Moreover,GC in the South part was significantly lower than the other GI cancer in the same region(8% vs 15%; OR = 1.8,95%CI: 1.3-3.1,P = 0.05). Among GC patients,38% were under the age of 60 years,mean age at diagnosis was 62.3(± 12.1) years with male-to-female ratio 1:0.5. The mean age among patients with all other type GI cancer was 60 years(± 13.2) and male-tofemale ratio of 1:0.7. At time of diagnosis of GC,342(93%) were at stage 3 and 4 of and by the year 2013; 80(23%) GC patients died compared to 31% death among patients with the all other GI cancer(P = 0.08).CONCLUSION: The incidence rate of GC in Bhutan is twice as high in the United States but is likely an underestimate rate because of unreported and undiagnosed cases in the villages.
