Tumor-protein 53(p53)is a transcription factor(TF)encoded by TP53(Trp53 in mice)and is a master regulator of tumor-suppressive programs'.Importantly,TP53 is the most frequently mutated gene in human cancer,and its loss of function contributes to tumor development,exemplified by variable p53 mutations detected in 50%-75%of pancreatic cancers.2 From these perceptions,a number of targets transcribed by p53 that confer tumor suppression have been ***,our understanding of how p53 prevents normal cells from initiating tumorigenesis is still *** complement this limitation,herein,we established a new approach to identify underlying effectors transcribed by p53 in normal physiology.

Helicobacter pylori(***)is currently recognized as the primary carcinogenic pathogen associated with gastric tumorigenesis,and its high prevalence and resistance make it difficult to tackle.A graph neural network-based deep learning model,employing different training sets of 13,638 molecules for pre-training and fine-tuning,was aided in predicting and exploring novel molecules against ***.A positively predicted novel berberine derivative 8 with 3,13-disubstituted alkene exhibited a potency against all tested drug-susceptible and resistant *** strains with minimum inhibitory concentrations(MICs)of 0.25-0.5μg/*** studies demonstrated an ideal gastric retention of 8,with the stomach concentration significantly higher than its MIC at 24 h post *** administration of 8 and omeprazole(OPZ)showed a comparable gastric bacterial reduction(2.2-log reduction)to the triple-therapy,namely OPZ+amoxicillin(AMX)+clarithromycin(CLA)without obvious disturbance on the intestinal flora.A combination of OPZ,AMX,CLA,and 8 could further decrease the bacteria load(2.8-log reduction).More importantly,the mono-therapy of 8 exhibited comparable eradication to both triple-therapy(OPZ+AMX+CLA)and quadrupletherapy(OPZ+AMX+CLA+bismuth citrate)*** and BamD,playing a major role in outer membrane protein(OMP)transport and assembling,were identified and verified as the direct targets of 8 by employing the chemoproteomics technique.n summary,by targeting the relatively conserved OMPs transport and assembling system,8 has the potential to be developed as a novel *** candidate,especially for the eradication of drug-resistant strains.

Background:Sickle Cell Disease(SCD)presents a significant global health burden,characterised by inflammation and oxidative stress,contributing to its complex pathophysiology and substantial socioeconomic ***-3 has been shown to alleviate SCD and has been found to be richly available in traditional medicinal plants such as perilla seed,sea buckthorn and ***:This research highlights the potential of omega-3 fatty acid supplementation as a therapeutic intervention for ***:We conducted an extensive search across databases such as PubMed,MEDLINE,and Google Scholar to gather pertinent articles that explore the effect of omega-3 fatty acid on inflammatory and oxidative stress markers in *** such as oxidative stress,and inflammation were also used in the ***-3 fatty acids,particularly EPA and DHA,exhibit anti-inflammatory and antioxidant properties that hold promise in mitigating these pathological ***:Clinical studies demonstrate reductions in pain episodes,hospitalisations,and oxidative stress markers with omega-3 supplementation,indicating its therapeutic ***:Further research is needed to determine optimal dosage,duration,and population-specific *** effectiveness studies and long-term safety assessments are essential for informing clinical *** omega-3 supplementation into comprehensive treatment regimens alongside other modalities may offer a holistic approach to managing ***,ongoing research efforts in omega-3 fatty acid supplementation offer hope for optimizing treatment strategies and improving outcomes for individuals affected by SCD.

Severe acute respiratory syndrome coronavirus-2(SARS-CoV-2)emerged at the end of 2019 and is responsible for the coronavirus disease(COVID-19)pandemic which shook the healthcare system *** April 2024,over 775 million positive cases and 7 million deaths have been reported globally[1].Since its emergence,the virus is constantly accumulating mutations in its genome and evolving frequently over *** the last 3 years,we have witnessed the emergence of numerous variants from Alpha to Omicron and other subvariants,and some of them are more virulent escaping the natural and/or vaccine induced immunity and some are much less virulent than earlier variants and contributed to the pandemic[2,3].The World Health Organization(WHO)announced that the COVID-19 is no longer a public health emergency of international concern on May 5,2023[4].
