Transforming growth factor-beta 1(TGF-β1)has been extensively studied for its pleiotropic effects on central nervous system *** neuroprotective or neurotoxic effects of TGF-β1 in specific brain areas may depend on the pathological process and cell types ***-gated sodium channels(VGSCs)are essential ion channels for the generation of action potentials in neurons,and are involved in various neuroexcitation-related ***,the effects of TGF-β1 on the functional properties of VGSCs and firing properties in cortical neurons remain *** this study,we investigated the effects of TGF-β1 on VGSC function and firing properties in primary cortical neurons from *** found that TGF-β1 increased VGSC current density in a dose-and time-dependent manner,which was attributable to the upregulation of Nav1.3 *** VGSC current density and Nav1.3 expression were significantly abolished by preincubation with inhibitors of mitogen-activated protein kinase kinase(PD98059),p38 mitogen-activated protein kinase(SB203580),and Jun NH2-terminal kinase 1/2 inhibitor(SP600125).Interestingly,TGF-β1 significantly increased the firing threshold of action potentials but did not change their firing rate in cortical *** findings suggest that TGF-β1 can increase Nav1.3 expression through activation of the ERK1/2-JNK-MAPK pathway,which leads to a decrease in the firing threshold of action potentials in cortical neurons under pathological ***,this contributes to the occurrence and progression of neuroexcitatory-related diseases of the central nervous system.

Objective:To detect some common microbial agents of vaginal discharge and improve the current syndromic management of abnormal vaginal ***:A prospective study of female genital swabs collected from obstetrics and gynecology units of Aminu Kano Teaching Hospital,Kano,Nigeria and an analysed of microscopy,culture and sensitivity in the medical microbiology and parasitology laboratory of this hospital were conducted from December 2007 to December *** on epidemiologic indices were collected from the patients,using structured interviewer-administered ***:Eight hundred and forty Candida species were detected,constituting 60%(n=840) of 1 400 female genital discharge samples of microbial etiology in a total of 2 000 female genital samples *** distribution of vaginal candidiasis was tbe highest in young adults aged 21 to 30 years with 43%(n=360) of the total 840 cases. Pregnant women that presented with vaginal candidiasis constituted 40%(n=360) of the total 840 *** risk groups included the immuno-suppression with 24%(n=202),group on hormonal therapy with 15%(n=126) and broad spectrum antibiotics users with 16%(n=134).Conclusions: The results show that Candida is the most common cause of vaginitis and vulvo-vaginal candidiasis followed by *** in the young adults aged 21 to 30 years,pregnant mothers, immuno-suppression,contraceptive and broad spectrum antibiotic *** management of vaginal candidiasis and vulvo-vaginal candidiasis is recommended especially among the risk groups in order to avoid complications and reduce HIV transmission.

Skeletal muscle plays a significant role in both local and systemic energy metabolism. The current investigation aims to explore the role of the Bambi gene in skeletal muscle, focusing on its implications for muscle hypertrophy and systemic metabolism. We hypothesize that skeletal muscle-specific deletion of Bambi induces muscle hypertrophy, improves metabolic performance, and activates thermogenic adipocytes via the reprogramming of progenitor of i WAT, offering potential therapeutic strategies for metabolic syndromes. Leveraging the Chromatin immunoprecipitation(ChIP)-seq and bioinformatics analysis, Bambi gene is shown to be a direct target of HIF2α, which is further confirmed by ChIP-qPCR and promoter luciferase assay. Skeletal muscle-specific Bambi deletion led to significant muscle hypertrophy and improved metabolic parameters, even under high-fat diet conditions. This deletion induced metabolic reprogramming of stromal vascular fractions(SVFs) into thermogenic adipocytes, contributing to systemic metabolic improvements, potentially through the secretory ***, mice with skeletal muscle-specific Bambi deletion demonstrate resistance to high-fat diet-induced metabolic disorders, highlighting a potential therapeutic pathway for metabolic syndrome management. Thus, skeletal muscle-specific deletion of Bambi triggers muscle growth, enhances metabolic performance, and activates thermogenic adipocytes. These findings suggest Bambi as a novel therapeutic target for metabolic syndromes, providing new insights into the interaction between muscle hypertrophy and systemic metabolic improvement. The study underscores the potential of manipulating muscle physiology to regulate whole-body metabolism, offering a novel perspective on treating metabolic disorders.

Cell death can be classified into two categories: apoptosis and necrosis. Apoptotic pathway can beeither caspase-dependent or caspase-independent. Caspase-independent cytopathic effect (CPE) has beendescribed. In order to evaluate the pattern of HeLa cell death induced by Coxsackievirus B3 (CVB3)and whether apoptosis involves caspase activation, we co-cultivated HeLa cells with CVB3 and detectedthe cytopathic changes, the alteration of mRNA and protein expression of caspase-3 gene plus caspase-3activity, as well as analyzing DNA fragmentation before and after caspase-3 activity inhibition. Accordingto the results, we propose that CVB3 may induce apoptosis and necrosis in HeLa cells, the latter appearingmuch earlier. Caspase-3 is activated at the levels of both transcription and translation, and procaspase-3 isproteolytically cleaved, thus leading to the continuous increasing of both caspase-3 precursor protein and itssubunit. Objective To identify the antimicrobial resistance of commercial lactic acid bacteria present in microbial foods and drug additives by analyzing their isolated strains used for fermentation and probiotics. Methods Antimicrobial susceptibility of 41 screened isolates was tested with disc diffusion and E-test methods after species-level identification. Resistant strains were selected and examined for the presence of resistance genes by PCR. Results Distribution of resistance was found in different species. All isolates were susceptible to chloramphenicol, tetracycline, ampicillin, amoxicillin/clavulanic acid, cephalothin, and imipenem. In addition, isolates resistant to vancomycin, rifampicin, streptomycin, bacitracin, and erythromycin were detected, although the incidence of resistance to these antibiotics was relatively low. In contrast, most strains were resistant to ciprofloxacin, amikacin, trimethoprim/sulphamethoxazole, and gentamycin. The genes msrC, vanX, and dfrA were detected in strains of Enterococcus faecium, Lactobacillus plantarum, Streptococcus thermophilus, and Lactococcus lactis. Conclusion Antibiotic resistance is present in different species of probiotic strains, which poses a threat to food safety. Evaluation of the safety of lactic acid bacteria for human consumption should be guided by established criteria, guidelines and regulations.

Objective:To investigate the relationship between enterobiasis and enuresis before and after albendazole therapy among 632 children,aged,5-14 years,in Calabar,***:The scotch tape(cellotape) technique was used for the detection of eggs of Enterobius vermicularis (***) while questionnaire-based interviews were used for screening for anal itching and/or enuresis among study *** subjects found positive for Enterobius infection as confirmed by the presence of eggs and those with persistent anal itching(both enuretic and nonenuretic) were treated with 400 mg of albendazole given as a single *** treated subjects were re-assessed post-treatment to ascertain whether they were cured and also to determine their enuretic status(for the entireties).Results:The overall prevalences of Enterobius infection, anal itching,and enuresis prior to albendazole therapy were 6.8%,42.9%,and 35.6%respectively. There was no statistically significant difference in the prevalence of these parameters by the socio-economic class of subjects(P = 0.462,P - 0.647,and P = 0.610,respectively).The pretreatment prevalences of enuresis among Enterobius and anal itching-positive subjects were 53.5%and 49.8%,respectively versus 34.3%and 24.9%,respectively among their negative counterpart(P = 0.012 and P < 0.001,respectively).Four(20%) out of the 20 enuretic children found Enterobius egg-negative after albendazole therapy were equally cured while 8(40%) had reduction of their enuresis,thus giving a total resolution rate of 60%.Similarly,64.2%resolution (25%cure and 32.8%reduction) of enuresis was observed amongst 120 anal itching-cured/ enuretic *** status of the Enterobius-lanal itching-uncured subjects,on the other hand,remained unchanged ***:This study is suggestive of the involvement of *** in the aetiology of enuresis in *** presenting with, especially,uncomplicated enuresis should be screened for en

Objective:To determine the common bacterial causes of urinary tract infection and their antibiotic susceptibility pattern in AIDS patients versus non-AIDS ***:One thousand consecutive AIDS patients with signs and symptoms of AIDS and non-AIDS patients (served as control) each on admission were recruited into the study between January 2005 to January 2008,in Federal medical Center,*** samples were collected with sterile universal bottles and analysed with appropriate laboratory methods and antibiotic susceptibility test was carried out by disk diffusion technique in accordance with National Committee for Clinical Laboratory Standards(NCCLS,now CLSI) *** results were analysed using SPSS 11.0 statistical ***:Urine samples of AIDS patients with urinary infection had a more spectrum of micro-organisms including Candida organisms,Pseudomonas aeruginosa and Staphylococcus ***,Ceftazidime or Ciprofloxacin had a remarkably high anti-bacterial activity across the two study groups.A general resistance was recorded in *** and *** was no significant difference in antibiotic susceptibility patterns between AIDS and non- AIDS patients(P>0.05).Conclusions:A reduction in unnecessary use of antibiotics as well as infection control should be encouraged in our health facilities.
