Estimating SVCV waterborne transmission and predicting experimental epidemic development:A modeling study using a machine learning approach

Viral infectious diseases significantly threaten the sustainability of freshwater fish *** lack of studies on epidemic transmission patterns and mechanisms inhibits the development of containment strategies from the viewpoint of veterinary public *** study raises an epidemic mathematical model considering water transmission with the aim of analyzing the transmission process more *** basic reproduction number R0 was derived by the model parameter including the water transmission coefficient and was used for the analysis of the virus *** viremia of carp virus(SVCV)and zebrafish were used as model viruses and animals,respectively,to conduct the transmission *** through water was achieved by connecting two aquarium tanks with a water channel but blocking the fish movement between the *** the collected experimental data,we determined the optimal hybrid machine learning algorithm to analyze the transmission process using an established mathematical *** addition,future transmission was predicted and validated using the epidemic model and an optimal ***,the sensitivity of model parameters and the simulations of R0 variation were performed based on the modified complex epidemic *** study is of significance in providing theoretical guidance for minimizing R0 by manipulating model parameters with containment *** importantly,since the modified model and algorithm demonstrated better performance in handling freshwater fish transmission problems,this study advances the future application of transmissible disease modeling with larger datasets in freshwater fish aquaculture.

Pollen wall pattern in Arabidopsis

The pollen wall is a solid and variously sculptured structure. This pattern is determined inside a tetrad. During meiosis, the callose wall is formed outside of the meiocyte/microspore to form a tetrad. Then, primexine is deposited between the callose wall and the microspore plasma membrane which will become undulated. The sporopollenin deposits on top of the undulated membrane and develops into the pollen wall pattern, while the callose wall is gradually degraded. In recent years, much progress has been made in the study of pollen wall pattern formation, at both molecular and genetic levels. In this review,we summarize these achievements mainly in Arabidopsis.

Hepatocellular carcinoma is a common and fatal malignancy for which there is no effective systemic therapeutic ***(DHA),a derivative of artemisinin,has been shown to exert anti-tumor effects through the production of reactive oxygen species(ROS)and resultant mitochondrial ***,clinical translation is limited by several drawbacks,such as insolubility,instability and low ***,based on a nanomedicine-based delivery strategy,we fabricated mitochondria-targeted carrier-free nanoparticles coupling DHA and triphenylphosphonium(TPP),aiming to improve bioavailability and mitochondrial ***-TPP nanoparticles can be passively delivered to the tumor site by enhanced penetration and retention and then *** cytometry and Western blot analysis showed that DHA-TPP nanoparticles increased intracellular ROS,which increased mitochondrial stress and in turn upregulated the downstream Bcl-2 pathway,leading to *** vivo experiments showed that DHA-TPP nanoparticles exhibited anti-tumor effects in a mouse model of hepatocellular *** findings suggest carrier-free DHA-TPP nanoparticles as a potential therapeutic strategy for hepatocellular carcinoma.

Contrast-enhanced magnetic resonance imaging(CE-MRI) is a pivotal tool for global disease diagnosis and management. Since its clinical availability in 2009, the off-label use of ferumoxytol for ferumoxytol-enhanced MRI(FE-MRI) has significantly reshaped CE-MRI practices. Unlike MRI that is enhanced by gadolinium-based contrast agents, FE-MRI offers advantages such as reduced contrast agent dosage, extended imaging windows, no nephrotoxicity, higher MRI time efficiency and the capability for molecular imaging. As a leading superparamagnetic iron oxide contrast agent, ferumoxytol is heralded as the next generation of contrast agents. This review delineates the pivotal clinical applications and inherent technical superiority of FE-MRI, providing an avant-garde medical-engineering interdisciplinary lens, thus bridging the gap between clinical demands and engineering innovations. Concurrently, we spotlight the emerging imaging themes and new technical breakthroughs. Lastly, we share our own insights on the potential trajectory of FE-MRI, shedding light on its future within the medical imaging realm.

Polyphyly in 16S rRNA-based LVTree Versus Monophyly in Whole-genome-based CVTree

We report an important but long-overlooked manifestation of low-resolution power of 16S rRNA sequence analysis at the species level, namely, in 16S rRNA-based phylogenetic trees polyphyletic placements of closely-related species are abundant compared to those in genomebased phylogeny. This phenomenon makes the demarcation of genera within many families ambiguous in the 16S rRNA-based taxonomy. In this study, we reconstructed phylogenetic relationship for more than ten thousand prokaryote genomes using the CVTree method, which is based on wholegenome information. And many such genera, which are polyphyletic in 16S rRNA-based trees, are well resolved as monophyletic clusters by CVTree. We believe that with genome sequencing of prokaryotes becoming a commonplace, genome-based phylogeny is doomed to play a definitive role in the construction of a natural and objective taxonomy.

Regulation of tumor angiogenesis by the microtubule-binding protein CLIP-170

Angiogenesis,the expansion of preexisting blood vessels,is a complex process required for tumor growth and *** current antiangiogenic strategies have shown promising results in several cancer types,identification of additional antiangiogenic targets is required to improve the therapeutic ***,we show that the microtubule-binding protein CLIP-170(cytoplasmic linker protein of 170 kDa)is highly expressed in breast tumor samples and correlates positively with blood vessel *** of CLIP-170 signifi cantly impaired vascular endothelial tube formation and sprouting in vitro and inhibited breast tumor growth in mice by decreasing tumor *** data further show that CLIP-170 is important for the migration but not the proliferation of vascular endothelial *** addition,CLIP-170 promotes the polarization of endothelial cells in response to the angiogenic *** fi ndings thus demonstrate a critical role for CLIP-170 in tumor angiogenesis and suggest its potential as a novel antiangiogenic target.

Snake toxin Calciseptine as a natural antagonist of L-type calcium channel has potential drug values, but its structural information remains unknown. Here, we report the total chemical synthesis of Calciseptine by using hydrazide based native chemical ligation. The crystal structure of Calciseptine was determined by racemic protein crystallography technique. Compared to the structure of its homologous family protein, we found that Calciseptine is adopting a typical three-finger structure.

Background:Excessive heat exposure can lead to hyperthermia in humans,which impairs physical performance and disrupts cognitive *** heat is a known physiological stressor,it is unclear how severe heat stress affects brain physiology and ***:Eleven healthy participants were subjected to heat stress from prolonged exercise or warm water immersion until their rectal temperatures(T_(re))attained 39.5℃,inducing exertional or passive hyperthermia,*** a separate trial,blended ice was ingested before and during exercise as a cooling *** were compared to a control condition with seated rest(normothermic).Brain temperature(T_(br)),cerebral perfusion,and task-based brain activity were assessed using magnetic resonance imaging ***:T_(br)in motor cortex was found to be tightly regulated at rest(37.3℃±0.4℃(mean±SD))despite fluctuations in T_(re).With the development of hyperthermia,T_(br)increases and dovetails with the rising T_(re).Bilateral motor cortical activity was suppressed during high-intensity plantarflexion tasks,implying a reduced central motor drive in hyperthermic participants(T_(re)=38.5℃±0.1℃).Global gray matter perfusion and regional perfusion in sensorimotor cortex were reduced with passive *** function was poorer under a passive hyperthermic state,and this could relate to compromised visual processing as indicated by the reduced activation of left lateral-occipital ***,ingestion of blended ice before and during exercise alleviated the rise in both T_(re)and T_(bc)and mitigated heat-related neural ***:Severe heat exposure elevates T_(br),disrupts motor cortical activity and executive function,and this can lead to impairment of physical and cognitive performance.

Chaetocin reactivates the lytic replication of Epstein-Barr virus from latency via reactive oxygen species

Oxidative stress, regarded as a negative effect of free radicals in vivo, takes place when organisms suffer from harmful stimuli. Some viruses can induce the release of reactive oxygen species （ROS） in infected cells, which may be closely related with their pathogenicity. In this report, chaetocin, a fimgal metabolite reported to have antimicrobial and cytostatic activity, was studied for its effect on the activation of latent Epstein-Barr virus （EBV） in B95-8 cells. We found that chaetocin remarkably up-regulated EBV lytic transcription and DNA replication at a low concentration （50 nmol L-l）. The activation of latent EBV was accompanied by an increased cellular ROS level. N-acetyl-L-cysteine （NAC）, an ROS inhibitor, suppressed chaetocin-induced EBV activation. Chaetocin had little effect on histone H3K9 methylation, while NAC also significantly reduced H3K9 methylation. These results suggested that chaetocin reactivates latent EBV primarily via ROS pathways.
