Medium effects on micropropagation and genetic stability of Citrullus lanatus oleaginous type

To regenerate adventitious shoots from the cotyledon proximal parts of Citrullus lanatus (Thunb.) Matsum. and Nakai ssp. mucosospermus (Fursa) oleaginous type, different concentrations of MS mineral elements, sucrose, 6-benzylaminopurine (BAP) and agar were tested. Shoot induction proved to depend on the interaction between levels of sucrose, BAP and MS mineral elements in the medium. The medium containing 3/2 strength of MS mineral elements, 35 g/l sucrose and 1 mg/l BAP solidified with 6 g/l agar allowed the production of numerous shoots without a callus phase. After 3 weeks of culture, 76.7% of the cotyledon proximal parts induced shoots with an average of 12.26 shoots per explant and a mean shoot length of 17.13 mm. The induced shoots were directly rooted and thus complete plants ready for acclimatization were obtained using a two steps procedure. Depending on the genotype, the shoot induction from cotyledon proximal parts ranged from 54% to 96%. Rooted plantlets were acclimatized and transferred to field, where they grew well, developed flowers and fruits like seeded plants. The assessment of the genetic stability of the in-vitro-regenerated plantlets by means of an Amplified Fragment Length Polymorphism (AFLP) analysis with the combination of 5 primers revealed no differences between regenerated plantlets and mother plants.

Antibiotics/antibacterial drug use, their marketing and promotion during the post-antibiotic golden age and their role in emergence of bacterial resistance

During the post-antibiotic golden age, it has seen a massive antibiotic/antibacterial production and an increase in irrational use of these few existing drugs in the medical and veterinary practice, food industries, tissue cultures, agriculture and commercial ethanol production globally. The irrational drug use has been further exacerbated by the increased marketing and promotion of these drugs by the pharmaceutical companies thus increasing their accessibility in the public and hence their improper use. The lack of production and introduction of the newer and effective antibiotic/antibacterial drugs in clinical practice in the post-antibiotic golden age has seen an increase in the emergence of the resistant pathogenic bacterial infections creating a significant problem in the global health of humankind. The massive productions of the antibiotic/antibacterial drugs have contributed to the poor disposal of these drugs and hence many of them are discharged in various water bodies contributing to the environmental antibiotic/antibacterial drug pollution. In the environment, these drugs exert pressure on the environmental bacteria by destroying useful bacteria that are responsible for the recycling of the organic matter and as well as promoting the selection of the resistant pathogenic bacteria that can spread in human and animal population thus causing an increase in the observed bacterial disease burden and hence a significant global public health problem. The resistant bacterial diseases lead to the high cost, increased occurrence of adverse drug reactions, prolonged hospitalization, the exposure to the second- and third-line drugs like in MDR-TB and XDR-TB that leads to toxicity and deaths as well as the increased poor production in agriculture and animal industry and commercial ethanol production.

Single top production associated with a neutral scalar at LHC in topcolor-assisted technicolor

The topcolor-assisted technicolor （TC2） model predicts a number of neutral scalars like the top-pion （πt^0） and the top-Higgs （ht^0）. These scalars have flavor-changing neutral-current （FCNC） top quark couplings, among which the top-charm transition couplings may be sizable. Such FCNC couplings induce single top productions associated with a neutral scalar at the CERN Large Hadron Collider （LHC） through the parton processes cg→ tπt^0 and cg→ tht^0. In this paper we examine these productions and find their production rates can exceed the 3σ sensitivity of the LHC in a large part of parameter space. Since in the Standard Model and the minimal supersymmetric model such rare productions have unobservably small production rates at the LHC, these rare processes will serve as a good probe for the TC2 model.

Assessing Crop Yield Sustainability under the Climatic and Bioclimatic Change in the Area of Palestine

We analyzed plant production such as olive, grape and fig in several Palestinian Occupied Territories, and studied the correlation analyses between productions and climatic, bioclimatic features in the territory. This work examines the following climate factors: average temperature, precipitation, soil water reserve and water deficit;and bioclimatic parameters such as compensated thermicity index, ombrothermic index and continentally index. The data used were from nine meteorological stations of the Palestinian Meteorological department. The study was based on the correlation analyses between olive, grape and fig production in nine plots: Hebron, Ramallah, Nablus, Jenin, Jericho, Bethlehem, Jerusalem and Tulkarem over fourteen years. The analysis of variance revealed a significant influence of the variables temperature and compensated thermicity index on the production of olive and fig, and a low influence on grape production. When we applied canonical correspondence analysis (CCA), the Nablus, Jenin, Jericho and Gaza plots were most affected by the climate factors as temperature, and bioclimate factors as compensated thermicity index, whereas, the Hebron, Bethlehem, Jerusalem, Tulkarem and Ramallah plots showed the influence of ombrothermic index, continentally index, precipitation, water deficit and soil water reserve.

J/ψ Production in Ultra-Peripheral pp Collisions through aTwo-Photon Process

Using the photon spectrum of a high energy charged patton, we investigate the J/ψ production via the two photon interaction mechanism in ultra-peripheral pp collisions. It is shown that the contributions from the direct electromagnetic and the corresponding fragmentation processes are meaningful for the J/ψ productions in ultra-peripheral pp collisions at LHC energy.

Repeated-batch Cultivation of Encapsulated Monascus purpureus by Polyelectrolyte Complex for Natural Pigment Production

In general,productions of natural pigment in submerged microorganism culture were much less than that in solid-state fermentation,because the solid-state culture can provide a support carrier for the mycelium. To improve natural pigment production,the cultivation of Monascus purpureus in submerged encapsulated cell was investigated. Monascus purpureus immobilized in polyelectrolyte complex(PEC) microcapsules,which were pre-pared by sodium cellulose sulphate(NaCS) and poly-dimethyl-diallyl-ammonium chloride(PDMDAAC),was a good substitute for submerged cell culture because it mimicked the solid-state environment. The repeated-batch process with encapsulated cells was studied in flasks and a bubble column. The results indicated that the bubble column was more suitable for the encapsulation culture than the shaking flasks because of its good mass transfer performance and minor shear stress on cells. Owing to the protection of the microcapsule's membrane,Monascus purpureus in microcapsules increased approximately three times over that in free cell culture with negligible cell leakage to the medium. The pigment production in the bubble column finally reached 3.82(OD500) ,which was two times higher than in free cell culture. In addition,the duration of each batch was shortened to 15% of that in free cell culture.

白介素-17在沙眼衣原体呼吸道感染中的早期产生可提高局部白介素-6和巨噬细胞炎性蛋白-2的表达

目的:探讨炎症性细胞因子白介素-17(interleukin-17,IL-17)在沙眼衣原体呼吸道感染中的早期产生与细胞分泌白介素-6(interleukin-6,IL-6)和巨噬细胞炎性蛋白-2(macrophage inflammatory protein2,MIP-2)之间的关系。方法:在体内,用沙眼衣原体小鼠肺炎株(MoPn)通过鼻腔感染小鼠,用免疫荧光法(immunofluoresent assay,IFA)检测衣原体在肺组织的生长;通过酶联免疫吸附测定法(enzyme-linked immunosorbent assay,ELISA)检测小鼠肺组织IL-17,IL-6和MIP-2的表达,未感染小鼠作为对照组;在体外,用重组鼠IL-17(recombinant murine IL-17,rmIL-17)预处理L929细胞24h,rmIL-17预处理并感染MoPn的细胞作为实验组,未用rmIL-17预处理但感染MoPn的细胞作为对照组,24h后收集上清液,利用ELISA检测IL-6和MIP-2的产生,细胞则通过IFA进行衣原体包涵体计数(inclusion-forming unit,IFU)。结果:体内实验表明,MoPn感染后第1天,肺组织有衣原体生长,感染后第8天达高峰,以常用对数(lg)计数每个肺组织的IFU为6.49±0.19。感染后第2天,IL-17在小鼠肺组织中的产生达峰值(83.0±35.8)ng/L,并很快下降。IL-6在感染后第3天达到峰值(3.98±0.04)μg/L,而MIP-2则在第8天出现高峰(2.19±0.71)μg/L。体外实验发现,分别用20,100和500μg/L不同剂量的rmIL-17预处理细胞后再感染衣原体MoPn24h,细胞上清液中IL-6含量分别为(531.65±24.40),(629.95±7.71)和(646.51±35.92)ng/L,与对照组(55.10±16.54)ng/L比较差异有统计学意义(P<0.01),细胞上清液中MIP-2含量分别为(107.21±28.40),(181.95±25.51)和(221.9±17.32)ng/L,与对照组(13.71±0.84)ng/L比较差异有统计学意义(P<0.05)。单独rmIL-17对细胞并无刺激作用,rmIL-17对感染细胞衣原体包涵体生长无直接抑制作用。结论:IL-17在衣原体呼吸道感染中早期出现,rmIL-17能诱导感染MoPn的L929细胞分泌IL-6和MIP-2。IL-17在衣原体呼吸道感染早期可能通过诱导IL-6和MIP-2的产生,在宿主抵御细胞内病原菌感染中发挥重要的作用。
