Gastrodin (GAS)， the principal bioactive composition of Gastrodia elata Blume， has potential for pharmaceutical applications. Several studies in recent years have shown that GAS may enhance neurotrophic benefits， reduce inflammation， and act as an antioxidant. In this study， we sought to identify the molecular mechanisms underlying the protective benefits of GAS against colitis induced by dextran sodium sulfate (DSS) in mice. GAS (200 mg/kg) significantly ameliorated the severity of colitis in mice caused by DSS， as evidenced by an increase in colon length， a reduction in disease activity index， a decrease in tissue damage， and a reduction in body weight loss. Additionally， GAS inhibited DSS-induced hyperactivation of inflammation-related NF-κB signaling pathways to reduce the production of inflammatory mediators， thereby mitigating the inflammatory response in mice. Furthermore， the administration of GAS restored the function of the gastrointestinal barrier by increasing the count of goblet cells， as well as the levels of tight junction-associated proteins， including ZO-1， Occludin， and Claudin-3. GAS also influenced the overall richness of the gut microbiota， as shown by 16S rRNA sequencing analysis， consequently boosting the proliferative rate of probiotic species， such as Lachnospiraceae and Muribaculaceae， while reducing the richness of harmful bacteria including Escherichia＿Shigella， Enterobacteriaceae， Bacteroidaceae， and Bacteroides. GAS (200 mg/kg) alleviated ulcerative colitis (UC) by modulating gut dysbiosis， as demonstrated by a fecal microbial transplantation (FMT) test. Furthermore， inflammatory damage induced by lipopolysaccharide (LPS) was averted in RAW264.7 cells by GAS administration， hence preventing the NF-κB signaling pathway from being activated in these experimental conditions conducted in vitro. Neurological injury caused by ischemic stroke is a major cause of permanent disability and death. The currently available neuroprotective drugs fail to achieve desired therapeutic efficacy mainly due to short circulation half-life and poor blood−brain barrier (BBB) permeability. For that, an edaravone-loaded pH/glutathione (pH/GSH) dual-responsive poly(amino acid) nanogel (NG/EDA) was developed to improve the neuroprotection of EDA. The nanogel was triggered by acidic and EDA-induced high-level GSH microenvironments, which enabled the selective and sustained release of EDA at the site of ischemic injury. NG/EDA exhibited a uniform sub-spherical morphology with a mean hydrodynamic diameter of 112.3 ± 8.2 nm. NG/EDA efficiently accumulated at the cerebral ischemic injury site of permanent middle cerebral artery occlusion (pMCAO) mice, showing an efficient BBB crossing feature. Notably, NG/EDA with 50 µM EDA significantly increased neuron survival (29.3%) following oxygen and glucose deprivation by inhibiting ferroptosis. In addition, administering NG/EDA for 7 d significantly reduced infarct volume to 22.2% ± 7.2% and decreased neurobehavioral scores from 9.0 ± 0.6 to 2.0 ± 0.8. Such a pH/GSH dual-responsive nanoplatform might provide a unique and promising modality for neuroprotection in ischemic stroke and other central nervous system diseases.

Among minimally invasive surgical procedures,colorectal surgery is associated with a notably higher incidence of incisional hernia(IH),ranging from 1.7% to 24.3%.This complication poses a significant burden on the healthcare system annually,necessitating urgent attention from *** a study published in the World Journal of gastrointestinal surgery,Fan et al compared the incidence of IH among 1614 patients who underwent laparoscopic colorectal surgery with different extraction site locations and evaluated the risk factors associated with its *** editorial analyzes the current risk factors for IH after laparoscopic colorectal surgery,emphasizing the impact of obesity,surgical site infection,and the choice of incision location on its ***,we summarize the currently available preventive measures for *** the low surgical repair rate and high recurrence rate associated with IH,prevention deserves greater research and attention compared to treatment.

Background Stomach cancer is among the most commonly occurring malignancies worldwide. It would be beneficial to develop a urine-based assay whereby patients with undiagnosed stomach cancer could be screened and their cancer detected in the eadiest stages. Methods A urinary metabonomics method based on ultra-performance liquid chromatography combined with quadruple time-of-flight mass spectrometry was used to analyze urine samples from patients with stomach cancer and healthy controls. Results Statistical analysis revealed a clear separation of patients and healthy controls using the aforementioned methodology. Some significantly changed metabolites were identified. Conclusions Use of the metabonomics method in patients with stomach cancer could effectively detect distinct changes in urinary metabolites and had the capacity to detect cancer; therefore, it may be a valuable tool in earlier diagnosis. Furthermore, the detection and identification of altered metabolites in the current study may help elucidate possible mechanisms involved in stomach cancer.

Double Emulsion Droplets as Microreactors for Synthesis of Magnetic Macroporous Polymer Beads

An easy method is presented to fabricate monodisperse magnetic macroporous polymer beads（MMPBs）. Waterin-oil high internal phase emulsion（HIPE） is prepared by emulsifying aqueous iron ions solution in an oil phase containing monomers. The HIPE is introduced into a simple microfluidic device to fabricate monodisperse（water-in-oil）-in-water double emulsion droplets. The droplets serve as microreactors to synthesize Fe3O4 nanoparticles and are on-line polymerized to form MMPBs. The prepared MMPBs display uniform size, interconnected porous structure, superparamagnetic behavior and uniform distribution of Fe3O4 in polymer matrix. The MMPBs are characterized by scanning electron microscopy（SEM）, Fourier transform infrared spectroscopy（FTIR）, X-ray diffraction（XRD）, transmission electron microscopy（TEM）, vibrating sample magnetometry（VSM）. We believe that this method is a universal technique in preparing macroporous nanocomposite beads.

Influence of Mixed Common Solvent on the Co-assembled Morphology of PS-b-PEO and CdS Quantum Dots

The hybrid structures of polystyrene-b-poly（ethylene oxide）（PS-b-PEO） block copolymer and inorganic nanoparticles with good stability and biocompatibility have potential applications in drug delivery and bioimaging. Spherical co-assemblies of PS120-b-PEO318 and oleylamine-capped CdS quantum dots （QDs） are produced successfully in this work by adding water to a mixed common solvent, such as N,N-dimethylmethanamide （DMF）/chloroform, DMF/tetrahydrofuran （THF）, or DMF/toluene. The energy dispersive X-ray （EDX） spectrum indicates that QDs are located at the interface between the core and shell of the spherical co-assemblies. The co-assembly process during water addition is traced by transmission electron microscopy （TEM） and turbidity measurement. Spherical co-assemblies are formed through budding from bilayers of the block copolymer and QDs. The morphology of the co-assemblies is related to the miscibility of the QD-dispersing solvents with water and the morphology changes from a spherical to a vesicle-like structure with DMF/toluene. Increasing THF content in the mixed solvent causes morphological transitions from spherical co-assemblies to multi-branched cylinders and micelles where QDs are located in the central core. Increasing chloroform content yields vesicle-like structures with protruding rods on the surface. The mechanism of the morphological transitions is also discussed in detail.
