BACKGROUND In microwave ablation(MWA), although computed tomography(CT) scanning can overcome gas interference, it cannot achieve real-time localization. Therefore, the puncture technique is more important in CT-guided *** To compare the fine needle-assisted puncture(FNP) positioning technique and the conventional puncture(CP) technique for the safety and efficacy of CT-guided MWA in treating hepatocellular carcinoma(HCC).METHODS This retrospective study included 124 patients with 166 tumor nodules from February 2018 and June 2021. Seventy patients received CT-guided MWA under the FNP technique(FNP group), and 54 patients received MWA under the CP technique(CP group). Intergroup comparisons were made regarding local tumor progression(LTP), recurrence-free survival(RFS), overall survival(OS), and complications. The influencing variables of LTP and RFS were analyzed through univariate and multivariate *** The 1-, 2-, and 3-year cumulative incidences of LTP in the FNP group were significantly lower than those in the CP group(7.4%, 12.7%, 21.3% vs 13.7%, 32.9%, 36.4%;P = 0.038). The 1-, 2-, and 3-year RFS rates in the FNP group were significantly higher than those in the CP group(80.6%, 73.3%, 64.0% vs 83.3%,39.4%, and 32.5%, respectively;P = 0.008). The FNP technique independently predicted LTP and RFS. Minor complications in the FNP group were lower than those in the CP group(P < 0.001). The difference in median OS was insignificant between the FNP and CP groups(P = 0.229).CONCLUSION The FNP technique used in CT-guided MWA may improve outcomes in terms of LTP, RFS, and procedure-related complications for HCC.

Purpose: Triple-negative breast cancer(TNBC) is a breast cancer type with poor prognosis due to the absence of targets for the treatment of endocrine and anti-HER-2 therapy. Furthermore, only about 20% of TNBC are sensitive to conventional chemotherapy. Therefore, this study aimed to evaluate the clinical efficacy of adjuvant dendrtic cell(DC)-cytokineinduced killer cells(CIK) immunotherapy combined with chemotherapy with/without radiotherapy in patients with postoperative ***: From December 1,2006 to February28,2011, 68 patients with postoperative TNBC were included in this retrospective study: 43 cases received chemotherapy with/without radiotherapy;25 cases received DC-CIK cells infusion combined with chemotherapy with/without ***: Survival analysis showed the 5-years disease-free survival(DFS) rate was significantly higher in the DC-CIK group compared with the non- DC-CIK group(P=0.049). Further subgroup analysis stratified according to tumor–node–metastasis(TNM) stages showed that the 5-years DFS was significantly increased in DC-CIK group with advanced TNM stages(including IIb,III,IV TNM stage) than the non- DC-CIK, and 5-year OS rate in advanced-stage subgroup was also improved although it only approved borderline statistical significance(P=0.051).Conclusions: Our results demonstrated that TNBC patients with advanced TNM stages could benefitted from adjuvant DC-CIK cells infusion combined with conventional therapy in terms of preventing disease relapse and increasing overall survival.

AIM To investigate the effects of different levels of expression of CDK5RAP3 and DDRGK1 on long-term survival of patients undergoing radical *** The expression of CDK5RAP3 and DDRGK1 was detected by immunohistochemistry in 135 patients who received standard gastrectomy were enrolled in the study. Western Blot was used to detect the expression of CDK5RAP3 and DDRGK1 in gastric cancer and its adjacent tissues and cell lines. The correlations between the expression of CDK5RAP3 and DDRGK1 and clinicopathological factors were analyzed, and the value of each parameter to the prognosis of the patients was compared. Receiver operating characteristic analysis was used to compare the accuracy of the prediction of clinical outcome by the *** CDK5RAP3 and DDRGK1 expression was downregulated in the gastric cancer compared to its respective adjacent non-tumor tissues. The expression of CDK5RAP3 was closely related to the age of the patients(P = 0.035) and the T stage of the tumor(P = 0.017). The expression of DDRGK1 was correlated with the sex of the patients(P = 0.080), the degree of tumor differentiation(P = 0.036), the histological type(P = 0.036) and the N stage of the tumor(P = 0.014). Low expression CDK5RAP3 or DDRGK1 is a poor prognostic factor for gastric cancer patients. Prognostic analysis showed that the co-expression of CDK5RAP3 and DDRGK1 was an independent prognostic factor correlating with the overall survival of gastric cancer patients. Combined expression analysis of CDK5RAP3 and DDRGK1 may provide a more accurate prognostic value for overall survival.

Objective: ARA55(androgen receptor coactivator 55 kDa protein) was first identified as a TGF β1-inducible protein and function as a molecular scaffold in coordinating protein-protein interactions. Herein, we focused on validating the functional role of ARA55 in TGF β1-induced epithelial-mesenchymal transition(EMT) in human CNE2 nasopharyngeal carcinoma cells. Methods: Expression of ARA55 in the CNE2 cells was stimulated by TGF β1(5 ng/ml), and a commercial RNA interference plasmid(siRNA-ARA55) was utilized to silence ARA55 expression in response to TGF β1 induction. Results: Our results manifested that forced expression of ARA55 enhances growth as well as migration and invasion of the CEN2 cells. In contrast, cells depleted of ARA55 resulted in suppressed cell proliferation and metastasis capability, along with a down-regulation of N-cadherin and up-regulation of Claudin-1. Further co-immunoprecipitation analysis exhibited that induced ARA55 yields a direct physical interaction with Smad7 in TGF β1 signaling. Conclusions: Our data demonstrate that ARA55 exerts a causative role and functions as a critical regulator in TGF β1-mediated EMT in CNE2 nasopharyngeal carcinoma cells through binding with Smad7.
