The temporal-spatial expression and functional analysis of three gustatory receptor genes in Solenopsis invicta using sweet and bitter compounds

The insect gustatory system participates in identifying potential food sources and avoiding toxic compounds.During this process,gustatory receptors(GRs)recognize feeding stimulant and deterrent compounds.However,the GRs involved in recognizing stimulant and deterrent compounds in the red imported fire ant,Solenopsis invicta,remain unknown.Therefore,we conducted a study on the genes SinvGR1,SinvGR32b,and SinvGR28a to investigate the roles of GRs in detecting feeding stimulant and deterrent compounds.In this current study,we found that sucrose and fructose are feeding stimulants and the bitter compound quinine is a feeding deterrent.The fire ant workers showed significant behavior changes to avoid the bitter taste in feeding stimulant compounds.Reverse transcription quantitative real-time polymerase chain reaction results from developmental stages showed that the SinvGR1,SinvGR32b,and SinvGR28a genes were highly expressed in fire ant workers.Tissue-specific expression profiles indicated that SinvGR1,SinvGR32b,and SinvGR28a were specifically expressed in the antennae and foreleg tarsi of workers,whereas SinvGR32b gene transcripts were also highly accumulated in the male antennae.Furthermore,the silencing of SinvGRI or SinvGR32b alone and the co-silencing of both genes disrupted worker stimulation and feeding on sucrose and fructose.The results also showed that SinvGR28a is required for avoiding quinine,as workers with knockdown of the SinvGR28a gene failed to avoid and fed on quinine.This study first identified stimulant and deterrent compounds of fire ant workers and then the GRs involved in the taste recognition of these compounds.This study could provide potential target gustatory genes for the control of the fire ant.

Syntheses and properties of associative acrylamide copolymers containing short hydrophobic chains used in a friction reducer for slick-water fracturing

Two allyldimethylalkyl quaternary ammonium salt（AQAS） monomers, N,N-dimethylallylphenylpropylammonium bromide（AQAS1） and N,N-dimethylallylnonylammonium bromide（AQAS2）, were synthesized and used to prepare modified polyacrylamide materials. Two new drag reducers were synthesized from acrylamide（AM）, sodium acrylate（Na AA） and a cationic modified monomer（AQAS1 or AQAS2） via aqueous solution polymerization, and the copolymers were named P（AM/Na AA/AQAS1） and P（AM/Na AA/AQAS2）, respectively. The structures of the drag reduction agents were confirmed by IR and1H NMR spectroscopies. The molecular weight（Mw） of P（AM/Na AA/AQAS1） was 1.79 × 10～6g/mol. When the copolymer concentration was 1000 mg/L and the flow rate was 45 L/min, in fresh water the highest drag reduction rate was 75.8%, in 10,000 mg/L Na Cl solution the drag reduction rate decreased to 72.9%. The molecular weight of P（AM/Na AA/AQAS2） was 3.17 × 10～6g/mol. When the copolymer concentration was500 mg/L and the flow rate was 45 L/min, the drag reduction rate reached 75.2%, and in 10,000 mg/L Na Cl solution the drag reduction rate was 73.3%, decreased by approximately 1.9%. The drag reduction rate for partially hydrolyzed polyacrylamide（HPAM） was also investigated, and the results showed that the drag reduction rates for 500 and 1000 mg/L HPAM solutions were merely 43.2% and 49.0% in brine, respectively. Compared with HPAM, both of the above copolymers presented better drag reduction capacities.

As an environmental pollutant,ambient fine particulate matter (PM_(2.5)) was linked to cardiovascular diseases.The molecular mechanisms underlying PM_(2.5)-induced extrapulmonary disease has not been elucidated clearly.In this study the ambient PM_(2.5)exposure mice model we established was to explore adverse effects of vessel and potential mechanisms.Long-term PM_(2.5)exposure caused reduced lung function and vascular stiffness in mice.And chronic PM_(2.5)induced migration and epithelial-mesenchymal transition (EMT) phenotype in BEAS-2B cells.After PM_(2.5)treatment,the circRNAs and mRNAs levels of exosomes released by BEAS-2B cells were detected by competing endogenous RNA (ceRNA) array,which contained 1664 differentially expressed circRNAs (DE-circRNAs) and 308 differentially expressed m RNAs (DE-mRNAs).By bioinformatics analysis on host genes of DE-circRNAs,vascular diseases and some pathways related to vascular diseases including focal adhesion,tight junction and adherens junction were enriched.Then,ceRNA network was constructed,and DE-mRNAs in ceRNA network were conducted functional enrichment analysis by Ingenuity Pathway Analysis,which indicated that hsa_circ_0012627,hsa_circ_0053261and hsa_circ_0052810 were related to vascular endothelial dysfunction.Furthermore,it was verified experimentally that Exo^(PM2.5)could induce endothelial dysfunction by increased endothelial permeability and decreased relaxation in vitro.In present study,we investigated in-depth knowledge into the molecule events related to PM_(2.5) toxicity and pathogenesis of vascular diseases.

Juvenile hormone induces reproduction via miR-1175-3p in the red imported fire ant, Solenopsis invicta

Juvenile hormone(JH)acts in the regulation of caste differentiation between queens and workers(i.e.,with or without reproductive capacity)during vitellin synthesis and oogenesis in social insects.However,the regulatory mechanisms have not yet been elucidated.Here,we identified a highly expressed microRNA(miRNA),miR-1175-3p,in the red imported fire ant,Solenopsis invicta.We found that miR-1175-3p is prominently present in the fat bodies and ovaries of workers.Furthermore,miR-1175-3p interacts with its target gene,broad-complex core(Br-C),in the fat bodies.By utilizing miR-1175-3p agomir,we successfully suppressed the expression of the Br-C protein in queens,resulting in reduced vitellogenin expression,fewer eggs,and poorly developed ovaries.Conversely,decreasing miR-1175-3p levels led to the increased expression of Br-C and vitellogenin in workers,triggering the“re-development”of the ovaries.Moreover,when queens were fed with JH,the expression of miR-1175-3p decreased,whereas the expression of vitellogenin-2 and vitellogenin-3 increased.Notably,the suppression of fertility in queens caused by treatment with agomir miR-1175-3p was completely rescued by the increased vitellogenin expression induced by being fed with JH.These results suggest the critical role of miR-1175-3p in JH-regulated reproduction,shedding light on the molecular mechanism underlying miRNA-mediated fecundity in social insects and providing a novel strategy for managing S.invicta.

The storage and utilization of carbohydrates in response to elevation mediated by tree organs in subtropical evergreen broad-leaved forests

Global climate change can affect tree growth and carbon sink function by influencing plant carbohydrate synthesis and utilization,while elevation can be used as an ideal setting under natural conditions to simulate climate change effects.The effect of elevation on tree growth may depend on organ type.However,the allocation patterns of nonstructural and structural carbohydrates(NSCs and SCs,respectively)in different tree organs and their response to elevation remain unclear.We selected four dominant tree species,Schima superba,Castanopsis eyrei,Castanopsis fargesii and Michelia maudiae,along an elevation gradient from 609 to 1,207 m in subtropical evergreen broad-leaved forests and analyzed leaf,trunk,and fine root NSCs,carbon(C),nitrogen(N)and phosphorus(P)concentrations and the relative abundance of SCs.Leaf NSCs increased initially and then decreased,and trunk NSCs increased with increasing elevation.However,root NSCs decreased with increasing elevation.The relative abundance of SCs in leaves and trunks decreased,while the relative abundance of root SCs increased with increasing elevation.No significant correlations between SCs and NSCs in leaves were detected,while there were negative correlations between SCs and NSCs in trunks,roots,and all organs.Hierarchical partitioning analysis indicated that plant C/N and C/P were the main predictors of changes in SCs and NSCs.Our results suggest that tree organs have divergent responses to elevation and that increasing elevation will inhibit the aboveground part growth and enhance the root growth of trees.A tradeoff between the C distribution used for growth and storage was confirmed along the elevation gradient,which is mainly manifested in the"sink"organs of NSCs.Our results provide insight into tree growth in the context of global climate change scenarios in subtropical forest ecosystems.

Investigation of rarely studied gravel layers found in the loess in Shandong Province,eastern China,reveals the fabric characteristics of two gravel layers(G1,G2)and the sedimentary characteristics of loess at the typical and well-preserved Heiyu section(HY),where,to determine the paleoclimatic changes during Marine Isotope Stage 3a.Optically stimulated luminescence dates of the HY formation range from 0.26±0.02 ka to 39.00±2.00 ka.In addition,the ages of G1 and G2 were estimated using the Bayesian model to be 39.60-40.50 and 29.00-29.50 ka.G1 and G2 are mainly composed of fine and medium gravel,both of which were subangular to subrounded limestone,with gravel directions to NE and E.The average flow velocity,average depth,and flood peak flow of G1 are 1.10 m/s,0.49 m,and 37.04 m^(3)/s,respectively,calculated using the flow energy method,whereas those of G2 are 0.98 m/s,0.38 m,and 18.38 m^(3)/s,respectively.Analysis of climate proxy indices show that the sedimentary environment of the gravel and loess in HY might be a regional response to global change.

The biosynthesis of alarm pheromone in the wheat aphid Rhopalosiphum padi is regulated by hormones via fatty acid metabolism

Aphids are major insect pests in agriculture and forestry worldwide.Following attacks by natural enemies,many aphids release an alarm pheromone to protect their population.In most aphids,the main component of the aphid alarm pheromone(AAP)is the sesquiterpene hydrocarbon(E)-β-farnesene(EβF).However,the mechanisms behind its biosynthesis and regulation remain poorly understood.In this study,we used the bird cherry–oat aphid Rhopalosiphum padi,which is an important wheat aphid,to investigate the regulatory mechanisms of EβF biosynthesis.Our results showed that EβF biosynthesis occurs during the mature embryo period and the molting period of the 1st-and 2nd-instar nymphs.Triglycerides provide the prerequisite material for EβF production and release.Based on transcriptome sequencing,RNAi analysis,hormone treatments,and quantitative measurements,we found that the biosynthesis of EβF utilizes acetyl coenzyme A produced from fatty acid degradation,which can be suppressed by juvenile hormone but it is promoted by 20-hydroxyecdysone through the modulation of fatty acid metabolism.This is the first systemic study on the modulation of EβF production in aphids.The results of our study provide insights into the molecular regulatory mechanisms of AAP biosynthesis,as well as valuable information for designing potential aphid control strategies.

High temperature treatment induced production of unreduced 2n pollen in Camellia oleifera

Unreduced gametes through chromosome doubling play a major role in the process of plant polyploidization.Our previous work confirmed that Camellia oleifera can produce natural 2n pollen,and it is possible to induce the 2n pollen formation by high temperature treatment.This study focused on the optimization of the 2n pollen induction technique and the mechanisms of high temperature-induced2n pollen formation in C.oleifera.We found that the optimal protocol for inducing 2n pollen via high temperature was to perform 45℃with4 h at the prophaseⅠstage of the pollen mother cells(PMCs).Meanwhile,high temperature significantly decreased the yield and fertility of2n pollen.Through the observation of meiosis,abnormal chromosome and cytological behaviour was discovered under high-temperature treatment,and we confirmed that the formation of 2n pollen is caused by abnormal cell plate.Based on weighted gene co-expression network analysis,fifteen hub genes related to cell cycle control were identified.After male flower buds were exposed to heat shock,polygalacturonase gene(CoPGX3)was significantly upregulated.We inferred that high temperature causes the CoPGX3 gene to be overexpressed and that CoPGX3 is redistributed into the cytosol where it degrades cytoplasmic pectin,which leads to an abnormal cell plate.Furthermore,abnormal cytokinesis resulted in the formation of dyads and triads,and PMCs divided to produce 2n pollen.Our findings provide new insights into the mechanism of 2n pollen induced by high temperature in a woody plant and lay a foundation for further ploidy breeding of C.oleifera.

Synergistic effects of the entomopathogenic fungus Isaria javanica and low doses of dinotefuran on the efficient control of the rice pest Sogatella furcifera

The rice planthopper,Sogatella furcifera,is a piercing-sucking insect pest of rice,Oryza sativa.It is responsible for significant crop yield losses,and has developed moderate to high resistance to several commonly used chemical insecticides.We investigated the effects of the insect fungal pathogen Isaria javanica,alone and in combination with the chemical insecticide dinotefuran,on S.furcifera under both laboratory and field conditions.Our results show that I.javanica displays high infection efficiency and mortality for different stages of S.furcifera,reducing adult survival,female oviposition and ovary development.Laboratory bioassays showed that the combined use of I.javanica with a low dose(4-16 mg L^(-1))of dinotefuran resulted in higher mortality in S.furcifera than the use of I.javanica or dinotefuran alone.The combined treatment also had more significant effects on several host enzymes,including superoxide dismutase,catalase,peroxidase,and prophenol oxidase activities.In field trials,I.javanica effectively suppressed populations of rice planthoppers to low levels(22-64%of the level in untreated plots).Additional field experiments showed synergistic effects,i.e.,enhanced efficiency,for the control of S.furcifera populations using the combination of a low dose of I.javanica(1×10^(4) conidia mL^(-1))and a low dose of dinotefuran(~4.8-19.2%of normal field use levels),with control effects of>90%and a population level under 50 insects per 100 hills at 3-14 days post-treatment.Our findings indicate that the entomogenous fungus I.javanica offers an attractive biological control addition as part of the integrated pest management(IPM)practices for the control of rice plant pests.

Differential colour-ring loss among Dalmatian Pelican(Pelecanus crispus) colonies and its consequence on survival estimates

The analysis of bird ringing data often comes with some potential sources of error and bias,as ring wear and/or loss could affect mark-recapture analyses and produce erroneous estimates of survival.Furthermore,ring wear and loss rates may differ between and within species based on the habitat they use or the species’ life-history traits and behaviour as well as the type of the ring.In this study we use resighting data from a long-term double marking experiment to directly estimate the rate of colour-ring loss among different Dalmatian Pelican colonies over time,evaluate any possible factors that could contribute to differential ring loss and assess how it may bias the results of mark-resighting analyses.Based on 14,849 resightings from 1275 individuals and using multi-state continuous-time hidden Markov models(HMMs) we showed that probability of ring loss was markedly different among colonies,ranging from 0.10 to 0.42 within the first year of marking,whereas the cumulative probability of losing a ring after ten years ranged 0.64 to 0.99.These rates are among the highest estimated when compared to previous studies in waterbirds.Our approach assessing the intra-specific variance in ring loss provided several factors potentially involved,such as the use of glue and the fledgling age accuracy and we could further hypothesise the effect of environmental factors.Finally,our results showed that ring loss can be a significant challenge for the assessment of the species’ population dynamics using mark-recapture methods as survival was consistently underestimated when not accounting for ring loss and varied significantly among different colonies.
