CO liberated from CORM-2 modulates the inflammatory response in the liver of thermally injured mice

AIM:To explore the effects of CO-releasing molecules tricarbonyldichlororuthenium(Ⅱ)dimer,CORM-2-liberated CO on attenuation of inflammatory responses in liver of an experimental animal model of thermal injury and to investigate the associated potential mechanisms.METHODS:Thirty-six mice were assigned to three groups in three respective experiments.In each experiment,mice in sham group(n=4)received sham thermal injury,whereas mice in burn group(n=4) received a 15%of total body surface area(TBSA)full-thickness thermal injury,and mice in burn+CORM-2 group(n=4)received the same thermal injury with immediate administration of CORM-2(8 mg/kg,iv).Hepatic tissue sections were stained with hematoxylin and eosin and examined under a light icroscope.Levels of aminotransferases(ALT and AST)and nitric oxide(NO) were measured by biochemical methods.Tumor necrosis factor-α(TNF-α)and interleukin(IL-1β)activity,and the protein expression of iNOS and HO-1 in serum and tissue homogenates were assessed.In in vitro experiments,Kupffer cells were stimulated with LPS(10μg/mL)for 4 h in the presence or absence of CORM-2(10-100μmol/L).Subsequently,the expression levels of TNF-αand NO production were assessed.RESULTS:Pro-inflammatory mediators(TNF-α,IL-1β,NO)in serum and liver homogenates of thermally injured mice were significantly reduced by CORM-2 administration.This was accompanied by a decrease in the expression of iNOS while an increase in the expression of HO-1 in the liver tissue.In parallel,the concentrations of TNF-αand NO in supernatants of LPS-stimulated Kupffer cells co-incubated with CORM-2 (10-100μmol/L)were also markedly decreased.Histological examination demonstrated that CORM-2 could attenuate the leukocytes infiltration to the liver tissue. CONCLUSION:CORM-released CO modulates liver inflammation and significantly protects liver injury in burn mice by inhibiting the expression of iNOS and NO production,down-regulating the expression of pro-inflammatory mediators(TNF-α,IL-1β).

Effects of different nerve autografts on greater auricular nerve deficit in rabbits

BACKGROUND: Autograft is commonly used to repair nerve deficit. Usually, the choice of donor nerves is based on their similarities in form and structures to the injured nerves. For the reason, the cutaneous antebrachii lateralis nerve is currently considered the most suited for digital nerve repair. OBJECTIVE: To compare early nerve regeneration after transplantation of three different autografts: the greater auricular nerve (GAN), the saphenous nerve (SN) and the lateral femoral cutaneous nerve (LFCN). DESIGN: Observational contrast study. SETTING: department of plastic surgery and Burns, Tangdu Hospital, Fourth Military Medical University of Chinese PLA. MATERIALS: A total of 42 New Zealand rabbits, of both genders, 12–14 months old and weighing 2.0– 2.5 kg, were used in this study. In addition, Moller-spetra 900 operating microscope (Germany), Olympus BX 51 microscope, DP 70 image collecting System (Japan), BL-420E+ Biologic function testing System (China), JEM-100 electron microscope (Japan), Reichet-JunG820 Cryostat (Swiss), and Libror-AEG-120 precision analytical Balance (Japan) were also used in this study. METHODS: The experiment was carried out in the department of plastic surgery and Burns, Tangdu Hospital, Fourth Military Medical University of Chinese PLA from April to November 2005. After anaesthesia, the GAN were dissected bilaterally and a 1.2 cm deficit was made in each nerve. The animals were randomly divided into three groups, including GAN group, SN group and LFCN group with 14 in each group. ① Nerve pinch test: At 1, 2, and 4 weeks after operation, three animals in each group were tested. The nerve grafts, along with the proximal and distal GAN segments were exposed and pinched with microsurgical forceps in distal-proximal orientations. Applications of human amniotic fluid stem cells in wound healing

Complete wound regeneration preserves skin structure and physiological functions,including sensation and perception of stimuli,whereas incomplete wound regeneration results in fibrosis and scarring.Amniotic fluid stem cells(AFSCs)would be a kind of cell population with self-renewing and non-immunogenic ability that have a considerable role in wound generation.They are easy to harvest,culture,and store;moreover,they are non-tumorigenic and not subject to ethical restrictions.They can differentiate into different kinds of cells that replenish the skin,subcutaneous tissues,and accessory organs.Additionally,AFSCs independently produce paracrine effectors and secrete them in exosomes,thereby modulating local immune cell activity.They demonstrate anti-inflammatory and immunomodulatory properties,regulate the physicochemical microenvironment of the wound,and promote full wound regeneration.Thus,AFSCs are potential resources in stem cell therapy,especially in scar-free wound healing.This review describes the biological characteristics and clinical applications of AFSCs in treating wounds and provide new ideas for the treatment of wound healing.

Role of adipose tissue grafting and adipose-derived stem cells in peripheral nerve surgery

The application of autologous fat grafting in reconstructive surgery is commonly used to improve functional form.This review aims to provide an overview of the scientific evidence on the biology of adipose tissue,the role of adipose-derived stem cells,and the indications of adipose tissue grafting in peripheral nerve surgery.Adipose tissue is easily accessible through the lower abdomen and inner thighs.Non-vascularized adipose tissue grafting does not support oxidative and ischemic stress,resulting in variable survival of adipocytes within the first 24 hours.Enrichment of adipose tissue with a stromal vascular fraction is purported to increase the number of adipose-derived stem cells and is postulated to augment the long-term stability of adipose tissue grafts.Basic science nerve research suggests an increase in nerve regeneration and nerve revascularization,and a decrease in nerve fibrosis after the addition of adipose-derived stem cells or adipose tissue.In clinical studies,the use of autologous lipofilling is mostly applied to secondary carpal tunnel release revisions with promising results.Since the use of adipose-derived stem cells in peripheral nerve reconstruction is relatively new,more studies are needed to explore safety and long-term effects on peripheral nerve regeneration.The Food and Drug Administration stipulates that adipose-derived stem cell transplantation should be minimally manipulated,enzyme-free,and used in the same surgical procedure,e.***.adipose tissue grafts that contain native adipose-derived stem cells or stromal vascular fraction.Future research may be shifted towards the use of tissue-engineered adipose tissue to create a supportive microenvironment for autologous graft survival.Shelf-ready alternatives could be enhanced with adipose-derived stem cells or growth factors and eliminate the need for adipose tissue harvest.

Non-microsurgical skin flaps for reconstruction of difficult wounds in distal leg and foot

Purpose: To express the versatility of a variety of non-microsurgical skin flaps used for coverage of difficult wounds in the lower third of the leg and the foot over 4 years period. Five kinds of flaps were used. Each flap was presented with detailed information regarding indication, blood supply, skin territory and technique. Methods: Altogether 26 patients underwent lower leg reconstruction were included in this study. The reconstructive procedures applied five flaps, respectively distally based posterior tibial artery perforator flap ( n = 8), distally based peroneal artery perforator flap ( n = 4), distally based sural flap ( n = 6), medial planter artery flap ( n = 2) and cross leg flaps ( n = 6). Results: In all cases, there were no signs of osteomyelitis of underlying bones or discharge from the undersurface of the flaps. Fat necrosis occurred at the distal end of posterior tibial artery perforator flap in one female patient. The two cases of medial planter artery flap showed excellent healing with closure of donor site primarily. One cross leg flap had distal necrosis. Conclusion: Would at lower third of leg can be efficiently covered by posterior tibial, peroneal artery and sural flaps. Heel can be best covered by nearby tissues such as medial planter flap. In presence of vascular compromise of the affected limb or exposure of dorsum of foot, cross leg flap can be used.

Optimal surgical approach for the treatment of Quervains disease: A surgical-anatomical study

AIM To determine which of the common used incision techniques has the lowest chance of iatrogenic damage to the nerves which at risk are the superficial branch of the radial nerve(SBRN) and the Lateral Antebrachial Cutaneous Nerve(LABCN).METHODS Twenty embalmed arms were dissected and the course of the SBRN and the LABCN in each individual arm was marked and the distance between the two branches of the SBRN at the location of the First Extensor Compartment(FEC) was measured. This data was used as input in a visualization tool called Computer Assisted Anatomy Mapping(CASAM) to map the course of the nerves in each individual arm. RESULTS This image visualizes that in 90% of the arms, one branch of the SBRN crosses the FEC and one branch runs volar to the compartment. The distance between the two branches was 7.8 mm at the beginning of the FEC and 10.2 mm at the end. Finally the angle of incision at which the chance of damage to the nerves is lowest, is 19.4 degrees volar to the radius.CONCLUSION CASAM shows the complexity of the course of the SBRN over the FEC. None of the four widely used incision techniques has a significantly lower chance of iatrogenic nerve damage. Surgical skills are paramount to prevent iatrogenic nerve damage.

Electric field-driven building blocks for introducing multiple gradients to hydrogels

Gradient biomaterials are considered as preferable matrices for tissue engineering due to better simulation of native tissues.The introduction of gradient cues usually needs special equipment and complex process but is only effective to limited biomaterials.Incorporation of multiple gradients in the hydrogels remains challenges.Here,betasheet rich silk nanofibers(BSNF)were used as building blocks to introduce multiple gradients into different hydrogel systems through the joint action of crosslinking and electric field.The blocks migrated to the anode along the electric field and gradually stagnated due to the solution-hydrogel transition of the systems,finally achieving gradient distribution of the blocks in the formed hydrogels.The gradient distribution of the blocks could be tuned easily through changing different factors such as solution viscosity,which resulted in highly tunable gradient of mechanical cues.The blocks were also aligned under the electric field,endowing orientation gradient simultaneously.Different cargos could be loaded on the blocks and form gradient cues through the same crosslinking-electric field strategy.The building blocks could be introduced to various hydrogels such as Gelatin and NIPAM,indicating the universality.Complex niches with multiple gradient cues could be achieved through the strategy.Silk-based hydrogels with suitable mechanical gradients were fabricated to control the osteogenesis and chondrogenesis.Chondrogenic-osteogenic gradient transition was obtained,which stimulated the ectopic osteochondral tissue regeneration in vivo.The versatility and highly controllability of the strategy as well as multifunction of the building blocks reveal the applicability in complex tissue engineering and various interfacial tissues.

Pathogenic roles of long noncoding RNAs in melanoma: Implications in diagnosis and therapies

Melanoma is one of the most dangerous types of cutaneous neoplasms,which are pigment-producing cells of neuroectodermal origin found all over the body.A great deal of research is focused on the mechanisms of melanoma to promote better diagnostic and treatment options for melanoma in its advanced stages.The progression of melanoma involves alteration in different levels of gene expression.With the successful implementation of next-generation sequencing technology,an increasing number of long noncoding RNAs(lncRNAs)sequences have been discovered,and a significant number of them have phenotypic effects in both in vitro and in vivo studies,implying that they play an important role in the occurrence and progression of human cancers,particularly melanoma.A number of evidence indicated that lncRNAs are important regulators in tumor cell proliferation,invasion,apoptosis,immune escape,energy metabolism,drug resistance,epigenetic regulation.To better understand the role of lncRNAs in melanoma tumorigenesis,we categorize melanomaassociated lncRNAs according to their cellular functions and associations with gene expression and signaling pathways in this review.Based on the mechanisms of lncRNA,we discuss the possibility of lncRNA-target treatments,and the application of liquid biopsies to detect lncRNAs in melanomadiagnosisandprognosis.

Exosomes derived from Nr-CWS pretreated MSCs facilitate diabetic wound healing by promoting angiogenesis via the circIARS1/miR-4782-5p/VEGFA axis

Mesenchymal stem cell(MSC)-derived exosomes(Exos)were reported to a prospective candidate in accelerating diabetic wound healing due to their pro-angiogenic effect.MSCs pretreated with chemistry or biology factors were reported to advance the biological activities of MSC-derived exosomes.Hence,this study was designed to explore whether exosomes derived from human umbilical cord MSCs(hucMSCs)preconditioned with Nocardia rubra cell wall skeleton(Nr-CWS)exhibited superior proangiogenic effect on diabetic wound repair and its underlying molecular mechanisms.The results showed that Nr-CWS-Exos facilitated the proliferation,migration and tube formation of endothelial cells in vitro.In vivo,Nr-CWS-Exos exerted great effect on advancing wound healing by facilitating the angiogenesis of wound tissues compared with Exos.Furthermore,the expression of circIARS1 increased after HUVECs were treated with Nr-CWS-Exos.CircIARS1 promoted the pro-angiogenic effects of Nr-CWS-Exos on endothelial cells via the miR-4782-5p/VEGFA axis.Taken together,those data reveal that exosomes derived from Nr-CWS-pretreated MSCs might serve as an underlying strategy for diabetic wound treatment through advancing the biological function of endothelial cells via the circIARS1/miR-4782-5p/VEGFA axis.
