Polydopamine-coated photoautotrophic bacteria for improving extracellular electron transfer in living photovoltaics

Living photovoltaics are microbial electrochemical devices that use whole cell–electrode interactions to convert solar energy to electricity.The bottleneck in these technologies is the limited electron transfer between the microbe and the electrode surface.This study focuses on enhancing this transfer by engineering a polydopamine(PDA)coating on the outer membrane of the photosynthetic microbe Synechocystis sp.PCC6803.This coating provides a conductive nanoparticle shell to increase electrode adhesion and improve microbial charge extraction.A combination of scanning electron microscopy(SEM),transmission electron microscopy(TEM),UV–Vis absorption,and Raman spectroscopy measurements were used to characterize the nanoparticle shell under various synthesis conditions.The cell viability and activity were further assessed through oxygen evolution,growth curve,and confocal fluorescence microscopy measurements.The results show sustained cell growth and detectable PDA surface coverage under slightly alkaline conditions(pH 7.5)and at low initial dopamine(DA)concentrations(1 mM).The exoelectrogenicity of the cells prepared under these conditions was also characterized through cyclic voltammetry(CV)and chronoamperometry(CA).The measurements show a three-fold enhancement in the photocurrent at an applied bias of 0.3 V(vs.Ag/AgCl[3 M KCl])compared to non-coated cells.This study thus lays the framework for engineering the next generation of living photovoltaics with improved performances using biosynthetic electrodes.

Transcriptional regulation of adult neural stem/progenitor cells: tales from the subventricular zone

In rodents,well characterized neurogenic niches of the adult brain,such as the subventricular zone of the lateral ventricles and the subgranular zone of the hippocampus,support the maintenance of neural/stem progenitor cells(NSPCs)and the production of new neurons throughout the lifespan.The adult neurogenic process is dependent on the intrinsic gene expression signatures of NSPCs that make them competent for self-renewal and neuronal differentiation.At the same time,it is receptive to regulation by various extracellular signals that allow the modulation of neuronal production and integration into brain circuitries by various physiological stimuli.A drawback of this plasticity is the sensitivity of adult neurogenesis to alterations of the niche environment that can occur due to aging,injury or disease.At the core of the molecular mechanisms regulating neurogenesis,several transcription factors have been identified that maintain NSPC identity and mediate NSPC response to extrinsic cues.Here,we focus on REST,Egr1 and Dbx2 and their roles in adult neurogenesis,especially in the subventricular zone.We review recent work from our and other laboratories implicating these transcription factors in the control of NSPC proliferation and differentiation and in the response of NSPCs to extrinsic influences from the niche.We also discuss how their altered regulation may affect the neurogenic process in the aged and in the diseased brain.Finally,we highlight key open questions that need to be addressed to foster our understanding of the transcriptional mechanisms controlling adult neurogenesis.

Elucidating the pathological mechanisms of neurodegeneration in the lethal serpinopathy FENIB

The term serpinopathies was introduced to describe a family of diseases caused by point mutations in serine protease inhibitors,or serpins.Serpins inhibit their cognate protease by an irreversible suicide mechanism starting with the attack of the active site serine on the reactive center loop of the inhibitor,followed by formation of a covalent complex between both proteins,insertion of the reactive center loop of the serpin into its own beta-sheet A,and culminating in distortion of the active site of the serine protease and thus irreversible inactivation.This inhibitory mechanism,reminiscent of the movement of a mousetrap,requires a structural flexibility that proves to be unfavorable when the folding of the serpin is altered by mutations responsible for conformational rearrangements,allowing an intermolecular domain exchange characterized by the insertion of the C-terminal domain of a molecule into a second one,thus forming a dimer.Expansion of this insertion reaction leads to the formation of serpin polymers that accumulate within the endoplasmic reticulum(ER)of the cell and consequently reduces the secretion of the wild-type serpin(Greene et al.,2016).

The Ever Expanding Role of Ubiquitin and SUMO in Plant biology

When it was discovered in the 1970s as a ubiquitous protein, ubiquitin, a 76-aminoacid peptide, had no assigned function. It was not until later that ubiquitin was found to be a necessary cofactor in a vital cellular process： the degradation of proteins. Work by Avram Hershko, Aaron Ciechanover, and Irwin Rose （for which they received the 2004 Nobel Prize in Chemistry） showed that the covalent attachment of ubiquitin provide pro-teins with a tag that dispatches them to the proteasome, the main cell degradation machinery （Ciechanover 2012）. Subsequent work, has revealed that ubiquitin belongs to a broader family of ubiquitin-like proteins that can modify not only protein stability, but also subcellular localization and function.

BACKGROUND Cardiovascular diseases particularly myocardial infarction(MI)are the leading cause of mortality and morbidity around the globe.As cardiac tissue possesses very limited regeneration potential,therefore use of a potent small molecule,inhibitor Wnt production-4(IWP-4)for stem cell differentiation into cardiomyocytes could be a promising approach for cardiac regeneration.Wnt pathway inhibitors may help stem cells in their fate determination towards cardiomyogenic lineage and provide better homing and survival of cells in vivo.Mesenchymal stem cells(MSCs)derived from the human umbilical cord have the potential to regenerate cardiac tissue,as they are easy to isolate and possess multilineage differentiation capability.IWP-4 may promote the differentiation of MSCs into the cardiac lineage.AIM To evaluate the cardiac differentiation ability of IWP-4 and its subsequent in vivo effects.METHODS Umbilical cord tissue of human origin was utilized to isolate the MSCs which were characterized by their morphology,immunophenotyping of surface markers specific to MSCs,as well as by tri-lineage differentiation capability.Cytotoxicity analysis was performed to identify the optimal concentration of IWP-4.MSCs were treated with 5μM IWP-4 at two different time intervals.Differentiation of MSCs into cardiomyocytes was evaluated at DNA and protein levels.The MI ratmodel was developed.IWP-4 treated as well as untreated MSCs were implanted in the MI model,then the cardiac function was analyzed via echocardiography.MSCs were labeled with 1,1′-dioctadecyl-3,3,3′,3′-tetramethylindocarbocyanine perchlorate(DiI)dye for tracking,while the regeneration of infarcted myocardium was examined by histology and immunohistochemistry.RESULTS MSCs were isolated and characterized.Cytotoxicity analysis showed that IWP-4 was non-cytotoxic at 5μM concentration.Cardiac specific gene and protein expression analyses exhibited more remarkable results in fourteen days treated group that was eventually selected for in vivo transplantation.Cardiac functi

Plants show an unparalleled regenerative capacity,allowing them to survive severe stress conditions,such as injury,herbivory attack,and harsh weather conditions.This potential not only replenishes tissues and restores damaged organs but can also give rise to whole plant bodies.Despite the intertwined nature of development and regeneration,common upstream cues and signaling mechanisms are largely unknown.Here,we demonstrate that in addition to being activators of regeneration,ETHYLENE RESPONSE FACTOR 114(ERF114)and ERF115 govern developmental growth in the absence of wounding or injury.Increased ERF114 and ERF115 activity enhances auxin sensitivity,which is correlated with enhanced xylem maturation and lateral root formation,whereas their knockout results in a decrease in lateral roots.Moreover,we provide evidence that mechanical cues contribute to ERF114 and ERF115 expression in correlation with BZR1-mediated brassinosteroid signaling under both regenerative and developmental conditions.Antagonistically,cell wall integrity surveillance via mechanosensory FERONIA signaling suppresses their expression under both conditions.Taken together,our data suggest a molecular framework in which cell wall signals and mechanical strains regulate organ development and regenerative responses via ERF114-and ERF115-mediated auxin signaling.

Integrative taxonomy,a multi-disciplinary approach adding modern techniques to traditional morphology-based methods(e.***.molecular and morphological criteria),can play an important role in bioinvasion research to identify introduced taxa,discover pathways of introduction and inform authorities to control and prevent future introductions.The present study is the first on introduced populations of Callosciurus,Asiatic tree squirrels,known as potentially invasive species in Europe(Italy,Belgium and France).We combined molecular(mitochondrial DNA markers:CoxI,D-loop)and morphometric analysis on skulls,comparing them to the widest morphological and molecular datasets ever assembled for Callosciurus.Squirrels collected in Italy and Belgium share the same haplotypes and skull characteristics,but are conspicuously different from the French population in Antibes.Genetic data revealed close similarity between French squirrels and Pallas’s squirrels,Callosciurus erythraeus,from Taiwan,China.Italian and Belgian squirrels formed an independent taxonomic lineage in genetic analyses,whose taxonomic rank needs further investigation.The morphological and morphometric characteristics of these 2 populations are,however,similar to known specimens assigned to Callosciurus erythraeus.These results may indicate a common origin for the populations found in Belgium and Italy.In contrast,French specimens suggest an independent introduction event of squirrels originating from Asia.

Least speciose among the most speciose:Natural history correlates of monospecific and bispecific genera of Rodentia and Soricomorpha

Monospecific and bispecific genera are of special concern as they represent unique phylogenetic/evolutionary trajectories within larger clades.In addition,as phylogenetically older taxa are supposed to be exposed to higher rarity and extinction risk,monospecific and bispecific genera may be intrinsically more prone to extinction risks than multispecies genera,although extinction risks also depend on the ecological and biological strategy of the species.Here,the distribution across biogeographical zones and the levels of threat to 2 speciose orders of mammals(monospecific and bispecific genera of Rodentia and Soricomorpha)are investigated in order to highlight major patterns at the worldwide scale.In Rodentia,39.7%of the genera(n=490)were monospecific and 17.9%were bispecific.In Soricomorpha,44.4%of the total genera(n=45)were monospecific and 15%were bispecific.There was a positive correlation between the number of monospecific genera and the total number of genera per family.Peaks of monospecific and bispecific genera richness were observed in Neotropical,Oriental and Afrotropical regions in rodents and in the Palearctic region in soricomorphs.Range size was significantly uneven across biogeographic region in rodents(with larger ranges in Nearctic and Oriental regions and smaller ranges in the Australian region),but there was no difference across biogeographic regions in terms of range size in soricomorphs.Most of the monospecific and bispecific genera occurred in forest habitat in both taxa.The frequency distribution of the monospecific and bispecific genera across IUCN categories did not differ significantly from the expected pattern using the total rodent genera and the multispecies genera.

On the Observable Transition to Living Matter

In recent developments in chemistry and genetic engineering, the humble researcher dealing with the origin of life finds her（him）self in a grey area of tackling something that even does not yet have a clear definition agreed upon. A series of chemical steps is described to be considered as the life-nonlife transition, if one adheres to the minimalistic definition： life is self-reproduction with variations. The fully artificial RNA system chosen for the exploration corresponds sequence-wise to the reconstructed initial triplet repeats, presumably corresponding to the earliest protein-coding molecules. The demonstrated occurrence of the mismatches （variations） in otherwise complementary syntheses （＂self-reproduction＂）, in this RNA system, opens an experimental and conceptual perspective to explore the origin of life （and its definition）, on the apparent edge of the origin.
