Anti free radical &anti inflammatory effect of rebamipide in chronic gastritis

Background/Aim: Free radicals have a role in the development of chronic gastritis. The aim of this study to know the effect and efficacy of rebamipide on free radicals in chronic gastritis. Method: Forty five patients in the division gastroenterology Cipto Mangunkusumo Hospital Jakarta 2009-2010 with moderate and severe gastritis endoscopically were included in this study. Before and after rebamipide treatment the patient were performed endoscopical examination and were taken 5 biopsies for histopathological examination and free radicals (MDA & Carbonyl Compound) examination. All patients were given rebamipide 100 mg three times a day for 28 days. Data were analyzed with t test or wilcoxon signed rank test. Exclusion: GERD, Peptic ulcer, PPI treatment, NSAID consumption etc. The symptoms were recorded on day-0 and day-28. The severity symptoms were measured by VAS. Result: The mucosal damage on day-0 was 2.268 ± 0.45 vs day-28 was 1.707 ± 0.78 (P < 0.001). The antrum neutrophil: day-0: 0.12 ± 0.46 vs day-28: 0.10 ± 0.37 (P = 0.710) and corpus neutrophil: day-0: 0.12 ± 0.40 vs day-28: 0.07 ± 0.26 (P = 0.421). The mean endoscopical mucosal severity score was decreased significantly on day- 28 compared to day-0 (1.707 ± 0.78 vs 2.268 ± 0.45;P < 0.05). The other histopathological appearances between day-0 and day-28 were not different. Rebamipide can reduce the mean of MDA from 5.28 ± 3.54 on day-0 to 4.15 ± 2.71 on day-28 (P = 0.047). The mean of carbonyl compound on day-0 was 4.14 ± 3.01 and on day-28 was 5.12 ± 2.71 (P = 0.642). Conclusion: Rebamipide significantly reduced the extend of symptoms associated with chronic gastritis. The improvement in symptoms was associated with the decreased of endoscopic severity score and the mean gastric mucosal malondialdehyde (MDA) significantly but not the histopathologic appearance and carbonyl compound.

Objective:To investigate whether ethanol extracts of Chondracanthus tenellus(EECT)could improve immunomodulatory property of murine monocyte/macrophage RAW 264.7 ***:Cell viability,phagocytic ability,and nitric oxide were *** levels of prostaglandin E2 and cytokines were determined using enzyme-linked immunosorbent assay *** of immunoregulatory response protein was detected by Western blotting ***:As the concentration of EECT increased,the morphology of the cells changed to a typical active macrophage shape,and the phagocytic activity increased *** also effectively enhanced the production and secretion of immunomodulatory mediators,such as nitric oxide and prostaglandin E2,and *** addition,compared with the control group,EECT markedly stimulated the expression of Toll-like receptor 4(TLR4)and myeloid differentiation factor 88,one of the TLR4 adapter ***,EECT promoted the nucleus translocation of nuclear factor-kappa B(NF-κB)by increasing the phosphorylation and degradation of the inhibitor of NF-κB-α,indicating activation of the NF-κB signaling ***,similar trends were found in cells treated with lipopolysaccharide as a positive ***:Taken together,the results indicate that EECT has an immunomodulatory effect by increasing the production of immunomodulatory mediators and cytokines through activation of the TLR4/NF-κB signaling *** could be used as a potential candidate for medication or dietary supplements to increase immune activity.

Extrahepatic right portal vein ligation allows parenchyma-sparing en bloc resection of segments 7, 8 and 4a for liver tumors engaging the right and middle hepatic veins

Right trisectionectomy for posterior liver tumors engaging the right and middle hepatic veins may lead to post-hepatectomy liver failure if the anticipated liver remnant is small. In such patients we developed a parenchymasparing one-step approach, that includes extrahepatic right portal vein ligation accompanied by en bloc resection only of segments 7, 8 and 4a and resection of the right and middle hepatic veins. The technique was applied in 3 patients with normal liver function, where according to the preoperative computed tomography the volume of segments 1, 2 and 3 ranged between 17% and 20% of the total liver volume. In all patients liver biochemistry improved rapidly postoperatively and a doubling of volume of segments 1, 2 and 3 was achieved by the third postoperative week, as extrahepatic right portal vein ligation ameliorated reperfusion injury of the remaining segments 5 and 6 and induced hypertrophy of segments 1, 2, 3 and 4b. There was no mortality or long-term *** are alive and free of disease 74, 50 and 17 months after the operation, respectively. We propose that the term ＂extended upper right sectionectomy＂ may be considered for the en bloc resection of segments 7, 8 and 4a, in future revisions ofthe Brisbane 2000 terminology of hepatic anatomy and resections.

BACKGROUND The respiratory infection caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has evolved into a multi-organ disorder,with longterm effects known as post-acute sequelae of SARS-CoV-2 infection or long coronavirus disease(COVID).AIM To examine the current knowledge and outcomes of long-term neurological and gastrointestinal(GI)symptoms in adult cohorts,including United States minority *** PubMed and Google Scholar were searched using relevant terms,and data from five studies were analyzed,comprising 27383 patients with persistent neurological and GI *** The main symptoms included anxiety,depression,dysphagia,headache,vomiting,nausea,gastroesophageal reflux,fatigue,and abdominal *** with comorbidities and metabolic syndromes were at higher risk for long *** most patients were European Americans,there was a need for further study on African *** The underlying causes of these symptoms remain unclear,warranting more investigation into the long-term impact of the SARS-CoV-2 on different populations.

Lysosome and proteasome dysfunction in alcohol-induced liver injury

Objective Neurodegenerative diseases, such as ischemia, traumatic injury, Alzheimer＇s disease, and Parkinson＇s disease are characterized by neuronal loss and dysfunction. It is known that glutamate-induced toxicity plays an important role in neurodegenerative diseases. Glutamate toxicity seems to be mediated by excessive influx of Ca^（2＋） into neuronal cells through activation of N-methyl-D-aspartate（NMDA） receptor. To search for potential NMDA receptor inhibitors in traditional Chinese medicine. Methods A series of computer methods including drug-likeness evaluation, ADMET tests as well as molecular docking have been used. Results 1,5-O-dicaffeoyl-quinic acid was identified as NMDA receptor inhibitor by virtual screening. Its neuroprotective activity was further confirmed by in vitro test. 1,5-O-dicaffeoyl-quinic acid showed strong neuroprotection against NMDA-induced cell injury. Conclusion 1,5-O-Dicaffeoylquinic acid may be regarded as a potential NMDA receptor inhibitor for the prevention and treatment of neurodegenerative disorders.

This study sought to investigate the possible mechanisms for the use of phenolic extracts from grapefruit peels in the management/prevention of cardiovascular *** effects of the phenolic extracts on key enzymes relevant to cardiovascular diseases[3-hydroxy-methyl-3-glutaryl coenzyme A reductase(HMG-CoA reductase)and angiotensin-I converting enzyme(ACE)],cellular antioxidant activity in human endothelial cells(*** 926)and radicals[1,1-diphenyl-2 picrylhydrazyl(DPPH)and 2,2-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid)(ABTS)]scavenging abilities were *** phenolic contents of the extracts were investigated using HPLC–*** was no significant(P>0.05)difference in the HMG-CoA reductase inhibitory ability of the two extracts,while the bound phenolic extracts had a stronger ACE inhibitory ability than the soluble free *** extracts also showed intracellular antioxidant activity in human endothelial(*** 926)***,the bound phenolics had significantly higher radicals(DPPH*and ABTS*)scavenging abilities than the free *** HPLC analysis revealed the presence of flavonoids(quercetin and kaempferol),phenolics acids(resveratrol,gallic acid,ellagic acid and caffeic acid)and tannin(catechin).The cellular antioxidative properties and inhibition of enzymes relevant to the management of cardiovascular complications showed that grapefruit peels could be used as nutraceuticals for the management of such conditions.

Lead Induces Different Responses of Two Subpopulations of Phagocytes in the Holothurian Eupentacta fraudatrix

In view of increasing lead pollution(Pb^(2+)) of coastal waters, the compensatory abilities of holothurians need to be assessed. The goal of the work is to clarify the functional and phenotypical differences between two types of phagocytes(P1 and P2) in Eupentacta fraudatrix exposed to Pb(NO_3)_2. It has been shown that 2 mg L^(-1) lead exposure for 48 h increases the number of P2 phagocytes as compared to P1 cells, does not significantly affect cell viability in both P1 and P2 phagocyte fractions, and significantly enhances chromatin condensation in P2 but not in P1 phagocytes. A lead concentration of 4 mg L^(-1) increases the number of P1 phagocytes compared to that of P2 type, and does not change cell viability and chromatin condensation in P1 phagocytes. In the P2 type, it decreases cell viability and does not influence the level of apoptosis. The protection against lead-induced apoptosis is apparently mediated by the activities of antioxidant enzymes, especially glutathione S-transferase. The differences in labeling cell surface receptors of P1 and P2 phagocytes by plant lectins also indicate the specific phenotypic properties of these cells. The results clarify the potential and GSH-dependent mechanisms of immune adaptation in holothurians that have been shortly exposed to lead at concentrations close to the maximum environmentally relevant level in coastal waters. Additionally, P1 and P2 phagocytes are first shown to have different functions and phenotypes during the response to lead, which indicates the complexity of the phagocytic system in holothurians and contributes to understanding the immunity evolution.

Resveratrol（RSV） is a naturally-occurring polyphenol that exhibits pleiotropic health benefits,including anti-colitis and colon cancer-protective ***,we identified the novel imine RSV analog（IRA）,2-methoxyl-3,6-dihydroxyl-IRA 3,4,5,4-tetramethoxystilbene（C33）,as a putative activator of nuclear factor erythriod 2-related factor 2（Nrf2）.The present study was designed to evaluate the ability of C33 to activate the Nrf2 signaling pathway and its anti-colitis effect in comparison to *** anti-colitis action of C33 was assessed in a mouse model of colitis induced by dextran sulfate sodium（DSS）.The effect of C33 on the Nrf2 signaling pathway was examined in vitro and in *** to RSV,C33 triggered a more dramatic increase in the expression of genes downstream from Nrf2 in Hepalclc7 cells and wild-type（WT） mouse embryonic fibroblasts,as well as in the small intestine and colon of WT *** with its superior ability to activate the cytoprotective Nrf2 pathway,C33 was significantly better in ameliorating DSS-induced colitis by improving the inflammation score,as well as downregulating the markers of inflammation in WT ***,induction of the NOD-like receptor family pyrin 3（NLRP3） inflammasome by colitis was also significantly inhibited by the ***2-knockout completely abolished the effects of C33,indicating that Nrf2 is the important mechanistic target of C33 in *** conclusion,the novel IRA,C33,has stronger anti-colitis effects than *** studies are warranted to evaluate C33 as a potential therapeutic agent for inflammatory bowel disease and cancer chemoprevention.
