Sex Differences in Neuropathology and Cognitive Behavior in APP/PS1/tau Triple-Transgenic Mouse Model of Alzheimer's Disease

Alzheimer＇s disease （AD） is the most common form of dementia among the elderly, characterized by amyloid plaques, neurofibrillary tangles, and neuroinflam- mation in the brain, as well as impaired cognitive behaviors. A sex difference in the prevalence of AD has been noted, while sex differences in the cerebral pathology and relevant molecular mechanisms are not well clarified. In the present study, we systematically investigated the sex differences in pathological characteristics and cognitive behavior in 12-month-old male and female APP/PSI/tau triple-trans- genic AD mice （3xTg-AD mice） and examined the molecular mechanisms. We found that female 3×Tg-AD mice displayed more prominent amyloid plaques, neurofib- rillary tangles, neuroinflammation, and spatial cognitive deficits than male 3×Tg-AD mice. Furthermore, the expres- sion levels of hippocampal protein kinase A-cAMP response element-binding protein （PKA-CREB） and p38- mitogen-activated protein kinases （MAPK） also showed sex difference in the AD mice, with a significant increase in the levels of p-PKA/p-CREB and a decrease in the p-p38 in female, but not male, 3×Tg-AD mice. We suggest that an estrogen deficiency-induced PKA-CREB-MAPK signaling disorder in 12-month-old female 3×Tg-AD mice might be involved in the serious pathological and cognitive damage in these mice. Therefore, sex differences should be taken into account in investigating AD biomarkers and related target molecules, and estrogen supplementation or PKA-CREB- MAPK stabilization could be beneficial in relieving the pathological damage in AD and improving the cognitive behavior of reproductively-senescent females.

The stability,frontier orbitals,bond character,and static linear polarizability of the fluorinated fullerene H_(20)@C_(80)F_(60)

This paper uses the density functional theory under generalised gradient approximation to analyse the stability, frontier orbitals, bond character, and static linear polarizability of H20@C80F60, which has not been isolated, as well as those of the synthesised H20@C80H60. The H20@C80F60 should be considerably stable by analysing its energy and aromaticity. The inside H and outside X will play different role in the chemical reaction involving H20@C80X60 （X = H and F）. The covalence of C H bond is in the order that the inside C H bond of H20@C80F60〉 the inside C H bond of H20@C80H60〉 the outside C-H bond of H20@C80H60, whereas the C-F bond of H20@C80F60 have both the covalent and ionic characters. The static linear polarizabilities of Cso and H20@C80X60 （X =H and F） are all isotropic.

Protective effect of low dose of melatonin against cholestatic oxidative stress after common bile duct ligation in rats

AIM: To investigate the role of oxidative injury and the effect of exogenous melatonin administration on liver damage induced by bile duct ligation (BDL), and second, to evaluate the role of nitric oxide (NO), a free oxygen radical, in oxidative injury. METHODS: Thirty-two Sprague-Dawley rats were assigned to four groups: sham operation (SO), BDL, BDL+melatonin, and BDL+vehicle. Cholestasis was achieved by double ligature of the common bile duct. Melatonin was injected intraperitoneally 500 μg/(kg·d) for 8 d. Hepatic oxidative stress markers were evaluated by changes in the amount of lipid peroxides, measured as malondialdehyde (MDA), and reduced GSH. Total nitrite (NOx) concentrations were determined in hepatic homogenates. Histopathological examination was performed using a histological scoring system. RESULTS: The histopathological changes including portal inflammation, necrosis,apoptosis, focal inflammation and fibrosis were severe in the BDL and BDL+vehicle groups. There were numerous large areas of coagulation necrosis. Histological Activity Index scores of these groups were significantly higher than that of the SO group. Treatment with melatonin reduced these alterations significantly. The degree of necro-inflammation and fibrosis showed significant difference between the BDL and BDL+melatonin groups. BDL was accompanied by a significant increase in MDA and NOx, and a significant decrease in GSH levels. Mean±SE values of MDA, GSH and NOx levels of SO group were 147.47±6.69, 0.88±0.33 μmol/g and 180.70±6.58 nm/g, respectively. The values of BDL group were 200.14±21.30, 0.65±0.02 μmol/g, and 400.46±48.89 nm/g, respectively, whereas the values of BDL+melatonin group were 115.93±6.8,0.74±0.02 μmol/g, and 290.38±32.32 nm/g, respectively. Melatonin treatment was associated with a significant recovery of MDA, GSH and NOx levels. Local magneto-optical Kerr effect imaging by scanning near-field optical microscope in reflection-mode

A method for local magneto-optical Kerr effect imaging based on a home-made scanning near-field optical microscope working in reflection mode is presented. Shear force detection is carried out by using a symmetric piezoelectric bimorph sensor, which provides an easy way not only for probe-surface distance control but also for imaging. Polarization-preserving fiber probes used as a local optical detector are fabricated with a heating-pulling technique and the probes＇ polarization properties are measured. Shear force topographic and near-field magneto-optical images of magueto-optical disk taken with the proposed method are shown.

Dysregulation of synaptic and extrasynaptic N-methyl-D-aspartate receptors induced by amyloid-β

The toxicity of amyloid-beta（Aβ） is strongly associated with Alzheimer’s disease（AD）,which has a high incidence in the elderly *** evidence showed that alteration in the activity of N-methyl-D-aspartate receptors（NMDARs） plays a key role in Aβ-induced ***,the activation of synaptic and extrasynaptic NMDARs has distinct consequences for plasticity,gene regulation,neuronal death,and Aβ *** review focuses on the dysregulation of synaptic and extrasynaptic NMDARs induced by Aβ.On one hand,Aβ downregulates the synaptic NMDAR response by promoting NMDAR endocytosis,leading to either neurotoxicity or *** the other hand,Aβ enhances the activation of extrasynaptic NMDARs by decreasing neuronal glutamate uptake and inducing glutamate spillover,subsequently causing *** addition,selective enhancement of synaptic activity by low doses of NMDA,or reduction of extrasynaptic activity by memantine,a non-competitive NMDAR antagonist,halts Aβ-induced ***,future neuroprotective drugs for AD should aim at both the enhancement of synaptic activity and the disruption of extrasynaptic NMDAR-dependent death signaling.

DNA-assisted rational design of BaF_2 linear and erythrocyteshaped nanocrystals

The synthesis of inorganic materials with special morphologies with the assistance of biological molecules is a potential development in the field of controllable growth and assembly of nanomaterials. In this paper, BaF2 nanocrystals in patterns of well-defined linear and erythrocyte-shaped structure were synthesized with the assistance of Escherichia coil DNA. Morphology and the arrangement of BaF2 particles on DNA were controllable by altering the reaction condition. Square nanoparticles arranged in linear chains were gained with the assistance of normal DNA; while, erythrocyte-shaped BaF2 nanospheres were synthesized with the assistance of denatured DNA. Besides, the influences of solvent, reaction temperature, concentration of reactants and the heating time on the morphology of the BaF2 particles were studied.

Structural Analysis of Eu_2Mn_(2/3)Ta_ 4/3O_7 with a Pyrochlore-Related Structure

Eu 2Mn 2/3Ta 4/3O 7 specimen was synthesized from Eu 2O 3, MnO and Ta 2O 5 in a reducing atmosphere. Structure analysis was carried out by Rietveld method from X-ray diffraction data. The X-ray diffraction profile calculated with monoclinic C2/c model was in a good agreement with the observed X-ray diffraction patterns since several small peaks of super lattice could be also assigned with C2/c symmetry in addition to fundamental reflection peaks. Eu 2Mn 2/3Ta 4/3O 7 has two kinds of distorted (Mn, Ta)O 6 octahedra and HTB layers, which deviates from the regular forms. Europium atoms coordinate to eight or seven oxygen atoms and lead to two kinds of polyhedra, EuO 8 and EuO 7 in this compound.

Bipartite entanglement in spin-1/2 Heisenberg model

The bipartite entanglement of the two- and three-spin Heisenberg model was investigated by using the concept of negativity. It is found that for the ground-state entanglement of the two-spin model, the negativity always decreases as B increases if △ 〈γ- 1, and it may keep a steady value of 0.5 in the region of B 〈 J[（△＋ 1）2 -γ^2]^1/2 if △ 〉γ-1, while for that of the three-spin model, the negativity exhibits square wave structures if γ=0 or△=0. For thermal states, there are two areas showing entanglement, namely, the main region and the sub-region. The main region exists only when △ 〉 △c （△c =γ- 1 and （γ^2 - 1）/2 for the 2- and 3-spin model respectively） and extends in terms of B and T as A increases, while the sub-region survives only when γ≠0 and shrinks in terms of B and T as △ increases.
