目的:比较不同日龄蛋鸡血清雌激素的水平、肝脏雌激素受体基因和卵黄前体物生成相关基因表达量、"肝脏-血液-卵巢轴"中甘油三酯和胆固醇的含量和肝脏抗氧化能力的动态变化,揭示蛋鸡卵黄前体物生成随着年龄增长的变化规律。创新点:首次发现在蛋鸡衰老过程中雌激素作用衰退以及肝脏抗氧化能力降低致使肝脏中卵黄前体物合成下降,最终导致蛋鸡的产蛋性能下降。方法:取90、150、280和580日龄蛋鸡各10只,检测血清中雌激素水平、肝脏雌激素受体基因表达水平、肝脏中卵黄前体物质合成和脂质代谢相关基因表达水平、"肝脏-血液-卵巢轴"中甘油三酯和胆固醇的含量及肝脏抗氧化能力,揭示蛋鸡卵黄前体物生成在衰老过程中的变化规律。结论:在蛋鸡衰老过程中,血清雌激素水平、肝脏雌激素受体基因和卵黄前体物质合成相关基因表达量降低,肝脏抗氧化能力降低,引起肝脏合成甘油三酯的能力下降,最终导致卵黄前体物质合成能力降低。

Rabbit breeding has many critical aspects related to reproduction,production,and animal welfare,which reduce its profitability as well as consumer attractiveness.Dietary supplementation with n-3 polyunsaturated fatty acids(PUFA)seems to be a good nutritional strategy to improve several aspects of rabbit breeding,enhance animal welfare and produce a new functional food considered healthy for human consumption.For this reason,the main available scientific research regarding the physiological effects of n-3 PUFA rich products supplemented to the rabbit diet will be reviewed.In particular,consequences on the reproductive performances of both doe and buck,the productive parameters,and the meat quality will be analysed.

Mastitis often occurs during women’s lactation period， causing great psychological and physical pain to women. Fucoidan， a sulfated polysaccharide obtained from brown algae， has much broader biological properties. However， the roles of fucoidan in lipopolysaccharides (LPS)-induced mastitis are still undiscovered. The present study was aimed to evaluate the influences of fucoidan on LPS-induced mouse mastitis and investigate its possible mechanisms. The expression profiles of fucoidan acting on mastitis were analyzed by Network pharmacology. Additionally， mechanism experiments verified the mechanism of fucoidan on mastitis. The results of in vivo study displayed that the treatment of fucoidan to LPS-stimulated mouse mastitis decreased the inflammatory damage， proinflammatory cytokines level and repaired the completeness of blood-milk barrier. In the study of mouse mammary epithelial cells， fucoidan suppressed the secretion of reactive oxygen species (ROS)and enhanced the activity of antioxidant enzymes. Molecular experiments suggested that fucoidan promoted Nrf2 nuclear import and autophagy via activating the AMPK signaling pathway. The above results indicated that in mastitis， fucoidan promoted Nrf2 nuclear import and autophagy via activating the AMPK signaling pathway， thereby suppressing oxidative stress and inflammatory injury to the mammary glands and protecting the blood-milk barrier.

Background： Rumen microbiome has a great influence on ruminant health and productivity. Different plant extracts have been tested for their ability to modulate the rumen microbiome to improve feed digestion and fermentation. Among the evaluated plant extracts, essential oils, tannins, and saponins appeared to have positive effects on rumen protein metabolism, volatile fatty acids production, and methane and ammonia production. Methods： The objective of this study was to evaluate the effect of rosemary （Rosmarinus officinalis L.） leaves and essential oils on rumen microbial populations. Four ruminally cannulated sheep were used in a 4x4 Latin square design fed （21 d/period）： 1） a control diet composed of alfalfa hay and concentrate pellet （CTR）, 2） CTR supplemented with 7 g/d/sheep of rosemary essential oil adsorbed on an inert support （EO）, 3） CTR with 10 g/d/sheep of dried and ground rosemary leaves （RL）, and 4） CTR with 10 g/d of dried and ground rosemary leaves pelleted into concentrate （RL pellet）. Abundance of total bacteria, archaea, protozoa, and some select bacterial species or groups was quantified using qPCR, while the community of bacteria and archaea was profiled using denaturing gradient gel electrophoresis. Results： No difference in abundance was noted for total bacteria, protozoa, or Rurninococcus flavefaciens between the control and the treatments, but the rosemary leaves, either in loose form or in pellet, decreased the abundance of archaea and the genus Prevotello （P 〈 0.001）. The rosemary leaves in loose form also decreased （P 〈 0.001） the abundance of Ruminococcus albus and Clostridium aminophilum, while the EO increased （P 〈 0.001） the abundance of Fibrobocter sucdnogenes. The community of bacteria and archaea was not affected by any of the supplements. Conclusions： Being able to affect the abundance of several groups of rumen microbes that are known to be involved in degradation of protein and fiber and production of methane and ammonia, rosemary leaves may be used to modulate rumen microbio

Background Grape and winery by-products have nutritional values for cattle and also contain functional compounds like phenols,which not only bind to protein but can also directly affect microbiota and their function in the rumen.We characterized the nutritional and functional effects of grape seed meal and grape pomace as well as an effective dosage of grape phenols on ruminal microbiota and fermentation characteristics using a rumen simulation technique.Results Six diets(each n=8)were compared including a control diet(CON,no by-product),a positive control diet(EXT,CON+3.7%grape seed extract on a dry matter(DM)basis),two diets with grape seed meal at 5%(GS-low)and 10%(GS-high),and two diets with grape pomace:at 10%(GP-low)and 20%(GP-high),on a DM basis.The inclusion of the by-product supplied total phenols at 3.4%,0.7%,1.4%,1.3%,and 2.7%of diet DM for EXT,GS-low,GS-high,GP-low,and GP-high,respectively.Diets were tested in four experimental runs.All treatments decreased ammonia concentrations and the disappearances of DM and OM(P<0.05)compared to CON.EXT and GP-high lowered butyrate and odd-and branch-chain short-chain fatty acids while increased acetate compared to CON(P<0.05).Treatments did not affect methane formation.EXT decreased the abundance of many bacterial genera including those belonging to the core microbiota.GP-high and EXT consistently decreased Olsenella and Anaerotipes while increased Ruminobacter abundances.Conclusion The data suggest that the inclusion of winery by-products or grape seed extract could be an option for reducing excessive ammonia production.Exposure to grape phenols at a high dosage in an extract form can alter the rumen microbial community.This,however,does not necessarily alter the effect of grape phenols on the microbial community function compared to feeding high levels of winery by-products.This suggests the dominant role of dosage over the form or source of the grape phenols in affecting ruminal microbial activity.In conclusion,supplementing grape phenols at about 3%of diet D

Background:Bacillus cereus is an important pathogen that causes human food poisoning,specifically diarrhea and vomiting.B.cereus can also induce mastitis in dairy cows and has a strong survival ability in milk,as it cannot be inactivated by high-temperature short-time pasteurization.Therefore,B.cereus can enter the market through pasteurized milk and other dairy products,imposing enormous hidden dangers on food safety and human health.Results:In this study,B.cereus 2101(BC)was isolated from milk samples of cows with mastitis.BC grew rapidly with strong hemolysis,making it difficult to prevent mastitis and ensure food security.MAC-T cells were treated with BC and/or Lactobacillus rhamnosus GR-1(LGR-1).Pretreatment with LGR-1 protected the integrity of tight junctions and the expression of zonula occludens-1(ZO-1)and occludin destroyed by BC.Furthermore,LGR-1 pretreatment reduced the expression of NOD-like receptor family member pyrin domain-containing protein 3(NLRP3),caspase recruitment and activation domain(ASC),Caspase-1 p20,gasdermin D(GSDMD)p30,inflammatory factors(interleukin(IL)-1βand IL-18),and cell death induced by BC.Moreover,LGR-1 pretreatment reduced NLRP3 inflammasome activity and increased expressions of ZO-1 and occludin induced by lipopolysaccharides(LPS)+ATP stimulation.MAC-T cells were transfected with NLRP3 si RNA or MCC950 and/or treated with BC and/or LGR-1.NLRP3-si RNA transfection and MCC950 attenuated BC-induced NLRP3 inflammasome activity.Expression of inflammatory cytokines and cell death suggested that the inflammatory pathway might play an important role in the induction of the NLRP3 inflammasome by BC and the protection of LGR-1.Conclusions:These results suggest that LGR-1 might be a probiotic alternative to antibiotics and could be administered to prevent mastitis in dairy cows,thus ensuring food security.

AIM To determine how a normal human colon cell line reacts to microbial challenge as a way to study oxidative stress-induced responses associated with inflammatory bowel disease.METHODS Normal human colon epithelial cells(ATCC?CRL.1790?)were stimulated with either heat killed E.coli or heat killed murine cecal contents(HKC)and examined for several relevant biomarkers associated with inflammation and oxidative stress including cytokine production,mitochondrial autophagy and oxidant status.TNFα,IL-1βand IL-8 protein concentrations were measured within the supernatants.Fluorescent microscopy was performed to quantify the production of reactive oxygen species(ROS)using an oxidation responsive fluorogenic probe.Mitochondrial morphology and mitochondrial membrane potential was assessed by dual staining using COXIV antibody and a dye concentrating in active mitochondria.Mitochondrial ROS scavenger was used to determine the source of ROS in stimulated cells.Autophagy was detected by staining for the presence of autophagic vesicles.Positive controls for autophagy and ROS/RNS experiments were treated with rapamycin and chloroquine.Mitochondrial morphology,ROS production and autophagy microscopy experiments were analyzed using a custom acquisition and analysis microscopy software(Image J).RESULTS Exposing CRL.1790 cells to microbial challenge stimulated cells to produce several relevant biomarkers associated with inflammation and oxidative stress.Heat killed cecal contents treatment induced a 10-12fold increase in IL-8 production by CRL.1790 cells compared to unstimulated controls at 6 and 12 h(P<0.001).Heat killed E.coli stimulation resulted in a4-5 fold increase in IL-8 compared to the unstimulated control cells at each time point(P<0.001).Both heat killed E.coli and HKC stimulated robust ROS production at 6(P<0.001),and 12 h(P<0.01).Mitochondrial morphologic abnormalities were detected at 6 and12 h based on reduced mitochondrial circularity and decreased mitochondrial membrane potential,P<0.01.Microbial stimulation also

Understanding changes in pathogen behavior(e.***.increased virulence,a shift in transmission channel) is critical for the public health management of emerging infectious diseases.Genome degradation via gene depletion or inactivation is recognized as a pathoadaptive feature of the pathogen evolving with the host.However,little is known about the exact role of genome degradation in affecting pathogenic behavior,and the underlying molecular detail has yet to be examined.Using large-scale global avian-restricted Salmonella genomes spanning more than a century,we projected the genetic diversity of Salmonella Pullorum(bvSP)by showing increasingly antimicrobial-resistant ST92 prevalent in Chinese flocks.The phylogenomic analysis identified three lineages in bvSP,with an enhancement of virulence in the two recently emerged lineages(L2/L3),as evidenced in chicken and embryo infection assays.Notably,the ancestor L1 lineage resembles the Salmonella serovars with higher metabolic flexibilities and more robust environmental tolerance,indicating stepwise evolutionary trajectories towards avian-restricted lineages.Pan-genome analysis pinpointed fimbrial degradation from a virulent lineage.The later engineeredfim-deletion mutant,and all other five fimbrial systems,revealed behavior switching that restricted horizontal fecal-oral transmission but boosted virulence in chicks.By depleting fimbrial appendages,bvSP established persistent replication with less proinflammation in chick macrophages and adopted vertical transovarial transmission,accompanied by ever-increasing intensification in the poultry industry.Together,we uncovered a previously unseen paradigm for remodeling bacterial surface appendages that supplements virulence-enhanced evolution with increased vertical transmission.
