Effects of tannic acid on the immunity and intestinal health of broiler chickens with necrotic enteritis infection

Background In broiler chickens,necrotic enteritis(NE)infection can reduce production performance.Tannic acid has shown great potential as a treatment of NE in broilers.However,the appropriate dosage of tannic acid in NE of broilers and the improvement effect on intestinal health are not very clear.In this study,we aimed to investigate the effects of different doses of tannic acid on the production performance,immunity,and intestinal health of broilers by constructing an NE model with C.perfringens infection and determining the appropriate dosage of tannic acid with regard to NE.Results Challenged birds showed significant reduction in body weight,villus height,and the ratio of villus height to crypt depth(P<0.05)and increase in the feed consumption gain ratio,intestinal lesion score,and crypt depth(P<0.05).The infection significantly reduced the relative Bacteroidota and Ligilactobacillus abundance(P<0.05)and increased the ratio of Firmicutes/Bacteroidota and cecal content of C.perfringens(P<0.05).Challenged birds fed diets supplemented with tannic acid showed significantly increased mRNA expression of nutrient transport carriers and intestinal barrier genes and growth performance and reduced serum zonulin and endotoxin levels(P<0.05).Addi-tion of tannic acid to the diet inhibited the inflammatory response by reducing the number of coccidia oocysts in feces and the content of C.perfringens in the cecum.Specifically,tannic acid reduced the serum levels of C reactive protein,myeloperoxidase,and specific IgY and ileal mucosal secretory immunoglobulin A levels in the ileal mucosa compared with those in the NE-infected birds.NE-infected birds fed diets supplemented with tannic acid also showed significantly increased relative Anaerocolumna,Thermoanaerobacterium,and Thermosinus abundance(P<0.05);their microbial composition and functional predictions were similar to those of the NC group.Conclusions Tannic acid in the diet alleviated NE by enhancing the intestinal barrier and absorption function.The recommended dietary t

Embelin alleviates weaned piglets intestinal inflammation and barrier dysfunction via PCAF/NF-κB signaling pathway in intestinal epithelial cells

Background:Intestinal barrier plays key roles in maintaining intestinal homeostasis.Inflammation damage can severely destroy the intestinal integrity of mammals.This study was conducted to investigate the protective effects of embelin and its molecular mechanisms on intestinal inflammation in a porcine model.One hundred sixty 21-day-old castrated weaned pigs(Duroc×Landrace×Yorkshire,average initial body weight was 7.05 d to four groups and fed with a basal diet o±0.28 kg,equal numbers of castrated males and females)were allotter a basal diet containing 200,400,or 600 mg embelin/kg for 28 d.The growth performance,intestinal inflammatory cytokines,morphology of jejunum and ileum,tight junctions in the intestinal mucosa of piglets were tested.IPEC-1 cells with overexpression of P300/CBP associating factor(PCAF)were treated with embelin,the activity of PCAF and acetylation of nuclear factor-κB(NF-κB)were analyzed to determine the effect of embelin on PCAF/NF-κB pathway in vitro.Results:The results showed that embelin decreased(P<0.05)serum D-lactate and diamine oxidase(DAO)levels,and enhanced the expression of ZO-1,occludin and claudin-1 protein in jejunum and ileum.Moreover,the expression levels of critical inflammation molecules(interleukin-1β,interleukin-6,tumor necrosis factor-α,and NF-κB)were downregulated(P<0.05)by embelin in jejunal and ileal mucosa.Meanwhile,the activity of PCAF were down-regulated(P<0.05)by embelin.Importantly,transfection of PCAF siRNAs to IPEC-1 cell decreased NF-κB activities;embelin treatment downregulated(P<0.05)the acetylation and activities of NF-κB by 31.7%-74.6%in IPEC-1 cells with overexpression of PCAF.Conclusions:These results suggested that embelin ameliorates intestinal inflammation in weaned pigs,which might be mediated by suppressing the PCAF/NF-κB signaling pathway.

Melatonin promotes the development of the secondary hair follicles by regulating circMPP5

Background The quality and yield of cashmere fibre are closely related to the differentiation and development of secondary hair follicles in the skin of cashmere goats.The higher the density of secondary hair follicles,the higher the quality and yield of cashmere from the fleece.Development of secondary hair follicles commences in the embryonic stage of life and is completed 6 months after birth.Preliminary experimental results from our laboratory showed that melatonin(MT)treatment of goat kids after their birth could increase the density of secondary hair follicles and,thus,improve the subsequent yield and quality of cashmere.These changes in the secondary hair follicles resulted from increases in levels of antioxidant and expression of anti-apoptotic protein,and from a reduction in apoptosis.The present study was conducted to explore the molecular mechanism of MT-induced secondary hair follicle differentiation and development by using whole-genome analysis.Results MT had no adverse effect on the growth performance of cashmere kids but significantly improved the character of the secondary hair follicles and the quality of cashmere,and this dominant effect continued to the second year.Melatonin promotes the proliferation of secondary hair follicle cells at an early age.The formation of secondary hair follicles in the MT group was earlier than that in the control group in the second year.The genome-wide data results involved KEGG analysis of 1044 DEmRNAs,91 DElncRNAs,1054 DEcircRNAs,and 61 DEmiRNAs which revealed that the mitogen-activated protein kinase(MAPK)signaling pathway is involved in the development of secondary hair follicles,with key genes(FGF2,FGF21,FGFR3,MAPK3(ERK1))being up-regulated and expressed.We also found that the circMPP5 could sponged miR-211 and regulate the expression of MAPK3.Conclusions We conclude that MT achieves its effects by regulating the MAPK pathway through the circMPP5 sponged the miR-211,regulating the expression of MAPK3,to induce the differentiation and proliferation of seconda

Restricted feeding regimens improve white striping associated muscular defects in broiler chickens

The current study investigated the effects of intermittent feeding(IF)and fasting strategies at different times post–hatch on muscle growth and white striping(WS)breast development.In the first trial,32 one-day-old Abor Acre broilers were fed ad libitum(AL)for 3 d post–hatch and then randomly allotted into 4 feeding strategies including AL,1h-IF group(1 h IF,4 times feeding/d,1 h each time),1.5h-IF(1.5 h IF,4 times feeding/d,1.5 h each time),and fasting(1d acute fasting,6 d free access to feed)groups and fed for 7 d.Although angiogenic genes including VEGFA,VEGFR1,and VEGFR2,and myogenic genes including MYOG and MYOD were upregulated(P0.05).In the second trial,384 one-day-old birds were fed AL for 1 wk and then randomly allotted to the above 4 feeding strategies starting at 8 d of age until 42 d of age.Similarly,IF and fasting strategies upregulated the expression of angiogenic and myogenic genes(P0.05).Fasting and 1h-IF reduced the expression of adipogenic genes ZNF423 and PDGFRα(P<0.05).Moreover,IF and fasting strategies reduced fibrosis in breast muscle and reduced skeletal muscle-specific E3 ubiquitin ligases(TRIM63 and MAFBX)(P<0.05).Fasting significantly reduced CASPASE-3 in breast muscle(P<0.05).In conclusion,IF starting in the first week decreases SC number.Compared to AL,IF or fasting promotes muscular angiogenesis,increases SC number,prevents muscle degeneration,and prevents the d

Vitamin A injection at birth improves muscle growth in lambs

Vitamin A and its metabolite,retinoic acid(RA)play important roles in regulating skeletal muscle development.This study was conducted to investigate the effects of early intramuscular vitamin A injection on the muscle growth of lambs.A total of 16 newborn lambs were given weekly intramuscular injections of corn oil(control group,n=8)or 7,500 IU vitamin A palmitate(vitamin A group,n=8)from birth to 3 wk of age(4 shots in total).At 3 wk of age and weaning,biceps femoris muscle samples were taken to analyze the effects of vitamin A on the myogenic capacity of skeletal muscle cells.All lambs were slaughtered at 8 months of age.The results suggest that vitamin A treatment accelerated the growth rate of lambs and increased the loin eye area(P0.05).Moreover,vitamin A upregulated the expression of PAX7(P0.05)as evidenced by myogenic gene expression and fusion index.Taken together,neonatal intramuscular vitamin A injection promotes lamb muscle growth by promoting the myogenic potential of satellite cells.

Recent advances in fermented feeds towards improved broiler chicken performance, gastrointestinal tract microecology and immune responses: A review

Previously, fermentation has been associated with methods that improve the nutritional value of unconventional feed ingredients for broilers. In recent decades, the fermentation process has been employed to produce functional feeds that have the potential to improve broiler gastrointestinal tract microecology, health and production performance. Some of the functional ingredients found in fermented feed include lactic acid bacteria(LAB), lactic acid and other organic acids, and appear to play major roles in determining the beneficial effects of fermented feed on broiler gut health and performance. Unlike the pig, the available literature on broiler fermented feed is still rather limited. This review describes recent advances in the use of fermented feed(on the basis of conventional and unconventional feed ingredients) in broilers. Similarly, this review also shows that additional research is necessary to exploit fermented feed as a viable food source in broiler nutrition.

Standardized ileal digestible lysine requirements based on growth performance and histochemical characteristics of male broilers from 10 to 21 d of age

The growth performance and histochemical characteristics of breast muscle fibers were used to estimate the standardized ileal digestible(SID)Lys requirements for 10-to 21-d-old male broilers.Three hundred and sixty 10-d-old Ross 308 broilers(290±16.6 g)were allocated to 6 diets in a randomized complete block design with 6 replicate cages per treatment and 10 birds per cage.The 6 experimental diets were formulated to contain equally spaced increasing levels of SID Lys from 0.86%to 1.36%.The data were analyzed using the MIXED procedure of SAS.The Lys requirements were estimated by the NLIN procedure of SAS.An increase in dietary SID Lys from 0.86%to 1.36%resulted in a quadratic increase(P<0.05)in body weight gain(BWG),gain to feed ratio(G:F),breast weight,muscle cross-sectional area(MCSA),and fiber area.The SID Lys requirements based on the one-slope broken-line,quadratic line,the first intercept between the plateau of the one-slope broken-line and quadratic-line models and 95%of the upper asymptote of the quadratic-line model were estimated to be 1.01%,1.19%,1.08%,and 1.13%for BWG,1.06%,1.22%,1.11%,and 1.16%for G:F,1.10%,1.29%,1.19%,and 1.22%for breast weight,1.06%,1.22%,1.12%,and 1.16%for MCSA,and 1.14%,1.22%,1.16%,and 1.16%for breast muscle fiber area,respectively.It was concluded that the SID Lys requirements for broilers at the age of 10 to 21 d depended on the response variables used for estimation,and that histochemical characteristics of breast muscle fibers could be good indicators for estimating SID Lys requirements.

The ubiquitin ligase Nedd4-2 mediates the regulation of PepT2 by mTORC1 in bovine mammary epithelial cells

Peptide transporter 2(PepT2)transports short peptides from the blood into bovine mammary epithelial cells(BMEC)to stimulate milk protein synthesis.Despite the fact that the effect of PepT2 is acknowledged in BMEC,little is known about its regulation.This study was completed to investigate the role of mammalian target of the rapamycin(mTOR)signaling in regulating the expression and function of PepT2 in BMEC.The regulation of PepT2 by mTOR in BMEC was studied in vitro using peptide transport assay,gene silencing,Western blot.The membrane expression of PepT2 and the uptake of b-Ala-Lys-N-7-amino-4-methylcoumarin-3-acetic acid(b-Ala-Lys-AMCA),a model dipeptide,in BMEC were reduced by rapamycin(a mTOR inhibitor)and silencing of either mTOR complex 1(mTORC1)or mTOR complex 2(mTORC2),stimulated by DEP domain-containing mTOR-interacting protein(DEPTOR,endogenous inhibitor of mTORC1 and mTORC2)silencing.The trafficking of PepT2 to the membrane and the uptake of b-Ala-Lys-AMCA was promoted by neuronal precursor cell-expressed developmentally down-regulated 4 isoform 2(Nedd4-2)silencing.The effects of knockdown of mTORC1,but not mTORC2,on cell membrane expression and transport activity of PepT2 was abolished by Nedd4-2 silencing.With immunofluorescence staining,PepT2 was identified to be interacting with Nedd4-2.The Nedd4-2 expression and the interaction between PepT2 and Nedd4-2 was increased through mTORC1 knockdown,indicating an increased ubiquitination of PepT2.The results revealed that mTORC1 can regulate the expression and function of PepT2 through Nedd4-2 in BMEC.

Dietary calcium requirements of broilers fed a conventional corn-soybean meal diet from 1 to 21 days of age

Background:The current calcium(Ca)recommendation for broilers is primarily based on studies conducted more than 30 years ago with birds of markedly different productive potentials from those which exist today.And the response indicators in these studies are mainly growth performance and bone ash percentage.Therefore,the present study was carried out to investigate the effect of dietary Ca level on growth performance,serum parameters,bone characteristics and Ca metabolism-related gene expressions,so as to estimate dietary Ca requirements of broilers fed a conventional corn-soybean meal diet from 1 to 21 days of age.Methods:A total of 4201-day-old Arbor Acres male broilers were randomly assigned to 1 of 7 treatments with 6 replicates(10 birds per cage)and fed the corn-soybean meal diets containing 0.60%,0.70%,0.80%,0.90%,1.00%,1.10%or 1.20%Ca for 21 days.Each diet contained the constant non-phytate phosphorus content of about 0.39%.Results:The average daily gain decreased linearly(P<0.001)as dietary Ca level increased.The serum and tibia alkaline phosphatase(ALP)activities,tibia bone mineral density(BMD),middle toe BMD,tibia ash percentage,tibia breaking strength,and tibia ALP protein expression level were affected(P<0.05)by dietary Ca level,and showed significant quadratic responses(P<0.02)to dietary Ca levels.The estimates of dietary Ca requirements were 0.80 to 1.00%based on the best fitted broken-line or quadratic models(P<0.03)of the above serum and bone parameters,respectively.Conclusions:The results from the present study indicate that the Ca requirements would be about 0.60%to obtain the best growth rate,and 1.00%to meet all of the Ca metabolisms and bone development of broilers fed a conventional corn-soybean meal diet from 1 to 21 days of age.

Application of resistant starch in swine and poultry diets with particular reference to gut health and function

The immediate post-weaning period poses a major challenge on the survival of piglets. Similarly, newly hatched chicks face life threatening challenges due to enteric infections. In the past several years, in-feed antibiotics have been used to reduce these production problems and improve growth. However, in-feed antibiotics have been banned in many jurisdictions and therefore the most effective alternatives to infeed antibiotics must be developed. To date, several studies have been conducted to develop alternatives to antibiotics. One of the potential candidates as alternatives to in-feed antibiotics is resistant starch(RS). Resistance starch is a type of starch that resists enzymatic digestion in the upper parts of the gastrointestinal tract and therefore passes to hindgut where it can be fermented by resident microorganisms. Microbial fermentation of RS in the hindgut results in the production of short chain fatty acids(SCFA). Production of SCFA in turn results in growth and proliferation of colonic and cecal cells, increased expression of genes involved in gut development, and creation of an acidic environment. The acidic environment suppresses the growth of pathogenic microorganisms while selectively promoting the growth of beneficial microbes. Thus, RS has the potential to improve gut health and function by modifying and stabilising gut microbial community and by improving the immunological status of the host. In this review, we discussed the roles of RS in modifying and stabilising gut microbiota, gut health and function, carcass quality, and energy metabolism and growth performance in pigs and poultry.
