Conceptus elongation in ruminants: roles of progesterone, prostaglandin, interferon tau and cortisol

The majority of pregnancy loss in ruminants occurs during the first three weeks after conception, particularly during the period of conceptus elongation that occurs prior to pregnancy recognition and implantation. This review integrates established and new information on the biological role of ovarian progesterone （P4）, prostaglandins （PGs）, interferon tau （IFNT） and cortisol in endometrial function and conceptus elongation. Progesterone is secreted by the ovarian corpus luteum （CL） and is the unequivocal hormone of pregnancy. Prostaglandins （PGs） and cortisol are produced by both the epithelial cells of the endometrium and the trophectoderm of the elongating conceptus. In contrast, IFNT is produced solely by the conceptus trophectoderm and is the maternal recognition of pregnancy signa that inhibits production of luteolytic pulses of PGF2α by the endometrium to maintain the CL and thus production of P4. Available results in sheep support the idea that the individual, interactive, and coordinated actions of P4, PGs, IFNT and cortisol regulate conceptus elongation and implantation by controlling expression of genes in the endometrium and/or trophectoderm. An increased knowledge of conceptus-endometrial interactions during early pregnancy in ruminants is necessary to understand and elucidate the causes of infertility and recurrent early pregnancy loss and provide new strategies to improve fertility and thus reproductive efficiency.

Sperm immobilization activity of Allium sativum L. and other plant extracts

Aim: To identify possible spermicidal agents through screening a number of edible medicinal plants with antimicrobial activity. Methods: Initial screening was made on the basis of ram cauda epididymal sperm immobilization immediately after addition of extracts. The most potent extract was selected and was evaluated on both ram and human spermatozoa. To unravel its mode of action several sperm functional tests were carried out, namely viability of cells, hypo-osmotic swelling test for membrane integrity and assays of membrane-bound enzyme 5'-nucleotidase and acrosomal marker enzyme acrosin. Results: The crude aqueous extract of the bulb of Allium sativum L. showed the most promising results by instant immobilization of the ram epididymal sperm at 0.25 g/mL and human ejaculated sperm at 0.5 g/mL. Sperm immobilizing effects were irreversible and the factor of the extract responsible for immobilization was thermostable up to 90 癈. On boiling at 100 癈 for 10 minutes, this activity was markedly reduced. Moreover, this extract was able to cause aggregation of ram sperms into small clusters after 30 minutes of incubation at 37 癈. However this property was not found in human spermatozoa. More than 50 % reduction in sperm viability and hypo-osmotic swelling occurred in treated sperm as compared with the controls, indicating the possibility of plasma membrane disintegration which was further supported by the significant reduction in the activity of membrane bound 5'-nucleotidase and acrosomal acrosin. Conclusion: The crude aqueous extract of A. sativum bulb possesses spermicidal activity in vitro.

The secrctory materials from the uterine flushings of the pregnant, non-pregnant and pseudopreg-nant rabbits were analysed by the method of SDS-polyacrylamide gel electrophoresis. The results demon-strated that there are three specific peptides in the pregnant uterine flushings with molecular weightsof 3000, 4500 and 6000. No correspondent peptides were found in non-pregnant and pseudopregnantuterine flushings. The peptides were also isolated and identified from the blastocystic fluid. At least 4peaks of small peptides were shown by high pressure liquid chromatography. The range of their isoelec-trophoretic points is between pH 4.0-5.5 Concavalin A (Con A) affinitive electrophoresis indicated thatthey are glycopeptides in nature. The LH-RH A is able to exert inhibitory effects on the synthesis ofthese peptides. Their possible physiological function has been discussed.

UniPron is A Fully Effective Non-hormonal Reversible Contraceptive in Baboon Model (Papio Anubis)

Objective To determine the safety and efficacy of UniPron as a reversible contraceptive.Methods Vaginal swabs were obtained before and after UniPron administration, cultured onto appropriate culture media and bacteria identification was done based on type of media used, Gram stain reactions, colony morphology and biochemical tests. Vaginal biopsy tissues were processed using paraffin wax method, stained with hematoxylin and eosin and examined under light microscopy to determine the effect of the product on vaginal tissues. The effect of UniPron on sperm was examined by mixing the product with electroejaculated spermatozoa in vitro at different concentrations. For efficacy studies, male baboons of proven fertility were mated with UniPron treated or untreated females of proven fertility during the fertile stages. Results All the five females （100%） that were treated with UniPron did not conceive and they regained total fertility when the treatment was stopped while all the controls conceived. At a concentration of 40%, UniPron completely immobilized spermatozoa in an in-vitro system. UniPron mechanism of action was by lowering the vaginal pH and on application in baboon, the pH was lowered for at least 3 h after which it went back to normal.Conclusions As we plan for a study to test UniPron as a microbicide to prevent STIs including HIV, our current study has established that this novel product is effective in contraception and harmless to vaginal tissues and vaginal microbial flora in a baboon model （Papio anubis）.

Laboratory Models for the Study of the Peri-implantation Period

The greatest risks to mammalian pregnancy occur during the peri-implantation period (passage through the uterotubal junction, blastulation, zona shedding, embryonic signaling and recognition, blastocyst attachment, postattachment events initiating placentation). Conventional markers and screening methods do not provide the means to clarify the relationship between exposure of a reproductively competent woman to a xenobiotic compound and the specific error in the functional expression of an embryogenetic process identified by impairment or deletion of that process. Laboratory models which provide the flexibility of in vitro culture methods and allow integration of cellular and molecular techniques have identified cell-specific, stage-specific markers that could focus on the mechanism of xenobiotic action. In vitro models have been used to define postattachment trophoblast cell differentiation. Trophoblast specific peptide hormones prove to be valid markers of established pregnancy. They provide no assessment of the risk to the embryo during the peri-implantation period. The relationship between developmental time of exposure and risk is discussed in terms of trophoblast differentiation.

VARIATIONS OF LECTIN-BINDING GLYCOPROTEINS IN EARLY PREGNANT RABBIT EMBRYO

In the present study, the quantitative and qualitative changes of three kinds of lectinbinding glycoproteins of early pregnant rabbit embryos (D4—D12) were analyzed. The technique of Western blot, as well as video densitometer scanning and its analytic software was used for analysis. Results found that there were five specific lectin-binding glycoproteins in Day 4 to Day 6 blastocyst fluids: one ConA-binding glycoprotein about 70 kD, two WGA-binding glycoproteins respectively about 42 kD and 25kD and two PNA-binding glycoprotein respectively about 180kD and 75kD. They disappeared immediately after implan tation, It is demonstrated that there are stage-specific glycoproteins in rabbit blastocyst fluid which might be relevant to the recognition of pregnancy and implantation.

THE BIOLOGICAL FUNCTION OF RABBIT BLASTOCYST PEPTIDES (RBPs)

The inhibitory effect of rabbit blastocystic peptides (RBPs) on lymphocyte transfor-mation was studied by the method of measuring the incorporation of ~3H- thymidine intoDNA. The results indicated that RBPs inhibited PHA-stimulated rat and human lympho-cyte transformation in vitro. In the concentration ranging from 40 to 200 μg/2.5 ml, theinhibition was dose-dependent. No obvious inhibitory action was found with hCG (16- 128IU/2.5 ml), progesterone (250 ng- 1 μg/2.5 ml) and pregnant rabbit serum. It was furtherdemonstrated that RBPs at a higher dosage (200 μg/ml) was inhibitory to PGF_(2α) secretion.On the other hand, the ~3H- leucine incorporation of rabbit endometrium was enhanced bythese peptides, and this action could be blocked by the addition of actidine.

More evidence intratumoral DHT synthesis drives castration-resistant prostate cancer

Again-of-function stabilizing somatic mutation in 3β-hydroxysteroiddehydrogenase type 1 （3βHSDI, HSD3B1） was reported in castration-resistant prostate cancer. The A-C nucleotide polymorphism replaced asparagine-367 with threonine （3βHSD1-N367T） as a homozygous somatic mutation in a subset of castration-resistant prostate cancers by loss of heterozygosity of the wild-type allele. Increased stability of 3[HSD I-N367T was associated with decreased ubiquitin-mediated degradation and higher levels of dihydrotestosterone （DHT）. The studies suggest that genetic instability in castration-resistant prostate cancer favors the more stable 313HSD I-N367T mutant that contributes to drug resistance. A somatic mutation in a steroid metabolic enzyme required for DHT synthesis provides further support for intratumoral androgen synthesis contributing to prostate cancer progression.
