;~lasmid-based Survivin shRNA and GRIM-19 carried by attenuated Salmonella suppresses tumor cell growth

Persistent activation of Survivin and its overexpression contribute to the formation, progression and metastasis of several different tumor types. Therefore, Survivin is an ideal target for RNA interference mediated-growth inhibition. Blockade of Survivin using specific short hairpin RNAs （shRNA） can significantly reduce prostate tumor growth. RNA interference does not fully ablate target gene expression, owing to the idiosyncrasies associated with shRNAs and their targets. To enhance the therapeutic efficacy of Survivin-specific shRNA, we employed a combinatorial expression of Survivin-specific shRNA and gene associated with retinoid-interferon-induced mortality-19 （GRIM-19）. Then, the GRIM-19 coding sequences and Survivin-specific shRNAs were used to create a dual expression plasmid vector and were carried by an attenuated strain of Salmonella enteric serovar typhimurium （S. typhimurium） to treat prostate cancer in vitro and in vivo. We found that the co-expressed Survivin-specific shRNA and GRIM-19 synergistically and more effectively inhibited prostate tumor proliferation and survival, when compared with treatment with either single agent alone in vitroand in vivo. This study has provided a novel cancer gene therapeutic approach for prostate cancer.

Cord blood zinc status effects on pregnancy outcomes and its relation with maternal serum zinc levels:a systematic review and meta-analysis

Background The association between maternal and cord blood zinc level and pregnancy outcomes remains *** present study aims to assess whether maternal blood zinc level represents cord blood zinc level *** In this meta-analysis,systematic search was performed in PubMed,Web of Science,and Scopus databases for relevant available English articles which included mean and standard deviation values of cord blood zinc level up to April *** the assessment of the relation between cord blood zinc level and pregnancy outcomes,the pooled standard mean difference with 95%confidence interval(CI)was used and 23 studies were *** Cumulative analysis showed that cord blood zinc level was found significantly decreased in pregnancies with complications compared with healthy pregnancy controls[REM:P=0.0007,mean difference−7.9(−12.48,−3.31)].For further analysis,maternal serum zinc level status was determined from same studies to compare with cord blood levels and subgroups were detected as“Preterm”,“Preeclampsia”,“Small for gestational age/Intrauterine growth restriction and Low birth weight”.It was observed that cord blood zinc levels in subgroup analysis were also decreased and/or tend to be decreased compared to healthy pregnancies,except for preeclampsia ***,a correlation was seen between cord blood and maternal blood zinc level status(R=0.4365,95%CI−0.530,0.756;P=0.0351).Conclusion It was thought that cord blood zinc level might tend to decrease more than maternal serum zinc level in the pathological conditions during pregnancies.

The circadian rhythm of glucocorticoid receptor (GR)in human peripheral leukocytes has been reported by us previously. It remains to be shown if the circadian variation of GR has any physiological significance. In order to answer the above question, the present work studies the circadian rhythm of the reactivity of the polymorphonuclear leukocytes (PML)to glucocorticoid (GC)and analyzes the correlation between the circadian rhythm of GR and the reactivity of PML to GC.

c-Myc promotes tubular cell apoptosis in ischemia-reperfusion-induced renal injury by negatively regulating c-FLIP and enhancing FasL/Fas-mediated apoptosis pathway

The aim of this study was to investigate the role of p38 mitogen-activated protein kinase(MAPK)in cell migration induced by platelet-derived growth factor(PDGF).Western blot was performed to detect the phosphorylation of p38 in NIH3T3 cells treated with PDGF.A Transwell cell migration system was used to determine the effects of PDGF treatment on the migration of NIH3T3 cells and the influence of p38 deficiency on this process in a p38 gene knockout(p38^(−/−))mouse embryonic fibroblast cell *** the stimulation of PDGF,the migration of NIH3T3 cells was significantly increased(P<0.001)compared to the control and p38 MAP kinase was simultaneously ***,the PDGF-induced cell migration was significantly blocked in p38 gene knockout(p38^(−/−))mouse embryonic fibroblasts(MEFs)(P<0.001)as compared with the wild type cells(p38+/+).p38 MAPK plays an important role in the regulation of cell migration induced by PDGF.
