By using Ca2+ -sensitive fluorescent probe, Fura-2 , the effects of endothelial cell-conditioned medium and hypoxia on intracellular free calcium ( [Ca2+]i) in cultured pulmonary artery smooth muscle cell (PASMC) were studied. Normoxic porcine pulmonary artery endothelial cell-conditioned medium (NPAECCM) obviously elevated [Ca2+]i in PASMC,whereas the hypoxic porcine pulmonary artery endothelial cell conditioned medium (HPAECCM)significantly elevated [Ca2+]i in PASMC much more than NPAECCM. Both the effects of NPAECCM and HPAECCM were dependent on the cultured endothelial cell extracellular calcium concentrations, ranged from 1.8 mmol/L to 2. 4 mmol/***, hypoxia directly increased, which was partially inhibited by verapamil,[Ca2+]i in PASMC through Ca2+ influx *** data suggest that the augmented regulation of endothelial cell on PASMC via Ca2+ second messenger system and the hypoxia-induced Ca2+ influx into PASMC,particularly the former, may be components of mechanisms underlying hypoxic pulmonary vasoconstriction and chronic pulmonary hypertension.

The effects of 3, 4-DHAP on hypoxic vasoconstriction response in pulmonary (PA) and basilar arterial(BA) rings of rabbits and their mechanism were compared in vitro.3,4-DHAP in different concentration (2.64×10-4,7.92×10-4,2.376×10-3mol/L) decreased the basal tone of PA rings by 32. 39±9.4 mg, 68. 96±26. 54 mg and 145. 60±58.07 mg respectively,while the tension of the BA rings was decreased by 13. 80±5. 08 mg,17.18±3.36 mg and 25. 00±4.02 mg *** PA rings it also decreased the percentage increase in tension induced by hypoxia(TIH%)from the control value 48. 82±5.75% to 10.02±3. 62%,2.14±0.96%, and 0.00% respectively, while in BA rings from 27.27±5.78% to 11. 23±2.71%,7.49±1. 62%, and 1.45±1.13% respectively. The effects of 3,4-DHAP on TIH% were partially blocked by indomethacin 10 M and L-NWE 10 *** results showed that 3, 4-DHAP can decrease the hypoxic pulmonary and basilar vasoconstriction in vitro. which can be partially inhibited by cyclooxygenase inhibitor and NO/EDRF inhibitor.

Brain GABA and glutamate levels in workers of two ant species （Hymenoptera： Formicidae）： Interspecific differences and effects of queen presence/absence

Presence of amino acid neurotransmitters gamma-aminobutyric acid （GABA） and glutamate （Glu） in ant brains was reported in very few studies. To learn more about factors influencing GABA and Glu levels in ant brains, we applied high-performance liquid chromatography to measure levels of these compounds in single brains of workers of 2 ant species, Myrmica ruginodis （subfamily Myrmicinae） and Formica polyctena （subfamily Formicinae） taken from queenright/queenless colony fragments and tested in dyadic aggression tests consisting of an encounter with a nestrnate, an alien conspecific or a small cricket. Brain glutamate levels were higher than those of GABA in both tested species. Brain GABA levels （in μmol/brain） and GABA ： Glu ratio were higher in M. ruginodis （a submissive species） than in E polyctena （a dominant, aggressive species） in spite of smaller brain weight of M. ruginodis. Brain glutamate levels （in μmol/brain） did not differ between the tested species, which implies that glutamate concentration （in μmol/mg of brain tissue） was higher in M. ruginodis. Queen absence was associated with increased worker brain GABA levels in E polyctena, but not in M. ruginodis. No significant effects of opponent type were discovered. As GABA agonists enhance friendly social behavior in rodents, we hypothesize that elevated brain GABA levels of orphaned workers of *** facilitate the adoption of a new queen. This is the first report providing information on GABA and glutamate levels in single ant brains and documenting the effects of queen presence/absence on brain levels of amino acid neurotransmitters in workers of social Hymenoptera.

The pathophysiology of Huntington's disease involves high levels of the neurotoxin quinolinic acid. Quinolinic acid accumulation results in oxidative stress, which leads to neurotoxicity. However, the molecular and cellular mechanisms by which quinolinic acid contributes to Huntington's disease pathology remain unknown. In this study, we established in vitro and in vivo models of Huntington's disease by administering quinolinic acid to the PC12 neuronal cell line and the striatum of mice, respectively. We observed a decrease in the levels of hydrogen sulfide in both PC12 cells and mouse serum, which was accompanied by down-regulation of cystathionine β-synthase, an enzyme responsible for hydrogen sulfide production. However, treatment with NaHS(a hydrogen sulfide donor) increased hydrogen sulfide levels in the neurons and in mouse serum, as well as cystathionine β-synthase expression in the neurons and the mouse striatum, while also improving oxidative imbalance and mitochondrial dysfunction in PC12 cells and the mouse striatum. These beneficial effects correlated with upregulation of nuclear factor erythroid 2-related factor 2 expression. Finally, treatment with the nuclear factor erythroid 2-related factor 2inhibitor ML385 reversed the beneficial impact of exogenous hydrogen sulfide on quinolinic acid-induced oxidative stress. Taken together, our findings show that hydrogen sulfide reduces oxidative stress in Huntington's disease by activating nuclear factor erythroid 2-related factor 2,suggesting that hydrogen sulfide is a novel neuroprotective drug candidate for treating patients with Huntington's disease.

Background Intrarenal activation of the renin angiotensin system (RAS) plays an important role in mediating renal fibrosis. Both angiotensin converting enzyme inhibitors (ACEIs) and angiotensin Ⅱ (AngⅡ) receptor antagonists have been shown to exert a protective role against diabetic and non-diabetic nephropathy. However, the exact mechanism of how blocking local RAS prevents renal fibrosis is unclear. The present study was to investigate the influence of a new AngⅡ receptor antagonist, irbesartan (Irb), on AngⅡ-induced hypertrophy in human proximal tubular cell line (HK-2). Methods The cell line, HK-2, was grown in Dulbeccos’s Modified Eagle’s Medium containing 10% heat-inactivated fetal calf serum. After rested in serum-free medium for 24 hours, the effects of Irb on AngⅡ (10 -7 mol/L)-induced [ 3H]-leucine incorporation, total protein content (measured by the Coomassie brilliant blue G250 method), and change in cell size (determined by scanning electron microscopy) were observed. The influence of Irb on the cell cycle was analyzed by fluorescence activated cell sorter (FACS) flow cytometry. Results AngⅡ induced cell hypertrophy in a time and dose dependent manner. Stimulation of cells with AngⅡ for 48 hours resulted in a increase in [ 3H]-leucine incorporation [0 hour: (5584±1016) cpm/10 5cells vs 48 hours: (10741±802) cpm/10 5cells, P<0.05], which was significantly attenuated by treatment with Irb. AngⅡ significantly increased the total protein content in HK-2 cells [control: (0.169±0.011) mg/10 5cells vs AngⅡ group: (0.202±0.010) mg/10 5 cells, P<0.05], which was also markedly inhibited by cotreatment with Irb (P<0.01). Scanning electron microscopy showed that AngⅡ induced an increase in average physical cell size, which was significantly inhibited by Irb [control: (11.92±1.62) μm; AngⅡ group: (20.63±3.83) μm; AngⅡ+Irb group: (13.59±3.15) μm; P<0.01 vs control, respectively]. Although laparoscopy has the potential to reduce peritoneal trauma and post-operative peritoneal adhesion formation, only one randomized controlled trial and a few comparative retrospective clinical studies have addressed this issue. Laparoscopy reduces de novo adhesion formation but has no efficacy in reducing adhesion reformation after adhesiolysis. Moreover, several studies have suggested that the reduction of de novo post-operative adhesions does not seem to have a significant clinical impact. Experimental data in animal models have suggested that CO2 pneumoperitoneum can cause acute peritoneal inflammation during laparoscopy depending on the insufflation pressure and the surgery duration. Broad peritoneal cavity protection by the insufflation of a low-temperature humidified gas mixture of CO2, N2O and O2 seems to represent the best approach for reducing peritoneal inflammation due to pneumoperitoneum. However, these experimental data have not had a significant impact on the modification of laparoscopic instrumentation. In contrast, surgeons should train themselves to perform laparoscopy quickly, and they should complete their learning curves before testing chemical anti-adhesive agents and anti-adhesion barriers. Chemical anti-adhesive agents have the potential to exert broad peritoneal cavity protection against adhesion formation, but when these agents are used alone, the concentrations needed to prevent adhesions are too high and could cause major post-operative side effects. Anti-adhesion barriers have been used mainly in open surgery, but some clinical data from laparoscopic surgeries are already available. Sprays, gels, and fluid barriers are easier to apply in laparoscopic surgery than solid barriers. Results have been encouraging with solid barriers, spray barriers, and gel barriers, but they have been ambiguous with fluid barriers. Ferroptosis is a form of regulated cell death driven by the accumulation of lipid *** of ferroptosis might be beneficial to cancer ***-coding RNAs(ncRNAs)are a class of RNA transcripts that generally cannot encode proteins and have been demonstrated to play critical roles in regulating ***,we developed ncFO,the ncRNA-ferroptosis association database,to document the manually curated and predicted ncRNAs that are associated with ***,ncFO contains 90 experimentally verified entries,including 46 microRNAs(miRNAs),21 long non-coding RNAs(lncRNAs),and 17 circular RNAs(circRNAs).In addition,ncFO also incorporates two online prediction tools based on the regulation and co-expression of ncRNA and ferroptosis *** default parameters,we obtained 3260 predicted entries,including 598 miRNAs and 178 lncRNAs,by regulation,as well as 2,592,661 predicted entries,including 967 miRNAs and 9632 lncRNAs,by ncRNA-ferroptosis gene co-expression in more than 8000 samples across 20 cancer *** detailed information of each entry includes ncRNA name,disease,species,tissue,target,regulation,publication time,and PubMed *** also provides survival analysis and differential expression analysis for *** summary,ncFO offers a user-friendly platform to search and predict ferroptosisassociated ncRNAs,which might facilitate research on ferroptosis and discover potential targets for cancer treatment.
