New generation sequencing systems are changing how molecular biology is practiced. The widely promoted $1000 genome will be a reality with attendant changes for healthcare, including personalized medicine. More broadly the genomes of many new organisms with large samplings from populations will be commonplace. What is less appreciated is the explosive demands on computation, both for CPU cycles and storage as well as the need for new computational methods. In this article we will survey some of these developments and demands.

Valine is an essential amino acid and a type of branched-chain amino acid. Due to the involvement of branchedchain amino acids in various metabolic pathways, there has been a surge of interests in valine nutrition and its role in animal physiology. In pigs, the interactions between valine and other branched-chain amino acids or aromatic amino acids are complex. In this review, we delve into the interaction mechanism, metabolic pathways, and biological functions of valine. Appropriate valine supplementation not only enhances growth and reproductive performances, but also modulates gut microbiota and immune functions. Based on past observations and interpretations, we provide recommended feed levels of valine for weaned piglets, growing pigs, gilts, lactating sows, barrows and entire males. The summarized valine nutrient requirements for pigs at different stages offer valuable insights for future research and practical applications in animal husbandry.

As immunotherapy has gained increasing interest as a new foundation for cancer therapy,some atypical response patterns,such as pseudoprogression and hyperprogression,have garnered the attention of *** is a phenomenon in which an initial increase in tumor size is observed or new lesions appear,followed by a decrease in tumor burden;this phenomenon can benefit patients receiving immunotherapy but often leads to premature discontinuation of treatment owing to the false judgment of *** recognizing pseudoprogression is also a challenge for *** of the extensive attention on pseudoprogression,significant progress has been *** new criteria for immunotherapy,such as irRC,iRECIST and imRECIST,were proposed to accurately evaluate the response to *** new detection indexes,such as ctDNA and IL-8,have also been used to identify *** this review,the definition,evaluation criteria,mechanism,monitoring,management and prognosis of pseudoprogression are summarized,and diagnostic and treatment processes for patients with progression but with a suspicion of pseudoprogression are proposed;these processes could be helpful for physicians in clinical practice and enhances the understanding of pseudoprogression.

Cells producing cytokines often express the receptor for the same cytokine, which makes them prone to autocrine signaling. How cytokine release and signaling are regulated in the same cell is not understood? In this study, we demonstrate that signaling by exogenous and self-synthesized inflammatory cytokine interleukin-6 (IL-6) within endosomal compartments acts as a cellular brake that limits the synthesis of IL-6. Our data show that IL-6 is internalized by dendritic cells and signals from endosomal compartments containing the IL-6 receptor. Newly synthesized IL-6 also traffics via these endosomal compartments and signals in transit to the plasma membrane. This allows activation of STAT3 which in turn limits toll-like receptor 4 stimulant lipopolysaccharide (LPS) triggered transcription of IL-6. Long-term exposure to LPS removes this brake via inhibition of STAT3 by increased expression of suppressor of cytokine signaling 3 and results in fully fledged IL-6 production. This transient regulation could prevent excessive IL-6 production during early infections.

Camptothecin is a complex monoterpenoid indole alkaloid with remarkable antitumor *** that two C-10 modified camptothecin derivatives,topotecan and irinotecan,have been approved as potent anticancer agents,there is a critical need for methods to access other aromatic ringfunctionalized congeners(e.g.,C-9,C-10,etc.).However,contemporary methods for chemical oxidation are generally harsh and low-yielding when applied to the camptothecin scaffold,thereby limiting the development of modified *** herein,we have identified four tailoring enzymes responsible for C-9 modifications of camptothecin from Nothapodytes tomentosa,via metabolomic and transcriptomic *** consist of a cytochrome P450(nt CPT9H)which catalyzes the regioselective oxidation of camptothecin to 9-hydroxycamptothecin,as well as two methyltransferases(nt OMT1/2,converting 9-hydroxycamptothecin to 9-methoxycamptothecin),and a uridine diphosphate-glycosyltransferase(nt UGT5,decorating 9-hydroxycamptothecin to9-β-D-glucosyloxycamptothecin).Importantly,the critical residues that contribute to the specific catalytic activity of nt CPT9H have been elucidated through molecular docking and mutagenesis *** work provides a genetic basis for producing camptothecin derivatives through metabolic *** will hasten the discovery of novel C-9 modified camptothecin derivatives,with profound implications for pharmaceutical manufacture.

Efficient enrichment and selective recovery of rare earths from sulfate leachate of ion-adsorption type rare earth ore by extraction with HPOAc

The efficient enrichment and separation of rare earth elements（REEs） from sulfate leachates of ionadsorption type rare earth ore（IATREO） is still a *** work presents a novel extractant 2-(bis(（2-ethylhexyl）oxy)phosphoryl)-2-hydroxyacetic acid（HPOAc） for the selective extraction and efficie nt enrichment of REEs from sulfate leachates of *** exhibits higher extraction ability for all fifteen REEs（Ⅲ） than naphthenic acid（NA） at pHini=1,***,it has no drawbacks of di-（2-ethylhexyl） phosphoric acid（P204） and 2-ethylhexyl phosphoric acid mono-2-ethylhexylester（P507）such as weak extraction ability towards light REEs（Ⅲ） and high stripping acidity for heavy REEs（Ⅲ）.It has better separation performance for metal ion impurities than P204 and P507,especially for the typical impurity Al（Ⅲ）.Furthermore,the HPOAc system has better phase separation behavior and extraction pheno mena.A simulated operation with two-stage counte r-current extraction and single-stage stripping of REEs was carried out using unsaponified *** recovery of REEs reaches 98.7%.The concentration of REEs increases from 0.44 to 130.35 g/L,indicating a nearly 300-fold ***,the content of REEs increases from 77.8 wt% to 97.6 wt%.So HPOAc has the potential to selectively recover REEs from sulfate leachates of IATREO.
