Background: Major Depressive Disorder (MDD) is a mental disorder characterized by a pervasive and persistent low mood which is accompanied by low self-esteem and loss of interest or pleasure in day to day activities that adversely affects a person’s family, work, and personal life. There is no sufficient laboratory test for the diagnosis of MDD and it is expected that this investigation may be helpful for better diagnosis and management of MDD. We aimed to measure serum immunoglobulin levels in MDD patients and control subjects to meet the above demand. Methods: For this purpose, we recruited 88 MDD patients from the department of psychiatry, Bangabandhu Sheikh Mujib Medical University, Dhaka and 89 healthy volunteers from Dhaka city matched with age, sex and socioeconomic status to the patient group. Turbidimetry method was applied to measure serum levels of immunoglobulin A, G, and M where immunoglobulin kit was utilized. Results: The current study revealed that mean serum concentrations of immunoglobulin A, G, and M in patients were found to be 209.07 ± 104.93, 791.50 ± 235.67 and 107.92 ± 47.53 mg/dL while those were 195.34 ± 92.16, 763.81 ± 175.89 and 99.17 ± 48.78 mg/dL in control subjects, respectively. Conclusion: Our result indicates that serum concentrations of immunoglobulin A, G and M were not significant between the groups and further studies are required to establish these findings.

Objective:In this study,we focus on the hypoglycemic effects of chlorogenic acid(CGA)in vitro and in vivo and its mechanism base on regulate glucose and lipid metabolism via AMPK ***:The cytotoxicity,glucose consumption and intracellular triglyceride assay were been detected by commercial *** western blots were used to detection the associated protein levels after CGA treatment,and the inhibiter blocking experiments were also be *** vivo experiment,the fasting blood-glucose,lipid metabolism,liver function,insulin resistance,glucose tolerance,and pathological change were assessed on streptozocin induced diabetic ***:We found that CGA exhibited no cytotoxicity at concentrations of 100μM,it caused a significant increasing of glucose consumption and reducing of the PA-BSA induced intracellular TG level on HepG2 cells at 50μM and 100μM treatment,CGA exhibited up-regulating the level of p-AMPK(Thr172)and p-ACC(Ser79)in dose-dependent manners in vitro and in *** stimulating activities of CGA on AMPK were completely blocked by compound c(CC)on HepG2 *** the efficacies of CGA on glucose consumption and intracellular TG accumulation were also completely blocked by CC *** CGA also exhibited potent anti-diabetic effects with hypoglycemic activity,improve insulin resistance and glucose tolerance,regulate glucose and lipid metabolism and protect the liver function in ***:Our results suggested that CGA can regulate glucose and lipid metabolism by AMPK activation,and exhibit potent anti-hyperglycemic effect in streptozocin induced diabetes mice,and may be used as a potential effective anti-diabetes drug.

OBJECTIVE This study discusses the effects of scutellarin(SCU),which is one of effective component of Erigeron breviscapus(Vant.)Hand-Mazz,on cGMP dependent protein kinase(PKG)in human cardiac microvascular endothelial cells(HCMECs)after hypoxia roxygenation(HR).METHODS Based on a model of HCMECs cells with HR injury,cell viability is examined by MTT *** expression of PKG is analyzed by Western blotting and its enzyme activity is investigated by ELISA *** The results of MTT assay- indicate that SCU(1,10μmol·L1)could protect HCMECs against HR ***(0.1,1 and 10μmol·L-1)canenhance PKG-I expression in control and HR injury ***,SCU(1,10μmol·L-1 significantly increase PKG activity in control cells(P<0.01),and also SCU(100μmol·L-1)appears similar on enzyme activity in HR injury cells(P <0.05).CONCLUSION SCU appears protective effects on endothelial cells against HR *** its mechanisms may be related to the influence of SCU on PKG activity in HCMECs.

Effect of Topiramate on Serum Proteome of Mice Bearing Lewis Lung Carcinoma

Topiramate has been used in epilepsy patients with brain tumor recently. In our present research we used the animal model of Lewis lung carcinoma metastasis in C57BL/6 mice to confirm the anti-tumor and anti-metastasis activity of topiramate. Then by using two-dimensional electrophoresis (2-DE) combined with matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MS), we analyzed and compared the protein profile of neoplasm serum before and after treated with topiramate (25, 50, 100 mg·kg^-1·d^-1, for 21 days, p.o.), trying to discover its possible targets. The results showed that topiramate could dramatically reduce the primary tumor growth (P < 0.05) and damage of lung alveolar which caused by metastasic tumor deposits. The 2-DE and software analysis results revealed 401 protein spots in control gel and 396 protein spots in topiramate treated (50 mg·kg^-1·d^-1, 21 d) gel, and 228 matched protein spots between the two groups, indicating that intensive changes had occurred during the process of treatment. Four of those obviously down regulated protein spots after topiramate treatment were processed to the peptide massfingerprinting. Data search results showed that the protein peptides might be Splice isoform 2 of Tropomyosin or Chain 1 of Osteopontin, Chain 1 of Transthyretin, Chain 1 of Serum amyloid A-1 protein and Chain 1 of Serum amyloid A-2 protein respectively. The results suggest that topiramate has a strong anti-tumor and anti-metastasis effect on Lewis-lung-carcinoma. The mechanism may be related to its regulation on plenty of proteins; in particular, on down-regulation of osteopontin and serum amyloid A protein expression in serum.

Sildenafil and bosentan are often co-administered for pulmonary arterial hypertension (PAH) treatment. The plasma concentration of sildenafil can be decreased by half if co-administered with bosentan. Many patients take these agents simultaneously in the morning and the evening. The aim of this study was to examine the pharmacokinetics of sildenafil which was interfered with bosentan administration to ascertain whether these agents should be given concomitantly or separately. A two-way crossover study was conducted in 6 PAH patients with combination therapy of sildenafil and bosentan. Participants underwent the sequence of treatment phases: phase S (sildenafil administered 3 h before bosen-tan);phase B (bosentan administered 3 h before sildenafil);and phase C (administered concomitantly). Blood samples were collected on the last day of each phase. There was no significant difference in maximum plasma concentration or area under the plasma concentration-time curve (AUC0-8) between phase C and phase S (95.5 ± 24.8 vs. 72.9 ± 40.9 (p = 0.07), 209.7 ± 81.8 vs. 180.2 ± 126.4 (p = 0.24), respectively) or between phases C and B (87.8 ± 42.0 vs. 99.6 ± 33.9 (p = 0.59), 197.2 ± 88.2 vs. 240.7 ± 121.8 (p = 0.19), respectively) (ng/mL, mean ± standard deviation). Large intra-and inter-individual variability in sildenafil concentration was noted. The timing of administration of sildenafil and bosentan does not significantly influence the plasma concentration of sildenafil. Physicians do not need to be overly concerned about the timing of administration of these drugs to maximize the sildenafil concentration.

＜正＞ Aggretin,a heterodimeric platelet aggregation inducer purified from Calloselasma rhodostoma venom, was identified as a collagen - like integrin alpha2betal agonist. In these studies we explore the receptor and signal transduction involved in aggretin - stimulated migration and proliferation of VSMCs. Aggretin significantly increased VSMCs proliferation as determined by tetrazolium assay. Moreover. VSMCs migration toward immobilized aggretin was increased in a modified Boy- den chamber. Incubation of VSMCs with aggretin stimulated the phosphorylation of phosphatidylinositol 3 - kinase （PI3K）. Akt and extracellular - regulated kinase（ERK） in a time - dependent manner. In a similar fashion, aggretin also induced phosphorylation of eNOS and PDGF beta receptor. Focal adhesion kinase （FAK） was phosphorylated within the first 5 min. The Enos activating related signaling is also elucidated. In conclusion, aggretin activates FAK. PI3K/Akt. ERK. eNOS and PDGF pathways leading to promoting of migration and proliferation of VSMCs.

Background Cardiac hypertrophy is an adaptive enlargement of the myocardium in response to stress or inju- ry. Many studies have revealed that microRNAs participate in cardiac hypertrophy, but the role of microRNA-214 -3p （miR-214-3p） in cardiac hypertrophy has not been illustrated. The present study aimed to investigate the ex- pression and potential role of miR-214-3p in a mouse model of angiotensin-II （Ang-II） infusion-induced myocar- dium hypertrophy. Methods A mouse model of hypertrophy was established by Ang-II infusion through osmot- ic pump. And a cell model was also achieved based on Ang-Ⅱ-promoted hypertrophy in neonatal mouse ventricu- lar cardiomyocytes （NMVCs）. Enforced enhancement of miR-214-3p was obtained in a mouse Ang-II infusion model via tail vein injection of miR-214-3p agomir. Left-ventricular （LV） structure and function variables were assessed by transthoracic echocardiography. Wheat germ agglutinin （WGA） staining was used to show cell size of mouse myocardium eardiomyocytes. FITC- phalloidin staining was performed to show the cell area of NMVCs. Expressions of miR-214-3p, ANP and β-MHC were detected by real-time quantitative PCR and western -blot assay respectively. Results MiR-214-3p expression was markedly attenuated in hypertrophic myocardium of a mouse Ang-Ⅱ infusion model, but was increased in ang-Ⅱ-induced NMVCs. MiR-214-3p could significantly suppress the increase of cell size and protein expression of ANP and 1β-MHC in Ang-Ⅱ-treated NMVCs. Cardiac hypertrophy in a mouse Ang-Ⅱ infusion model was ameliorated by enforced increase of miR-214-3p, with the de- creases in cell size of cardiomyocyte and protein expression of ANP and β-MHC. Conclusion MiR-214-3p ex- pression is downregulated in the myocardium of a mouse Ang-Ⅱ infusion model, and miR-214-3p could suppress Ang-Ⅱ-induced cardiac hypertrophy.
