Systemic Lupus Erythematosus Complicated with Femoral Head Ischemic Necrosis Treated by Chinese medicine Therapy for Activating blood and Dredging Collaterals Method

Objective：To observe the effect and mechanism of Chinese medicine therapy for activating blood and dredging collaterals（ABDC） on treating systemic lupus erythematosus complicated with avascular necrosis of the femoral head（SLE-ANFH）.Methods：Thirty-four patients（51 joints） with SLE-ANFH were assigned by a random number table to two groups：22 patients（32 joints） in the treatment group and 12 patients （19 joints） in the control *** received Western medical conventional treatment for anti-inflammation and immunosuppression,but an additional Chinese medicine decoction prescribed based on ABDC principle was administered to patients in the treatment *** observation on the patients＇ condition and therapeutic effect lasted for 3 ***：The patients＇ conditions in the two groups,as assessed by Association for Research Circulation Osseous（ARCO） staging,were similar before *** treatment,comparison between groups showed significant difference（P0.05）,and the raised Harris functional scores in the treatment group were higher than that in the control group（P0.01）.The post-treatment symptom improving rate in the treated group was 72.73%,which was higher than that in the control group（50.00%,P0.05）.Moreover,the former was superior in improving hematologic and hemorrheologic parameters in terms of prolonging activated partial thromboplastin time,lowering whole blood middle/low shear viscosity,and plasma viscosity（P0.05 or P0.01）. Two patients in the control group but none in the treatment group received hip joint replacement operation during the observation ***：Chinese medicine ABDC therapy could effectively alleviate clinical symptoms and improve joint function of patients with *** mechanism may be related to its effects on improving high coagulation manner and trend for getting embolism.

Background: The effect of repeated blood donation on some biochemical values of healthy adult male blood donors in Calabar, Nigeria was studied. Methods: One hundred and fifty three (153) healthy repeat blood donors and 90 first time blood donors constituted the study population. Samples were analyzed using colorimetric procedures. Results: The BMI values of 24.4 ± 2.4 kg/ m2 in the first time donors was significantly higher than the 21.7 ± 1.7 kg/m2 obtained in repeat donors (P 2 = 0.7934, P Conclusion: A reduction in the values of some lipid profiles and high GGT activity is associated with repeated blood donations in this study population. Repeated blood donation may play a significant role in reducing the incidence of heart disease.

blood viscosity plays an important role in maintaining vascular homeostasis. Under normal physiological conditions, blood viscosity is kept at a relatively stable level by a variety of regulatory mechanisms. However, under pathological conditions, disorders in these regulatory mechanisms mediated by endothelial cell dysfunction result in the pathogenesis and development of certain diseases, such as cardio-cerebrovascular diseases, diabetes mellitus, and hemorrhagic shock. Therefore, monitoring and regulating blood viscosity are important for the diagnosis, treatment, and prognosis of disease. This article reviews the role of blood viscosity regulation and its importance in disease prevention and treatment.

The presence of mesenchymal progenitor cells within bone marrow has been known since the late nineteenth century. To date, mesenchymal stem cells (MSCs) have been isolated from several different connective tissues, such as adipose tissue, muscle, placenta, umbilical cord matrix, blood, liver, and dental pulp. Bone marrow, however, is still one of the major sources of MSCs for preclinical and clinical research. MSCs were first evaluated for regenerative applications and have since been shown to directly influence the immune system and to promote neovascularization of ischemic tissues. These observations have prompted a new era of MSC transplantation as a treatment for various diseases. In this review, we summarize the important studies that have investigated the use of MSCs as a therapeutic agent for regenerative medicine, immune disorders, cancer, and gene therapy. Furthermore, we discuss the mechanisms involved in MSC-based therapies and clinical-grade MSC manufacturing.

Genetical Study of Mutation in Maternal-fetal ABO Incompatibility

This study looked into a family involving a rare mother-child ABO blood type inconsistency and explored its genetic and molecular basis. In the family, the mother had type AB blood and the father was blood type B and they gave birth to a baby of blood type O. Their blood types were phenotypically identified by using different techniques, including micro-column gel test, immune inhibition test, absorption and elution tests. The sequences of all 7 exons of ABO allele from the core family members were determined by using PCR and clone-based sequencing. The loci of mutated gene were compared against normal human genes. The result showed that the mother＇s erythrocytes were agglutinable with monoclonal anti-A antibody（2＋） and had agglutination reaction with anti-B antibody（4＋）. The mother＇s serum registered agglutination action with standard blood type A cells. The findings showed an ABO inconsistency. When domestic antibodies were used, the mother＇s erythrocytes yielded agglutination reaction with humanized anti-B serum（4＋） and anti-B monoclonal antibody but were non-agglutinable with humanized anti-A serum and anti-A monoclonal antibody. Upon absorption and elution, the titer of anit-A antibody was 128 both before and after the absorption test, with no significant difference found between pre- and post-absorption values. Our results confirmed that the mother＇s allelic gene was type B and contained type A. The father＇s blood type was type B, and son＇s blood type was type O. Clone-based sequencing revealed that the mother carried a heterozygous gene of B101.01（nt A640→G）/O01, which contained an M214→V mutation that could express a weak expression of antigen A, resulting in blood type AB. However, their son did not have the M214→V mutation, which yielded a false ABO-inconsistency between him and his mother. We were led to conclude that type B gene with a M214→V mutation can encode both antigen B and weak antigen B that can lead to false ABO-inconsistencies.

Objective: To assess the knowledge, attitudes and practices regarding blood donation in the general population. Methods: A cross-sectional study was conducted and enrolled 416 people between 18 and 65 years old at random, living in three health zones in the city of Bukavu in eastern DRC. These subjects responded to a questionnaire on knowledge, attitudes and practices regarding blood donation. Results: Our sample consisted of 61.5% men, 70.9% of people from one level of education at the secondary level and 60% unemployed. According to the surveyed subjects blood was considered as a fuel of the body in 44.6% and as a source of life in 44.1%. Sixty-one percent of the population did not know the practice of blood donation;this knowledge differed significantly (p blood had negative effects and 27.4% did not know where the blood bags were stored. Channels of knowledge about blood donation practices were associations of blood donors (30.9%), awareness campaigns (18.2%), school (17.3%) and media (15.5%), churches (10%), and friends (8.1%). Approximately 85% of subjects who knew blood donation agreed with blood donation and blood transfusion but only 54.9% had donated blood in their lifetime, with a proportion of loyalty to blood donation (31.8%). In the population 59.4% were willing to donate blood. Motivation to donate blood most common (66%) was volunteerism while 19.1% were willing to donate blood for a family member and 12.6% willing to donate blood against money. The reasons for refusal to donate blood identified were fear of contracting diseases especially HIV, lack of information, religious beliefs, the sale of the collected blood by medical staff, and fear of test result for HIV. Conclusion: Factors of refusal to donate blood were identified. Efforts and new strategies tailored to these factors must be developed and implemented to increase blood donation.

Gene therapy has shown significant potential in treating various diseases,particularly inherited blood disorders such as hemophilia,sickle cell disease,and *** in understanding the regulatory network of disease-associated genes have led to the identification of additional therapeutic targets for treatment,especially for β-*** regulatory factor BCL11A offers the most promising therapeutic target for β-hemoglobinopathies,and reduction of its expression using the commercialized gene therapy product Casgevy has been approved for use in the UK and USA in ***,the emergence of innovative gene editing technologies has further broadened the gene therapy landscape,presenting possibilities for *** studies indicate that base editing and prime editing,built upon CRISPR technology,enable precise single-base modification in hematopoietic stem cells for addressing inherited blood disorders ex vivo and in *** this review,we present an overview of the current landscape of gene therapies,focusing on clinical research and gene therapy products for inherited blood disorders,evaluation of potential gene targets,and the gene editing tools employed in current gene therapy practices,which provides an insight for the establishment of safer and more effective gene therapy methods for a wider range of diseases in the future.
