S-adenosylmethionine Administration Attenuates Low Brain-Derived Neurotrophic Factor Expression Induced by Chronic Cerebrovascular Hypoperfusion or Beta Amyloid Treatment

Chronic cerebrovascular hypoperfusion is a high-risk factor for Alzheimer＇s disease（AD） as it is conducive to beta amyloid（Ab） over-production. Brainderived neurotrophic factor（BDNF） is a member of the neurotrophin family widely expressed in the central nervous system. The structure of the rat BDNF gene is complex, consisting of eight non-coding exons（I–VIII） and one coding exon（IX）. The BDNF gene is transcribed from multiple promoters located upstream of different 50 noncoding exons to produce a heterogeneous population of BDNF m RNAs. S-adenosylmethionine（SAM） produced in the methionine cycle is the primary methyl donor and the precursor of glutathione. In this study, a cerebrovascular hypoperfusion rat model and an Ab intrahippocampal injection rat model were used to explore the expression profiles of all BDNF transcripts in the hippocampus with chronic cerebrovascular hypoperfusion or Ab injection as well as with SAM treatment. We found that the BDNF m RNAs and protein were down-regulated in the hippocampus undergoing chronic cerebrovascular hypoperfusion as well as Ab treatment, and BDNF exons IV and VI played key roles. SAM improved the low BDNF expression following these insults mainly through exons IV and *** results suggest that SAM plays a neuroprotective role by increasing the expression of endogenous BDNF and could be a potential target for AD therapy.

Plexiform angiomyxoid myofibroblastic tumor of the stomach is a unique mesenchymal tumor that we first described in *** tumor is very rare,and to date,only 18 cases confirmed by immunohistochemistry have been reported in the *** patients' ages ranged from 7 to 75 years(mean,43 years),and the male-to-female ratio was approximately 1:*** clinical symptoms are ulceration,associated upper gastrointestinal bleeding(hematemesis),and *** tumors are located at the antrum in all cases,and grossly,the tumor is whitish to brownish or reddish,and forms a lobulated submucosal or transmural ***,the tumor is characterized by a plexiform growth pattern,the proliferation of cytologically bland spindle cells,and a myxoid stroma that is rich in small vessels and positive for Alcian blue ***,the tumor cells are positive for α-smooth muscle actin and negative for KIT and *** diagnoses include gastrointestinal stromal tumor and other mesenchymal tumors of the gastrointestinal *** authors proposed that this tumor should be designated as "plexiform fibromyxoma",but this designation might cause *** tumor is probably benign and thus far,neither recurrence nor metastasis has been reported.

Biological Significance and the Related Molecular Mechanism of Ets1 mRNA Expression in Lung Cancer by Tissue Microarray(TMA)

Objective： To investigate the expressions and proteins in the pathogenesis, progression of lung molecular mechanism of Ets-1 mRNA, and TGFβ1 and c-Met cancer by tissue microarray （TMA） method. Methods： The expressions of Ets-1 mRNA, and TGFβ1 and c-Met proteins were detected in 89 primary lung cancers, 12 lung cancer with lymph-node metastasis and 12 precancerous lesions by FISH（fluorescence in situ hybridization） and immunohistochemical method, and 10 normal lung tissues were used as controls. Results： The expressions of Ets-1 rnRNA, and TGFβ1 and c-Met proteins were significantly higher in 89 primary lung cancer than in the control group （P〈0.05）. The expressions of Ets-1 mRNA, and TGFβ1 and c-Met proteins were related to lymph node metastasis and clinical stages. There was a positive correlation between the Ets-1 mRNA expression and TGFβ1 and c-Met proteins （P〈0.05）. Conclusion： Ets-1 mRNA, TGFβ1 and c-Met proteins may be related to the pathogenesis, progression and malignant behavior of lung cancer. They may play an important role in prognosis assessment of lung cancer.

ABCG5-positivity in tumor buds is an indicator of poor prognosis in node-negative colorectal cancer patients

AIM:To analyze the expression of 8 putative cancer stem cell(CSC) markers within colorectal cancer tumor buds and to determine their prognostic impact in patients with this disease. METHODS:Immunohistochemistry was performed on 101 colorectal cancer resections for CK22(to identify tumor buds) as well as CD133,CD166,CD24,CD44s,CD90,EpCAM,ALDH1,and ABCG5,and their expression within tumor buds was evaluated. RESULTS:CD90,CD44s,and CD133 expression in tumor buds was found in less than 5%of all cases. ALDH1,CD24,CD166 were expressed in 16.5%,16.2%,and 34%cases,respectively,while ABCG5 and EpCAM expression was more frequent and found in 35%and 69%of cases,*** the 8 markers studied,EpCAM and ABCG5 positivity in tumor buds were significantly associated with poor prognosis(P=0.023,P=0.038,respectively) in multivariable analysis with pT and pN classificationP=0.048;hazard ratio(HR) :2.64;95%CI:1.0-6.9,for EpCAM and P=0.029;HR:2.22;95%CI:1.0-4.5,for *** survival time was particularly striking for lymph node-negative patients with ABCG5-positive buds(P<0.001) . CONCLUSION:Expression of putative stem cell markers EpCAM and ABCG5 within the tumor buds of colorectal cancer are frequently noted and are associated with poor prognosis.

Down-Regulation of Notchl and NF-κB by Curcumin in Breast Cancer Cells MDA-MB-231

Objective： To test whether the down-regulation of Notchl gene expression by curcumin could inhibit cell growth and induce apoptosis, which may be associated mechanistically with the down-regulation of NF-κB in breast cancer cells. Methods： Breast cancer cell lines MDA-MB-231 were cultured in vitro and treated with different dosages of curcumin （0, 1.25, 5.0, 20.0μmol/L） for dose-dependent assay and different time （0, 24, 48, 72 h） at the dosage of 5.0μmol/L for time course assay. The changes of the mRNA and protein expression of Notchl and NF-κB were measured by RT-PCR and Western Blot, and MTT assay was used to measure the change of proliferation. Results： The mRNA and protein levels of Notchl and NF-κB were decreased significantly in human breast cancer cell line with the increase of dosage of curcumin（P〈0.05）, and with the extension of time course（P〈0.05）. These changes suggested a dose- and time-dependent manner. The proliferation rate of cells also was significantly inhibited（P〈0.05）. Conclusion： The current results show that the Notch-1 signaling pathway is associated mechanistically with NF-κB activity during curcumin-induced cell growth inhibition and apoptosis of breast cancer cells. These results suggest that the down-regulation of Notch signaling by curcumin may be a novel strategy for the treatment of patients with breast cancer.

Several new treatment options for gastric cancer have been introduced but the prognosis of patients diagnosed with gastric cancer is still poor. Disease prognosis could be improved for high-risk individuals by implementing earlier screenings. Because many patients are asymptomatic during the early stages of gastric cancer,the diagnosis is often delayed and patients present with unresectable locally advanced or metastatic disease. Cytotoxic treatment has been shown to prolong survival in general,but not all patients are responders. The application of targeted therapies and multimodal treatment has improved prognosis for those with advanced ***,these new therapeutic strategies do not uniformly benefit all *** whether patients will respond to specific therapies would be of particular value and would allow for stratifying patients for personalized treatment *** imaging by positron emission tomography was the first technique with the potential to predict the response of esophagogastric cancer to neoadjuvant *** and validating tissue-based biomarkers are ongoing *** this review,we discuss the status of several targeted therapies for gastric cancer,as well as proteomic and metabolic methods for investigating biomarkers for therapy response prediction in gastric cancer.

BACKGROUND Acquired hemophilia A (AHA) is a rare and potentially severe bleeding disordercaused by circulating autoantibodies against factor Ⅷ (FⅧ). In approximately50% of the patients, the condition is associated with autoimmune diseases,cancers, medication use, pregnancy, and the post-partum period. Bullous pemphigoid(BP) is a chronic autoimmune subepidermal blistering disease associatedwith tissue-bound and circulating autoantibodies against BP antigens 180 (BP180)and 230 (BP230). AHA-associated BP has a high mortality rate;hence, the understandingof this disease must *** SUMMARY A 69-year-old man presented with erythema, blisters, blood blisters, and crustsaccompanied by severe pruritus for more than 20 days, and ecchymosis andswelling on his left upper arm for 3 days. Pathological examination revealed asubepidermal blister that contained eosinophils. Laboratory tests showed that theBP180 autoantibody levels had increased, isolated activated partial thromboplastintime was notably prolonged (115.6 s), and coagulation FⅧ activity wasextremely low (< 1.0%). Furthermore, the FⅧ inhibitor titer had greatlyincreased (59.2 Bethesda units). Therefore, the patient was diagnosed as having BP associated with AHA, prescribed 0.05% topical halometasone cream, and transferred to a higher-level hospitalfor effective treatment;however, he died after 2 *** AHA associated BP is rare, dangerous, and has a high mortality rate. Therefore, its timely diagnosis and effectivetreatment are necessary.

Perivascular epithelioid cell tumors （PEComas） are a family of rare mesenchymal neoplasms. The PEComas, composed of epithelioid and spindle cells, have the same cellular and immtmohistocherrtical features but are found in different visceral and soft tissue sites. Here, we report the histological and immunohistochemical features of one case of PEComa restricted in the pelvic visceral peritoneum of a male patient. The patient was treated with radical surgery, and was well and on follow-up visits without tumor recurrence.
