Immunosuppressive tumor microenvironment contributes to tumor progression in diffuse large B-cell lymphoma upon anti-CD19 chimeric antigen receptor T therapy

Anti-CD19 chimeric antigen receptor(CAR)-T cell therapy has achieved 40%–50%long-term complete response in relapsed or refractory diffuse large B-cell lymphoma(DLBCL)***,the underlying mechanism of alterations in the tumor microenvironments resulting in CAR-T cell therapy failure needs further investigation.A multi-center phase I/II trial of anti-CD19 CD28z CAR-T(FKC876,ChiCTR1800019661)was *** 22 evaluable DLBCL patients,seven achieved complete remission,10 experienced partial remissions,while four had stable disease by day ***-cell RNA sequencing results were obtained from core needle biopsy tumor samples collected from long-term complete remission and early-progressed patients,and compared at different stages of treatment.M2-subtype macrophages were significantly involved in both in vivo and in vitro anti-tumor functions of CAR-T cells,leading to CAR-T cell therapy failure and disease progression in *** tumor microenvironments persisted before CAR-T cell therapy,during both cell expansion and disease progression,which could not be altered by infiltrating CAR-T *** metabolism profile of M2-subtype macrophages and those of dysfunctional T cells also contributed to the immunosuppressive tumor ***,our findings provided a clinical rationale for targeting tumor microenvironments and reprogramming immune cell metabolism as effective therapeutic strategies to prevent lymphoma relapse in future designs of CAR-T cell therapy.

Spatial-temporal change in urban agricultural land use efficiency from the perspective of agricultural multi-functionality： A case study of the Xi＇an metropolitan zone

The excessive expansion of urbanized areas has resulted in haphazard land utili- zation, immoderate consumption of superior agricultural land and water resources, significant fragmentation of agricultural landscape, and gradual deterioration of the agro-ecological en- vironment. Combined, these factors cause poor land use efficiency. Under these circum- stances, comprehensively assessing land use efficiency for urban agriculture is a key issue in land use research. Currently, evaluation methods for agricultural land use efficiency narrowly concentrate on aspects of economic input and output. However, urban agro-ecosystems can provide diverse economic, social, and ecological services and functions. In particular, the social and ecological services and functions originating from agricultural land, which have a higher value than economic services, play a significant role in ensuring regional social, eco- logical, and environmental security. However, recent research has rarely taken these benefits into consideration. Therefore, land use value has been greatly underestimated, which has resulted in mishandled and poor land use policies. In this study, we apply Landsat imagery and social and economic statistical data for the Xi＇an metropolitan zone （XMZ） to investigate agricultural multi-functionality. We develop an evaluation framework for urban agricultural land use efficiency and identify agro-ecosystem services and functions as important outputs from agricultural land. The land use efficiency of urban agriculture is then evaluated using ecosystem services models, providing a mechanism for assessing spatial-temporal changes in land use efficiency in the XMZ from 1999 to 2015. Four important conclusions are reached from this analysis. First, the rapid urbanization and agricultural transformation from traditional cereal cultivation to modern urban agriculture has resulted in steadily increasing costs, out- puts, and land use efficiency of urban agriculture. Increasing wheat yield is a long-term goal for wheat breeders around the *** elite genetic resources and dissecting the genetic basis of important agronomic traits in wheat are the necessary methods for high-yield wheat *** study evaluated nine crucial agronomic traits found in a natural population of 156 wheat varieties and77 landraces from Sichuan,China in seven environments over two *** results of this investigation of agronomic traits showed that the landraces had more tillers and higher kernel numbers per spike (KNS),while the breeding varieties had higher thousand-kernel weight (TKW) and kernel weight per spike (KWS).The generalized heritability (H2) values of the nine agronomic traits varied from 0.74 to *** analysis suggested that the natural population could be divided into three groups using 43 198 single nucleotide polymorphism (SNP) markers from the wheat 55K SNP chip.A total of 67 quantitative trait loci (QTLs) were identified by the genome-wide association study (GWAS) analysis based on the Q+K method of a mixed linear *** important QTLs were analyzed in this *** haplotypes of ***-7BL.1 for fertile tillers number (FTN),three haplotypes of ***-1AL.2 for KNS,and four haplotypes of ***-3BS.1 for TKW were ***-Hap2,KNS-Hap1,and TKW-Hap2 were excellent haplotypes for each QTL based on the yield performance of 42 varieties in regional trials from 2002 to *** varieties with all three haplotypes showed the highest yield compared to those with either two haplotypes or one *** addition,the KASP-AX-108866053 marker of QTL ***-1AL.2 was successfully distinguished between three haplotypes(or alleles) in 63 varieties based on the number of kernels per spike in regional trials between 2018 and *** genetic loci and reliable makers can be applied in marker-assisted selection or map-based gene cloning for the genetic improvement of wheat yield.

Background:Large uncertainty in modeling land carbon(C)uptake heavily impedes the accurate prediction of the global C *** the uncertainty sources among models is crucial for model improvement yet has been difficult due to multiple feedbacks within Earth System Models(ESMs).Here we present a Matrix-based ensemble Model Inter-comparison Platform(MEMIP)under a unified model traceability framework to evaluate multiple soil organic carbon(SOC)*** the MEMIP,we analyzed how the vertically resolved soil biogeochemistry structure influences SOC prediction in two soil organic matter(SOM)*** comparing the model outputs from the C-only and CN modes,the SOC differences contributed by individual processes and N feedback between vegetation and soil were explicitly ***:Results showed that the multi-layer models with a vertically resolved structure predicted significantly higher SOC than the single layer models over the historical simulation(1900–2000).The SOC difference between the multi-layer models was remarkably higher than between the single-layer *** analysis indicated that over 80%of the SOC increase in the multi-layer models was contributed by the incorporation of depth-related processes,while SOC differences were similarly contributed by the processes and N feedback between models with the same soil depth ***:The output suggested that feedback is a non-negligible contributor to the inter-model difference of SOC prediction,especially between models with similar process *** analysis with TRenDY v7 and more extensive MEMIP outputs illustrated the potential important role of multi-layer structure to enlarge the current ensemble spread and the necessity of more detail model decomposition to fully disentangle inter-model *** stressed the importance of analyzing ensemble outputs from the fundamental model structures,and holding a holistic view in understanding the en

Linked to major volcanic eruptions around 536 and 540 CE, the onset of the Late Antique Little Ice Age has been described as the coldest period of the past two millennia. The exact timing and spatial extent of this exceptional cold phase are, however, still under debate because of the limited resolution and geographical distribution of the available proxy archives. Here, we use 106 wood anatomical thin sections from 23forest sites and 20 tree species in both hemispheres to search for cell-level fingerprints of ephemeral summer cooling between 530 and 550 CE. After cross-dating and double-staining, we identified 89Blue Rings(lack of cell wall lignification), nine Frost Rings(cell deformation and collapse), and 93Light Rings(reduced cell wall thickening) in the Northern Hemisphere. Our network reveals evidence for the strongest temperature depression between mid-July and early-August 536 CE across North America and Eurasia, whereas more localised cold spells occurred in the summers of 532, 540–43, and548 CE. The lack of anatomical signatures in the austral trees suggests limited incursion of stratospheric volcanic aerosol into the Southern Hemisphere extra-tropics, that any forcing was mitigated by atmosphere-ocean dynamical responses and/or concentrated outside the growing season, or a combination of factors. Our findings demonstrate the advantage of wood anatomical investigations over traditional dendrochronological measurements, provide a benchmark for Earth system models, support cross-disciplinary studies into the entanglements of climate and history, and question the relevance of global climate averages.

High entropy alloys(HEAs)contain near equimolar amounts of five or more elements and are a compelling space for materials *** the design of HEAs,great emphasis is placed on identifying thermodynamic conditions for single-phase and multi-phase stability regions,but this process is hindered by the difficulty of navigating stability relationships in high-component *** phase diagrams use barycentric coordinates to represent composition axes,which require(N–1)spatial dimensions to represent an N-component system,meaning that HEA systems with N>4 components cannot be readily ***,we propose forgoing barycentric composition axes in favor of two energy axes:a formation-energy axis and a‘reaction energy’*** Inverse Hull Webs offer an information-dense 2D representation that successfully captures complex phase stability relationships in N≥5 component *** use our proposed diagrams to visualize the transition of HEA solid-solutions from high-temperature stability to metastability upon quenching,and identify important thermodynamic features that are correlated with the persistence or decomposition of metastable HEAs.

2008年M_W7.9汶川地震导致龙门山断裂发生强烈地壳变形,同时引发的巨量同震滑坡加速了该地区的地表剥蚀和河流侵蚀.然而,目前尚缺少系统的数据定量研究滑坡物质的运移以及河流侵蚀速率随时间的演化规律,这些对理解龙门山前缘物质的再分配以及强震对活动造山带地形塑造的作用至关重要.为此,本研究在汶川地震后的6年间,对震区沱江上游3条支流湔江、石亭江、绵远河流域进行了多期次的定点现代河沙采样.通过系统测量河沙中的石英10Be浓度,并与震前已发表的数据进行对比,发现如下基本特点:(1)震后河沙10 Be浓度均有明显降低,表明同震滑坡物质对河沙的稀释作用;(2)震后河流对河沙的运移量增加为震前的1.3~18.5倍,因此震后龙门山地区侵蚀速率短期显著增加;(3)初步估计得到汶川地震产生的滑坡物质被完全运移出造山带所需要的时间至少为100~4000年,接近龙门山地区强震复发周期;(4)震间和同震产生的构造变形和地表剥蚀在空间上具有互补性.考虑到地表剥蚀引起的地壳均衡反弹效应,认为类似汶川地震的强震有利于龙门山的隆升.认识震前、震时和震后的地壳变形及侵蚀过程有助于更好地理解单次强震事件对高原边界龙门山地形演化的作用.

Cultivated potato is a clonally propagated autotetraploid species with a highly heterogeneous *** assemblies of six cultivars including two chromosome-scale phased genome assemblies revealed extensive allelic diversity,including altered coding and transcript sequences,preferential allele expression,and structural variation that collectively result in a highly complex transcriptome and predicted proteome,which are distributed across the homologous *** species contribute to the extensive allelic diversity in tetraploid cultivars,demonstrating ancestral introgressions predating modern breeding *** a clonally propagated autotetraploid that undergoes limited meiosis,dysfunctional and deleterious alleles are not purged in tetraploid *** a quarter of the loci bore mutations are predicted to have a high negative impact on protein function,complicating breeder’s efforts to reduce genetic *** StCDF1 locus controls maturity,and analysis of six tetraploid genomes revealed that 12 allelic variants of StCDF1 are correlated with maturity in a dosage-dependent *** of the complexity of the tetraploid potato genome with its rampant structural variation and embedded deleterious and dysfunctional alleles will be key not only to implementing precision breeding of tetraploid cultivars but also to the construction of homozygous,diploid potato germplasm containing favorable alleles to capitalize on heterosis in F1 hybrids.

In 2010,a major scientific milestone was achieved for tree fruit crops:publication of the first draft whole genome sequence(WGS)for apple(Malus domestica).This WGS,v1.0,was valuable as the initial reference for sequence information,fine mapping,gene discovery,variant discovery,and tool development.A new,high quality apple WGS,GDDH13 v1.1,was released in 2017 and now serves as the reference genome for *** the past decade,these apple WGSs have had an enormous impact on our understanding of apple biological functioning,trait physiology and inheritance,leading to practical applications for improving this highly valued *** gene identities for phenotypes of fundamental and practical interest can today be discovered much more ***-wide polymorphisms at high genetic resolution are screened efficiently over hundreds to thousands of individuals with new insights into genetic relationships and ***-density genetic maps are constructed efficiently and quantitative trait loci for valuable traits are readily associated with positional candidate genes and/or converted into diagnostic tests for *** understand the species,geographical,and genomic origins of domesticated apple more precisely,as well as its relationship to wild *** WGS has turbo-charged application of these classical research steps to crop improvement and drives innovative methods to achieve more durable,environmentally sound,productive,and consumer-desirable apple *** review includes examples of basic and practical breakthroughs and challenges in using the apple *** for“what’s next”focus on necessary upgrades to the genome sequence data pool,as well as for use of the data,to reach new frontiers in genomics-based scientific understanding of apple.
