Salicylic acid(SA) plays a pivotal role in delaying fruit ripening and senescence. However, little is known about its underlying mechanism of action. In this study, RNA sequencing was conducted to analyze and compare the transcriptome profiles of SA-treated and control pear fruits. We found a total of 159 and 419 genes differentially expressed between the SA-treated and control pear fruits after 12 and 24 h of treatment, respectively. Among these differentially expressed genes(DEGs), 125 genes were continuously differentially expressed at both treatment times, and they were identified as candidate genes that might be associated with SA-regulated fruit ripening and senescence. Bioinformatics analysis results showed that 125 DEGs were mainly associated with plant hormone biosynthesis and metabolism, cell wall metabolism and modification, antioxidant systems, and senescence-associated transcription factors. Additionally, the expression of several candidate DEGs in ripening and senescent pear fruits after SA treatments were further validated by quantitative real-time PCR(qRT-PCR). This study provides valuable information and enhances the understanding of the comprehensive mechanisms of SA-meditated pear fruit ripening and senescence.

Leaf growth and its interaction with the growing environment critically affect leaf area, distribution, and function, and ultimately affects grain yield of maize(Zea mays L.). To detect the effects of leaf area dynamics, growth periods, and the environment on maize grain yield, a three-year field experiment was conducted using two maize varieties, medium plant-size variety Zhengdan 958(ZD958) and large plant-size variety Zhongnongda 4(ZND4), and three to five sowing dates. The sowing date significantly affected maize yield as a result of changes in leaf area, growth stage, and growing environment. Prior to the 12 th leaf stage, significant correlations between leaf area dynamics, environment, and yield were seldom detected. The expansion of leaf area from 12 th leaf stage to silking stage was significantly positively correlated with growing degree days(GDD), solar radiation, and grain yield, indicating the importance of leaf area dynamics during this period. After silking, solar radiation played a more important role in inducing leaf senescence than GDD, particularly in the 2 nd half of the grain filling stage. Accelerated leaf senescence in late growth period can increase maize yield. The environment affected leaf area dynamics and yield of the large plant-size variety(ZND4) more easily than the medium plant-size variety(ZD958) at the optimum plant density, reflecting the difference in varietal capacity to adapt to the growing environment. This study indicates that optimizing the interaction among leaf area dynamics, growth periods, and environment is a sound strategy to increase maize yield. Favorable interactions are useful to determine the optimal sowing date of a given variety.

The photoperiod response of lowering is critical to the survival and successful reproduction in crops. The Hd6 gene, encoding a protein similar to the Arabidopsis α-subunit of CASEIN KINASE2α （CK2α） has been reported to affect flowering time through the photoperiodic pathway in rice. In this study, the maize ortholog, ZmHd6, has been cloned and its role in the photoperiodic pathway was investigated. ZmHd6 encodes 332 amino acids with six distinct domains： two CK2 active domains, a nuclear localization signal sequence, a N terminal domain, an ATP binding site, and an active site of serine/threonine protein kinase. ZmHd6 was constitutively expressed in the leaves and shoot apical meristem （SAM） with the highest expression in SAMs of plants with seven leaves under long and short day conditions. Delayed flowering time under long day （LD） conditions were observed in ZmHd6 overexpression transgenic Arabidopsis plants compared to the wild type. In transgenic Arabidopsis plants, ZmHd6 was expressed in a circadian oscillatory pattern, TOC1 peak expression was delayed, new GI, CO, and FT expressions reached to peak after 3 h while entering illumination period and oscillatory expression patterns of CO and FT were created during the dark phase. These results suggested that ZmHd6 might negatively regulate flowering time under LD conditions by delaying TOC1 peak expression and disrupting CO and FT expression in transgenic Arabidopsis plants.

Isolation and Characterization of Fucoidans from Five Brown Algae and Evaluation of Their Antioxidant Activity

In this study, we evaluated the chemical property and antioxidant activity of fucoidans isolated from brown algae, Laminaria japonica(LJF), Lessonia nigrescens(LNF), Lessonia trabeculata(LTF), Ascophyllum mackaii(AMF), and Ecklonia maxima(EMF). LJF was less in sulfate content(14.16%) and more in galactose and mannose content(1.08 and 0.68) than the documented early. EMF contained 20%–30% of sulfate and fucose, 0.97 in molar ratio which was lower than that of sulfate to other four fucoidans(1.21–1.41). AMF(162 kDa) and EMF(150 kDa) were the first two largest in molecular weight, which were followed by LJP(126 kDa), LNF(113 kDa) and LTF(105 kDa). The fucoidans isolated these algae showed a wide range of antioxidant activity in vitro. It was found that the reducing power of the isolated fucoidans was positively correlated with their sulfate content and molecular weight. In addition, LNF and LTF at low concentrations exhibited high superoxide and hydroxyl radical scavenging activity. This demonstrated that low molecular weight fucoidans may perform a high antioxidant activity.

Analysis of the Fungal Community in Apple Replanted Soil Around Bohai Gulf

Apple replant disease(ARD) is a frequently occurring plant disease in replanted orchards around Bohai Gulf, which causes growth inhibition and even death of plants. The aim of this study was to investigate the etiology of ARD around Bohai Gulf. In this study, the primary growth inhibition of apple seedlings was evaluated in ten replanted soils, sampled around Bohai Gulf. A fungal clone library was used to identify changes in the structure and composition of the soil fungal community. The results revealed that the Simpson diversity indices of Laizhou and Pulandian orchards were higher than others, presenting severe ARD. Ascomycota dominated around Bohai Gulf at the phyla level. Fusarium and Saccharomyces were abundant in all replanted soils. In addition, correlations between the relative abundance of fungal genera in soils and the severity of ARD were analyzed. The results showed that Fusarium was correlated positively with the severity of ARD, but Mortierella was negatively correlated. Furthermore, the quantitative PCR of Fusarium oxysporum, which was regarded as a factor of ARD, was performed. Overall, this study demonstrated that ARD was strongly associated with an unbalanced microbial ecosystem with more pathogenic fungi, while Fusarium in the apple replanted soil was the key factor for ARD around Bohai Gulf.

A hydroponics experiment was conducted to evaluate the role of potassium (K) and silicon (Si) in mitigating the deleterious effects of NaCl on sugarcane genotypes differing in salt *** salt-sensitive (CPF 243 and SPF 213) and two salt-tolerant (HSF 240 and CP 77-400) sugarcane genotypes were grown for six weeks in 1/2 strength Johnson's nutrient *** nutrient solution was salinized by two NaCl levels (0 and 100 mmol L 1 NaCl) and supplied with two levels of K (0 and 3 mmol L 1) and Si (0 and 2 mmol L 1).Applied NaCl enhanced Na + concentration in plant tissues and significantly (P ≤ 0.05) reduced shoot and root dry matter in four sugarcane ***,the magnitude of reduction was much greater in salt-sensitive genotypes than salt-tolerant *** salts interfered with the absorption of K + and Ca 2+ and significantly (P ≤ 0.05) decreased their uptake in sugarcane *** of K and Si either alone or in combination significantly (P ≤ 0.05) inhibited the uptake and transport of Na + from roots to shoots and improved dry matter yields under NaCl *** uptake,K + /Na + ratios,and Ca 2+ and Si uptake were also significantly (P ≤ 0.05) increased by the addition of K and/or Si to the root *** this study,K and Si-enhanced salt tolerance in sugarcane genotypes was ascribed to decreased Na + concentration and increased K + with a resultant improvement in K + /Na + ratio,which is a good indicator to assess plant tolerance to salt ***,further verification of these results is warranted under field conditions.

Human induced pluripotent stem cells （iPSCs） hold great promise for regenerative med- icine. Generating iPSCs from immunologically immature newborn umbilical cord blood mononu- clear cells （UCBMCs） is of great significance. Here we report generation of human iPSCs with great efficiency from UCBMCs using a dox-inducible lentiviral system carrying four Yamanaka factors. We generated these cells by optimizing the existing iPSC induction protocol. The UCBMC-derived iPSCs （UCB-iPSCs） have characteristics that are identical to pluripotent human embryonic stem cells （hESCs）. This study highlights the use of UCBMCs to generate highly functional human iPSCs that could accelerate the development of cell-based regenerative therapy for patients suffering from various diseases.

As an essential plant macronutrient, the low availability of phosphorus （P） in most soils imposes serious limitation on crop production. Plants have evolved complex responsive and adaptive mechanisms for acquisition, remobilization and recycling of phosphate （Pi） to maintain P homeostasis. Spatio-temporal molecular, physiological, and biochemical Pi deficiency responses developed by plants are the consequence of local and systemic sensing and signaling pathways. Pi deficiency is sensed locally by the root system where hormones serve as important signaling components in terms of developmental reprogramming, leading to changes in root system architecture. Root-to-shoot and shoot-to-root signals, delivered through the xylem and phloem, respectively, involving Pi itself, hormones, miRNAs, mRNAs, and sucrose, serve to coordinate Pi deficiency responses at the whole-plant level. A combination of chromatin remodeling, transcriptional and posttranslational events contribute to globally regulating a wide range of Pi deficiency responses. In this review, recent advances are evaluated in terms of progress toward developing a comprehen- sive understanding of the molecular events underlying control over P homeostasis. Application of this knowledge, in terms of developing crop plants having enhanced attributes for P use efficiency, is discussed from the perspective of agricultural sustainability in the face of diminishing global P supplies.

ARF2 coordinates with PLETHORAs and PINs to orchestrate ABA-mediated root meristem activity in Arabidopsis

Multiple hormones, including abscisic acid（ABA） and auxin, regulate cell division and differentiation of Arabidopsis root meristems. AUXIN RESPONSE FACTOR2（ARF2） functions as a negative regulator of ABA responses, as seed germination and primary root growth of arf2 mutants are hypersensitive to ABA. In this study, we found that ABA treatment reduced the expression levels of the PIN-FORMEDs（PIN） auxin efflux carriers, PIN1, PIN3,PIN4, and PIN7, to a greater extent in the root meristems of arf2-101 mutant than in the wild type. Also, arf2-101 pin1 and arf2-101 pin4 double mutants show less ABA-induced inhibition of root meristem activity than the arf2-101 mutants. Furthermore, ARF2 positively mediates the transcripts of transcription factor PLETHORA 1（PLT1）gene but negatively mediates PLT2 at protein level in root meristems. Using a dexamethasone（DEX）-inducible transgenic line, Pro35S：PLT2-GR, we showed that PLT2 greatly promotes cell division and completely inhibits cell differentiation in root meristems of the arf2-101 mutant once PLT2 is induced by DEX, which can be partially reversed by ABA treatment, suggesting that ABA regulates root meristem activity in both ARF2-dependent and independent pathways. Our results uncover a complex regulatory architecture in which ARF2 coordinates with PLTs and PINs to orchestrate ABA-mediated regulation of root meristem activity in Arabidopsis.
