Germination is the first and maybe the foremost growth stage in the life cycle of a plant. Herein, we report that initiation of germination in the Arabidopsis Columbia ecotype was accompanied by a sharp decrease in the amount of extractable total RNA. At the beginning of our germination experiment, we were usually able to obtain 35-40 IJg total RNA from 100 mg dry seeds. However, after 3 d of cold stratification, we could only obtain less than 5 μg total RNA from the same amount of starting material. Young seedlings contained approximately 100 μg total RNA per 100 mg fresh tissue. Further studies showed that inhibition of de novo RNA synthesis by actinomycin D prevented the degradation of parental RNA and, in the meantime, significantly delayed the germination process. Several ribonuclease-like genes that were highly expressed in dry seeds, and especially during the cold stratification period, were discovered. We propose that these enzymes are involved in the regulation of parental RNA degradation. These results indicate that parental RNA metabolism may be an important process for Arabidopsis seed germination.

Effects of Rice Straw and Its Biochar Addition on Soil Labile Carbon and Soil Organic Carbon

Whether the biochar amendment could affect soil organic matter （SOM） turnover and hence soil carbon （C） stock remains poorly understood. Effects of the addition of ^13C-labelled rice straw or its pyrolysed biochar at 250 or 350℃ to a sugarcane soil （Ferrosol） on soil labile C （dissolved organic C, DOC; microbial biomass C, MBC; and mineralizable C, MC） and soil organic C （SOC） were investigated after 112 d of laboratory incubation at 25℃. Four treatments were examined as （1） the control soil without amendment （Soil）; （2） soil plus ^13C-labelled rice straw （Soil＋Straw）; （3） soil plus 250℃ biochar （Soil＋B250） and （4） soil plus 350℃biochar （Soil＋B350）. Compared to un-pyrolysed straw, biochars generally had an increased aryl C, carboxyl C, C and nitrogen concentrations, a decreased O-alkyl C and C：N ratio, but similar alkyl C and δ^13C （1 742- 1 877 %）. Among treatments, significant higher DOC, MBC and MC derived from the new C （straw or biochar） ranked as Soil＋Straw〉Soil＋B250〉Soil＋B350, whilst significant higher SOC from the new C as Soil＋B250〉Soil＋Straw≈Soil＋B350. Compared to Soil, DOC and MBC derived from the native soil were decreased under straw or biochar addition, whilst MC from the native soil was increased under straw addition but decreased under biochar addition. Meanwhile, native SOC was similar among the treatments, irrespective of the straw or biochar addition. Compared to Soil, significant higher total DOC and total MBC were under Soil＋Straw, but not under Soil＋B250 and Soil＋B350, whilst significant higher total MC and total SOC were under straw or biochar addition, except for MC under Soil＋B350. Our results demonstrated that the application of biochar to soil may be an appropriate management practice for increasing soil C storage.

Diacetylenic glycero-phosphatidylcholine is a chiral molecule with amphiphilic property, and it can self-assembly into a lipid microtubular structure. The lipid microtubule is a stable structure formed by tightly wound helical ribbons, and the ribbon-wrapping patterns have a significant effect on their chemical deposition on the microtubules. The deposition of colloidal Pd catalyst occurs mainly on the helical edge of the wound helical ribbons to form helical deposition lines of colloidal Pd particles in the interior and exterior of the lipid microtubules, resulting in an uneven chemical deposition of Ni on the microtubules. Catalyzed by as-deposited colloidal Pd, metallized Ni microtubules are characterized by a helical form, which may be in relation to inner stress due to the thickness difference or the different deposition processes. The observation of microtom shows that metallized tubules have a hollow structure. Some metallized tubules have a kind of coaxial double layer structure observed in the direct experiment evidence, indicating that metallization can occur in the inner and outer surface of the lipid tubules. Both lipid microtubules and metallized microtubules can be used as vehicles for encapsulating biological active molecules to control their release and to develop micro-components in biological and mechanical systems. With the aim of designing and preparing the materials based on polyoxometalates(POMs)and the metal-organic frameworks(MOFs)toward desired applications,a facile approach was developed to incorporate the clusters of Mo7O24^6-(Mo7),a representative isopolyoxometalate,into the vacant cages of *** the so-called strategy of building MOF bottles around the POM ships,the powdered crystals and crystalline membrane of Mo7@ZIF-8 were synthesized,*** products were characterized by IR,PXRD,N2 adsorption,and *** Mo7@ZIF-8 crystals could effectively adsorb malachite green(MG)from water,and the maximum adsorption capacity reached 1963 mg·g^-*** Mo7@ZIF-8 crystals could also be used for selectively adsorbing MG from the mixed dye solution containing MG and methyl orange(MO).Moreover,the Mo7@ZIF-8 membrane exhibited the proton conductivity of 9.5×10^-2 S·cm^-1 at 75℃under 98%relative humidity.

Reproductive toxicity of brazilein in ICR mice

Brazilein is an active small molecular compound extracted from Caesalpinia sappan L. with favorable pharmacological properties on immune system, cardiovascular system, and nervous system. C. sappan has been used as a traditional medicine in China for hundreds of years for various diseases. However, the general reproductive toxicity of brazilein is still unknown. The purpose of the present study was to thoroughly evaluate the general reproductive toxicity of brazilein in ICR mice to support the future drug development and modernization of this potent traditional Chinese medicine. The results showed that, although no apparent toxicity on the reproducibility of the male was observed, brazilein might cause considerable risks to the fetuses and females as indicated by the ratios of dead fetuses and reabsorptions. In conclusion, our results from the present study provided some useful insights about the safety profile of brazilein, suggesting that brazilein should be used with caution in pregnant women.

DNA barcodes have been proposed as a shortcut to provide species identification and as a way to accelerate the discovery of new species. A number of candidate gene regions have been suggested as possible barcodes for animals and plants, but for the identification of recently diverged species and/or varieties with only a few genetic differences it has been reported to be problematic in some cases. This study selected widely cultivated cruciferous vegetables as the primary samples, after failure of discrimination of each species using current DNA barcodes, we performed the fluorescent amplified fragment length polymorphism （F-AFLP） and successfully discriminated each species, subspecies, variety and their cultivar in 74 samples. Then the non-qualitative results obtained from F-AFLP were transformed into two-dimensional barcodes image file of each cultivar via the PDF417 software. This method was also successfully applied to the discrimination of 17 Chinese indigenous pig breeds. The barcode we constructed which greatly reduces the information storage space is genotypes-specific, and can be conveniently decoded into the original data and thereby be conveniently shared and referred to. We believe that it is possible to construct a new data sharing molecular barcode system that could discriminate the subspecies, varieties, cultivars and even individuals with close genetic relationships.

Arabidopsis AtVPS15 Plays Essential Roles in Pollen Germination Possibly by Interacting with AtVPS34

VPS 15 protein is a component of the phosphatidylinositol 3-kinase complex which plays a pivotal role in the development of yeast and mammalian cells. The knowledge about the function of its homologue in plants remains limited. Here we report that AtVPS15, a homologue of yeast VPS15p in Arabidopsis, plays an essential role in pollen germination. Homozygous T-DNA insertion mutants of AtVPS15 could not be obtained from the progenies of self-pollinated heterozygous mutants. Reciprocal crosses between atvps15 mutants and wild-type Arabidopsis revealed that the T-DNA insertion was not able to be transmitted by male gametophytes. DAPI staining, Alexander＇s stain and scanning electron microscopic analysis showed that atvps15 heterozygous plants produced pollen grains that were morphologically indistinguishable from wild-type pollen, whereas in vitro germination experiments revealed that germination of the pollen grains was defective. GUS staining analysis of transgenic plants expressing the GUS reporter gene driven by the AtVPS15 promoter showed that AtVPS15 was mainly expressed in pollen grains. Finally, DUALmembrane yeast two-hybrid analysis demonstrated that AtVPS15 might interact directly with AtVPS34. These results suggest that AtVPS15 is very important for pollen germination, possibly through modulation of the activity of PI3-kinase.

ZmWRKY104 positively regulates salt tolerance by modulating ZmSOD4 expression in maize

Salinity impairs plant growth, limiting agricultural development. It is desirable to identify genes responding to salt stress and their mechanism of action. We identified a function of the Zea mays WRKY transcription factor, Zm WRKY104, in salt stress response. Zm WRKY104 was localized in the nucleus and showed transcriptional activation activity. Phenotypic and physiological analysis showed that overexpression of Zm WRKY104 in maize increased the tolerance of maize to salt stress and alleviated salt-induced increases in O;accumulation, malondialdehyde(MDA) content, and percent of electrolyte leakage. Further investigation showed that Zm WRKY104 increased SOD activity by regulating Zm SOD4 expression. Yeast onehybrid, electrophoretic mobility shift test, and chromatin immunoprecipitation–quantitative PCR assay showed that Zm WRKY104 bound directly to the promoter of Zm SOD4 by recognizing the W-box motif in vivo and in vitro. Phenotypic, physiological, and biochemical analysis showed that Zm SOD4 increased salt tolerance by alleviating salt-induced increases in O;accumulation, MDA content, and percent of electrolyte leakage under salt stress. Taken together, our results indicate that Zm WRKY104 positively regulates Zm SOD4 expression to modulate salt-induced O;accumulation, MDA content, and percent of electrolyte leakage, thus affecting salt stress response in maize.

Based on both the spin diffusion equation and the Landau-LlTshitz-Gilbert （LLG） equation, we demonstrate the influence of out-of-plane spin torque on magnetization switching and susceptibility in a magnetic multilayer system. The variation of spin accumulation and local magnetization with respect to time are studied in the magnetization reversal induced by spin torque. We also research the susceptibility subject to a microwave magnetic field, which is compared with the results obtained without out-of-plane torque.
